The deposition of lignin during plant-pathogen interactions is thought to play a role in plant defence. However, the function of lignification genes in plant disease resistance is poorly understood. In this article, we provide genetic evidence that the primary genes involved in lignin biosynthesis in Arabidopsis, CAD-C and CAD-D, act as essential components of defence to virulent and avirulent strains of the bacterial pathogen Pseudomonas syringae pv. tomato, possibly through the salicylic acid defence pathway. Thus, in contrast with cellulose synthesis, whose alteration leads to an increase in disease resistance, alteration of the cell wall lignin content leads directly or indirectly to defects in some defence components.

Expression of a poplar cDNA encoding a ferulate-5-hydroxylase/coniferaldehyde 5-hydroxylase increases S lignin deposition in Arabidopsis thaliana Inra

Ferulate-5-hydroxylase or coniferaldehyde 5-hydroxylase (F5H or Cald5H; CYP84A1) is the enzyme responsible for the lasthydroxylation of the syringyl-type lignin precursors. A cDNA clone highly homologous to the Arabidopsis thaliana F5H/Cald5H cDNAwas identified during the random sequencing of a poplar (Populus trichocarpa) differentiating xylem cDNA library. Present as a multigenicfamily in poplar, this gene (PopF5H) was found to be highly expressed in lignified tissues supporting its role in lignin biosynthesis. Benzodioxane structures are produced in lignins of transgenic poplar plants deficient in COMT, an O-methyltransferase required to produce lignin syringyl units. They result from incorporation of 5-hydroxyconiferyl alcohol into the monomer supply and confirm that phenols other than the three traditional monolignols can be integrated into plant lignins.

Quantitative 31P NMR spectroscopy of lignins from transgenic poplars Inra

Milled wood lignins isolated from genetically modified poplar trees were studied by quantitative 31P NMR spectroscopy, in combination with thioacidolysis and mild alkaline hydrolysis. The genetictransformation of the trees included down regulation of CAD and COMT enzyme activities.These analyses confirmed that moderate CAD down-regulation does not substantially alter ligninstructure. In contrast, severe CAD deficiency alters the lignin structure more profoundly by decreasingthe syringyl/guaiacyl ratio and increasing the degree of condensation of the lignin. Themost severe alterations were observed in the lignins from the COMT-transformed lines.

Stability of transgene expression in poplar: A model forest tree species Inra

Notre étude a pour but d'évaluer la stabilité de l'expression d'un transgène chez un peuplier hybride (Populus tremula x P. alba) transformé avec le gène reporter uidA sous le contrôle soit du promoteur 35S, soit du promoteur du gène cad2 de l'eucalyptus (EuCAD) à spécificité vasculaire. L'expression du transgène a été suivie quantitativement par fluorimétrie et qualitativement par histochimie à partir de matériel collecté in vitro, en serre ou en champ sur quelques centaines d'arbres, issus de 44 lignées transgéniques. Les auteurs ont étudié l’effet combiné de deux gènes chez des peupliers hybrides (genre Populus), CP4 et GOX, qui confèrent la tolérance au glyphosate (l’ingrédient actif de l’herbicide Roundup©).
