Les récepteurs olfactifs et le codage des odeurs Inra

La première étape de la détection olfactive implique l’activation de récepteurs olfactifs par les molécules odorantes. Ces récepteurs sont des protéines membranaires contenues dans la membrane des cils portés par les neurones sensoriels olfactifs localisés dans l’épithélium olfactif. Le codage de l’odeur résulte de l’activation combinatoire d’un ensemble de récepteurs et repose sur leur expression clonale. La connectique neuronale aboutit à la formation, dans le cortex, d’une image sensorielle olfactive qui est perçue consciemment en tant qu’odeur. Ce système combinatoire permet, avec seulement environ 340 récepteurs différents, de discriminer des myriades de molécules odorantes naturelles ou non (nouvelles flaveurs culinaires, molécules de synthèse,...

Structure and stability of a rat odorant-binding protein: another brick in the wall Inra

The effect of temperature on the structure of the rat odorant-binding protein was investigated by spectroscopic and in silico methodologies. In particular, in this work, we examined the structural features of the rat OBP-1F by Fourier-transform infrared spectroscopy and molecular dynamics investigations. The obtained spectroscopic results were analyzed using the following three different methods based on the unexchanged amide hydrogens of the protein sample: (1) the analysis of difference spectra; (2) the generalized 2D-IR correlation spectroscopy; (3) the phase diagram method. Sulfur single-wavelength anomalous diffraction crystal structure of a pheromone-binding protein from the honeybee Apis mellifera L. Inra

Pheromone binding proteins (PBPs) are small helical proteins (∼13-17 kDa) present in several sensory organs from moth and other insect species. They are involved in the transport of pheromones from the sensillar lymph to the olfactory receptors. We report here the crystal structure of a PBP (Amel-ASP1) originating from the honey-bee (Apis mellifera) antennae and expressed as recombinant protein in the yeast Pichia pastoris. Crystals of Amel-ASP1 were obtained at pH 5.5 using the nano-drops technique of crystallization with a novel optimization procedure, and the structure was solved initially with the single-wavelength anomalous diffraction technique using sulfur anomalous dispersion.
