Abiotic environmental conditions for germination and development of gametophytes of Cyathea phalerata Mart. (Cyatheaceae) Acta Botanica
Marcon,Catiuscia; Silveira,Tatieli; Schmitt,Jairo Lizandro; Droste,Annette.
ABSTRACT In order to successfully establish themselves in their natural environment, ferns need habitats with abiotic conditions that are suitable for spore germination and gametophyte development. The objective of this study was to assess the influence of abiotic factors on the initial development of Cyathea phalerata cultivated in vitro. Spore germination and gametophyte development were assessed under varying conditions of surface sterilization, pH, temperature and photoperiod. Exogenous contamination was eliminated by sterilizing spores with 2.5 % NaClO for 15 min and sowing them into a culture medium supplemented with nystatin. Spores germinated at all pHs tested. Gametophytic development was faster in acidic pHs. Cultures at 25 °C exhibited the...

Asymbiotic culture of Cattleya intermedia Graham (Orchidaceae): the influence of macronutrient salts and sucrose concentrations on survival and development of plantlets Acta Botanica
Sasamori,Márcio Hisayuki; Endres Júnior,Delio; Droste,Annette.
ABSTRACTCattleya intermediais an Atlantic Forest species endemic to Brazil that is classed as vulnerable on the list of threatened species. In this study, C. intermedia plantlets were micropropagated in an asymbiotic culture and the influence of different concentrations of sucrose (15, 30, 45 and 60 g L-1, plus a zero sucrose medium) and macronutrient salts (complete Murashige and Skoog (MS) medium and half MS medium (with half-strength macronutrients)) on survival and development of the plantlets was evaluated. In all treatments 100% plantlet survival was achieved. The integrated analysis of height of aerial part, number of leaves per plantlet, fresh mass, number of roots per plantlet and length of the longest root showed that the plantlets exhibited...

Carbon sources and polyethylene glycol on soybean somatic embryo conversion PAB
Körbes,Ana Paula; Droste,Annette.
Somatic embryogenesis is an efficient method for the production of target cells for soybean genetic transformation. However, this method still offers low percentages of plant regeneration, and perhaps is related to the maturation process and high morphological abnormalities of the matured embryos. This study aimed to identify a maturation medium that could contribute to the outcome of more efficient plant regeneration results. Embryogenic clusters, derived from cotyledons of immature seeds of the soybean cultivars Bragg and IAS5, were used as starting material for embryos development. Different maturation media were tested by using 6% maltose, 3% sucrose or 6% sucrose, combined with or without 25 g L-1 of the osmotic regulator polyethylene glycol...

Genotoxicity biomonitoring of sewage in two municipal wastewater treatment plants using the Tradescantia pallida var. purpurea bioassay Genet. Mol. Biol.
Thewes,Márcia Regina; Endres Junior,Delio; Droste,Annette.
The genotoxicity of untreated and treated sewage from two municipal wastewater treatment plants (WTP BN and WTP SJN) in the municipality of Porto Alegre, in the southern Brazilian state of Rio Grande do Sul, was evaluated over a one-year period using the Tradescantia pallida var. purpurea (Trad-MCN) bioassay. Inflorescences of T. pallida var. purpurea were exposed to sewage samples in February (summer), April (autumn), July (winter) and October (spring) 2009, and the micronuclei (MCN) frequencies were estimated in each period. The high genotoxicity of untreated sewage from WTP BN in February and April was not observed in treated sewage, indicating the efficiency of treatment at this WTP. However, untreated and treated sewage samples from WTP SJN had high...

In vitro culture of Vriesea gigantea and Vriesea philippocoburgii: two vulnerable bromeliads native to Southern Brazil BABT
Droste,Annette; Silva,Anelise Machado da; Matos,Adriana Vieira; Almeida,Júlia Winck de.
Micropropagation studies were carried out using the seeds for establishing an in vitro culture of Vriesea gigantea and Vriesea philippocoburgii. Germination rate of V. gigantea was higher than of V. philippocoburgii. Plantlets of V. philippocoburgii gave rise to many adventitious shoots when cultivated in Knudson basal medium. In contrast, for V. gigantea, a higher salts-concentration was needed, so that the number of shoots was increased by Murashige and Skoog medium. Addition of activated charcoal and naphthaleneacetic acid in regeneration medium allowed the growth of shoots and the formation of roots, confirming the success of in vitro culture. The differences in expression of the genotypes reinforce the need of more research in order to set up the...

Influence of antibiotics on embryogenic tissue and Agrobacterium tumefaciens suppression in soybean genetic transformation Bragantia
Wiebke,Beatriz; Ferreira,Fabricio; Pasquali,Giancarlo; Bodanese-Zanettini,Maria Helena; Droste,Annette.
The influence of different antibiotic treatments in soybean genetic transformation was evaluated. First, an assay was performed to verify how different antibiotic treatments affect soybean embryogenic tissues. The effect of carbenicillin at 500 mg L-1 was genotype-dependent. This antibiotic did not affect embryo survival of cv. IAS5, but a three-fold increase of embryo proliferation was observed for cv. Bragg, when compared to the control. On the other hand, cefotaxime at 350 and 500 mg L-1 caused death of embryogenic tissues of both cultivars. Finally, the association of cefotaxime (250 mg L-1) + vancomycin (250 mg L-1) did not affect negatively the somatic embryos of tested cultivars until 63 days of treatment. Thereafter, a second experiment was carried...

Megaspore germination and initial development of Regnellidium diphyllum Lindman (Pteridophyta, Marsileaceae) sporophytes in the presence of cadmium Rev. Bras. Bot.
Wunder,Denise A.; Droste,Annette; Windish,Paulo G..
Regnellidium diphyllum has its distribution restricted to Southern Brazil and adjoining localities in Uruguay and Argentina. Currently it is on the list of threatened species of Rio Grande do Sul. The conversion of wetlands into agricultural areas or soil contamination by the introduction of waste products and fertilizers may compromise the establishment and survival of this species. Among the pollutants are heavy metals, such as cadmium (Cd). Megaspores were germinated in liquid culture medium, with concentrations 0 (control), 0.39; 0.78; 1.56; 3.12; 6.25; 12.5; 25; 50 and 100 mg L-1 of Cd, starting from a standard solution of Titrisol® at 1000 mg L-1. The increase of Cd in the growth medium to 50 mg L-1 resulted in low germinability (58%), and no...

Regeneration of soybean via embryogenic suspension culture Scientia Agricola
Droste,Annette; Leite,Paulo César Pimentel; Pasquali,Giancarlo; Mundstock,Elsa Cristina de; Bodanese-Zanettini,Maria Helena.
In an attempt to establish an alternative plant regeneration system for soybean [Glycine max (L.) Merrill] cultivars used in Brazilian breeding programs, ten genotypes were tested for their embryogenic potential. Cotyledons were removed as explants from immature seeds harvested from field-grown plants. After 45 days on induction medium, the number of responding cotyledons and the number of somatic embryos per immature cotyledon were evaluated. The percentage of explants that produced somatic embryos varied from 1 to 70% among cultivars. The average number of somatic embryos produced per cotyledon pair ranged from 0.01 to 10.3 with a mean of 3.4. Suspension cultures were initiated with three Agrobacterium tumefaciens susceptible cultivars. Suspensions were...

Screening of Brazilian soybean genotypes with high potential for somatic embryogenesis and plant regeneration PAB
Droste,Annette; Silva,Anelise Machado da; Souza,Iliane Freitas de; Wiebke-Strohm,Beatriz; Bücker-Neto,Lauro; Bencke,Marta; Sauner,Márcia Viviane; Bodanese-Zanettini,Maria Helena.
The aim of this work was to identify Brazilian soybean (Glycine max) genotypes with potential to respond to in vitro culture stimuli for primary somatic embryo induction, secondary embryo proliferation and plant regeneration. Differences among eight tested cultivars were observed at each stage. Two cultivars, IAS-5 and BRSMG 68 Vencedora, were selected for the evaluation of the capacity for embryo differentiation and plant regeneration. These cultivars had high embryo induction frequencies, repetitive embryogenic proliferation, and low precocious embryo germination in the initial experiment. The effect of abscisic acid (ABA) and charcoal addition on plant regeneration was investigated. The addition of ABA to proliferation medium and of ABA and activated...

Survival and development of reintroduced Cattleya intermedia plants related to abiotic factors and herbivory at the edge and in the interior of a forest fragment in South Brazil Acta Botanica
Endres Júnior,Delio; Sasamori,Márcio Hisayuki; Schmitt,Jairo Lizandro; Droste,Annette.
ABSTRACT Biotic and abiotic factors, such as luminosity, temperature, air humidity, and herbivory, can affect the establishment of reintroduced plants in natural habitats. This study evaluated the effects of these factors on the survival and growth of Cattleya intermedia plants reintroduced into a forest fragment in South Brazil. Plants of C. intermedia were obtained from in vitro seed germination in asymbiotic culture. Eighty-eight plants were reintroduced at both the forest edge and forest interior. Plants with greater shoot heights and number of leaves and pseudobulbs suffered more damage from herbivores at the edge. There were no significant differences in morphometric parameters between damaged and non-damaged plants in the interior. Tenthecoris...
