La variabilité génétique du peuplier lui assure la place de modèle pour une étude fine des gènes impliqués dans des fonctions adaptatives ou de production de bois. Une meilleure connaissance des déterminismes génétiques doit permettre la création de cultivars performants.

Characterization of two major genetic factors controlling quantitative resistance to Melampsora larici-populina leaf rust in hybrid poplars : strain specificity, field expression, combined effects, and relationship with a defeated qualitative resistance gene Inra

Two genetic factors explain a significant proportion of the variability for quantitative resistance to Melampsora larici-populina leaf rust in a Populus deltoides × P. trichocarpa F1 progeny. One is inherited from P. deltoides and is associated with a defeated qualitative resistance geneR1, and the other, RUS, is inherited from P. trichocarpa. To assess the potential contribution of these two factors for durable resistance breeding, 284 genotypes from this F1 progeny were studied in laboratory experiments with three M. larici-populina strains and in a field experiment under natural inoculum pressure. • In order to elucidate the genetic control of resistance to Melampsora larici-populina leaf rust in hybrid poplars, a Populus deltoides × P. trichocarpa F1 progeny was analysed for qualitative and quantitative rust resistances.• This progeny was evaluated for three components of quantitative resistance (latent period, uredinia number and uredinia size) to seven M. larici-populina strains in controlled conditions, and for one component of field susceptibility (rust colonization on the most infected leaf).• One qualitative resistance locus inherited from P. deltoides, R1, was localized on the genetic map. Qualitative resistance to Melampsora larici-populina leaf rust inherited from North American species Populus deltoides did not allow for durable control of this pathogen in interspecific hybrid cultivars. Despite significant levels of strain-specificity, quantitative resistance would exert lower selection pressures on the pathogen populations, and hence could be more durable. Previous studies restricted to a large P. × interamericana (i.e., P. deltoides × Populus trichocarpa) F1 family revealed that the presence of R1, a segregating defeated qualitative resistance gene inherited from P. deltoides, had major beneficial effects on quantitative resistance. Resources and potential for candidate-gene association studies in European black poplar for leaf rust resistance Inra

High levels of variability for partial resistance to Melampsora larici-populina leaf rust in black poplar (Populus nigra L.) across Europe offers unique opportunity to identify favourable alleles through association studies.Nucleotide polymorphism (SNP and indels) within candidate genes has been explored in a discovery panel of 21 French P.nigra genotypes.Phenotyping for field and laboratory resistance components revealed significant diversity at both the intra and inter-metapopulation levels, with a suspected North vs. South geographical contrast.
