Brucella canis infection in dogs attended in veterinary clinics from patos, Paraíba state, Brazil BJM
Fernandes,Annielle Regina Fonseca; Azevedo,Sérgio Santos de; Piatti,Rosa Maria; Pinheiro,Eliana Scarcelli; Genovez,Margareth Élide; Azevedo,Adílio Santos de; Batista,Carolina de Sousa Américo; Alves,Clebert José.
To determine the frequency of anti-Brucella canis antibodies in dogs attended in veterinary clinics from Patos, Paraíba State, Brazil, as well as to identify risk factors and to isolate and identify the agent, 193 dogs were used. Agar gel immunodiffusion test (AGID) was used to detect B. canis antibodies in sera. Isolation of B. canis was carried out in blood and bone marrow from seropositive animals. Six animals tested seropositive in AGID, resulting in a frequency of 3.11%. B. canis was isolated from bone marrow of one seropositive animal, with confirmation by PCR. Lack of cleaning of the dog's environment was identified as risk factor (odds ratio = 7.91). This is the first report of isolation of B. canis in dogs from the Northeast region of Brazil.

Comparison of electrophoretic protein profiles of Campylobacter jejuni subsp. jejuni isolated from different animal species BJM
Scarcelli,Eliana; Costa,Elizabeth Oliveira da; Genovez,Margareth Élide; Cardoso,Maristela Vasconcellos; Bach,Erna Elizabeth; Torres,Ana Paula.
Electrophoretic protein profiles of Campylobacter jejuni subsp. jejuni strains isolated from feces of seven animal species, including man, were compared. Fourteen strains (two from each species) plus two human strains and the reference one, were ruptured by ultrasound and their total soluble proteins were analyzed by SDS-PAGE technique in a 12% polyacrylamide gel with computerized densitometric reading by the molecular analyst software. All the strains had bands in common that correspond to 45 and 66 Kda molecular weight. The disagreement corresponded to a 97 to 200 Kda molecular weight region. From the 17 strains, 13 (76.5%), were classified as biotype I, three (17.6%) as biotype II and one (5.8%) as biotype III. Since protein extracts were obtained from...

Detection of Leptospira spp. in the semen and urine of bulls serologically reactive to Leptospira interrogans serovar hardjo BJM
Magajevski,Fernanda Senter; Girio,Raul Jose Silva; Mathias,Luis Antonio; Myashiro,Simone; Genovez,Margareth Élide; Scarcelli,Eliana Pinheiro.
This study evaluated PCR for the detection of leptospires in semen and urine of ten serologically reactive bulls, comparing these results with those obtained with other diagnosis techniques. Two collections of materials were done in alternate days. Semen and urine samples were separated in aliquots for: direct visualization in dark field microscopy; inoculation in hamsters (for semen only); isolation in culture media; and PCR. No hamster was positive by the microscopic agglutination test (MAT); kidney and liver fragments from the hamsters were used in an isolation attempt in culture media, with one positive isolation from the kidney of a hamster inoculated with semen of one bull, and from liver of hamsters inoculated with semen of three bulls. Isolation in...

Evaluation of the counterimmunoelectrophoresis reaction as a serodiagnosis test for swine leptospirosis BJM
Genovez,Margareth Élide; Yasuda,Paulo Hideki; Vasconcellos,Silvio Arruda; Scarcelli,Eliana; Cardoso,Maristela Vasconcellos; Girio,Raul da Silva.
The Counterimmunoelectrophoresis reaction (CIE) was tested as a genus specific test for swine leptospirosis diagnosis using three soluble extracts obtained from Leptospira sp, serovars pomona, icterohaemorrhagiae and patoc. The extracts were treated with hot Triton X-100 and applied to serum samples of animals divided in three groups: Group 1, 10 swines experimentally infected with the Pomona strain; Group 2, 50 naturally infected swines and Group 3, 10 swines control. These animals were serologically evaluated by CIE and by the Microscopic Agglutination Test (MAT), the WHO reference technique. Groups 1 and 3 were monitored during a period of 93 days after infection (a.i.). Group 1 serum convertion took place around the 10th day a.i. by MAT but ocurred...

Leptospira spp detection by Polymerase Chain Reaction (PCR) in clinical samples of captive black-capped Capuchin monkey (Cebus apella) BJM
Scarcelli,Eliana; Piatti,Rosa Maria; Fedullo,José Daniel Luzes; Simon,Faiçal; Cardoso,Maristela Vasconcellos; Castro,Vanessa; Miyashiro,Simone; Genovez,Margareth Élide.
Leptospirosis is a widely distributed zoonosis that affects domestic and wild animals, and that has the man as the end point of its epidemiological chain. Leptospirosis diagnosis in primates is more difficult than in other animal species, as clinical signs and lesions are less evident and antibody response is detected only for short periods. The aim of this article was to describe the detection of Leptospira spp using polymerase chain reaction (PCR), in clinical samples from one captive black-capped Capuchin monkey (Cebus apella), which presented characteristics compatible with leptospirosis (jaundice and haemorrhagic kdney) in the macroscopic post-mortem examination. A friable kidney fragment and urine sample were cultured and submitted to experimental...

Molecular subtyping of Campylobacter jejuni subsp. jejuni strains isolated from different animal species in the state of São Paulo, Brazil BJM
Scarcelli,Eliana; Piatti,Rosa Maria; Harakava,Ricardo; Miyashiro,Simone; Fernandes,Flora Maria de Campos; Campos,Fabíola Ribeiro; Francisco,Waldemar; Genovez,Margareth Élide; Richtzenhain,Leonardo José.
The objective of the present trial was to characterize genetically strains of Campylobacter jejuni subsp. jejuni isolated from humans and several animal sources (bovines, swine, dogs, primates, wild boars and poultry). A total of 828 different animal samples (feces, carcass, aborted fetus and hysterectomized uterus) were analysed by means of routine bacteriological methods, and 36 C. jejuni strains were isolated. Thirty strains of human fecal origin were obtained in clinical analysis laboratories in the city of São Paulo. The 66 C. jejuni strains isolated were submitted to genetic characterization. Primers based on fla A gene were used in a polymerase chain reaction (PCR) and amplified a fragment of the 702 bp. PCR products were evaluated by means of...
