Comparative sublethal toxicity of nine pesticides on olfactory learning performances of the honeybee Apis mellifera Inra
Decourtye, A.; Devillers, J.; Genecque, E.; Le Menach, K.; Budzinski, H.; Cluzeau, S.; Pham-Delègue, M.H..
Using a conditioned proboscis extension response (PER) assay, honeybees (Apis mellifera L.) can be trained to associate an odor stimulus with a sucrose reward. Previous studies have shown that observations of conditioned PER were of interest for assessing the behavioral effects of pesticides on the honeybee. In the present study, the effects of sublethal concentrations of nine pesticides on learning performances of worker bees subjected to the PER assay were estimated and compared. Pesticides were tested at three concentrations. The highest concentration of each pesticide corresponded to the median lethal dose value (48-h oral LD50), received per bee and per day, divided by 20. Reduced learning performances were observed for bees surviving treatment with...

Detection of early effects of a single herbicide (diuron) and a mix of herbicides and pharmaceuticals (diuron, isoproturon, ibuprofen) on immunological parameters of Pacific oyster (Crassostrea gigas) spat ArchiMer
Luna-acosta, Andrea; Renault, Tristan; Thomas-guyon, Helene; Faury, Nicole; Saulnier, Denis; Budzinski, Helene; Le Menach, K.; Pardon, P.; Fruitier-arnaudin, I.; Bustamante, Paco.
In the context of massive summer mortality events of the Pacific oyster Crassostrea gigas, the aim of this study was to investigate the early effects on genes, enzymes and haemocyte parameters implicated in immune defence mechanisms in C. gigas oysters exposed to a potentially hostile environment, i.e. to an herbicide alone or within a mixture. Following 2 h of exposure to the herbicide diuron at 1 mu g L-1 the repression of different genes implicated in immune defence mechanisms in the haemocytes and the inhibition of enzyme activities, such as laccase-type phenoloxidase (PO) in the plasma, were observed. The inhibition of superoxide dismutase (SOD) activity in the plasma was also observed after 6 and 24 h of exposure. In the mixture with the herbicides...

Influence of seep emission on the non-symbiont-bearing fauna and vagrant species at an active giant pockmark in the Gulf of Guinea (Congo-Angola margin) ArchiMer
Olu, Karine; Caprais, Jean-claude; Galeron, Joelle; Causse, R.; Von Cosel, R.; Budzinski, H.; Le Menach, K.; Le Roux, C.; Levache, D.; Khripounoff, Alexis; Sibuet, Myriam.
Detailed surveying with an ROV found that a dense and diverse cold-seep community colonises a giant pockmark located at 3200 m depth, 8 km north from the deep Congo channel. Several types of assemblages, either dominated by Mytilidae and Vesicomyidae bivalves or Siboglinidae polychaetes, are distributed on the 800-m diameter active area. The site is characterised by a most active central zone in a depression with abundant carbonate concretions and high methane fluxes where high-density clusters of mussels and siboglinids dominate. In contrast, the peripheral zones display large fields of dead and live vesicomyids on soft sediment, with a lower mean density and lower methane concentration in seawater. The associated megafauna includes Alvinocarididae...

Molecular diversity studies of bacterial communities of oil polluted microbial mats from the Etang de Berre (France) Inra
Hernandez-Raquet, G.; Budzinski, H.; Caumette, P.; Dabert, P.; Le Menach, K.; Muyzer, G.; Duran, R..
The biodiversity of microbial mats inhabiting the oil-contaminated lagoon Etang de Berre was determined by molecular approaches. The fingerprint of denaturing gradient gel electrophoresis (DGGE) and automatic ribosomal intergenic spacer analysis (ARISA) of mats exposed to different pollution levels showed specific microbial communities for each site but similar diversity richness. Species composition of the mats were compared by constructing 16S rRNA libraries. Amplified rDNA restriction analysis (ARDRA) of clone libraries confirmed their similar level of diversity richness. Phylogenetic analysis of the 16S rRNA sequences showed that the classes γ and α of Proteobacteria were abundantly present in both sites whereas phylotypes related to the...

PAH metabolites in fish bile: from the Seine Estuary to Iceland ArchiMer
Kammann, U.; Akcha, Farida; Budzinski, H.; Burgeot, Thierry; Gubbins, M. J.; Lang, T.; Le Menach, K.; Vethaak, A. D.; Hylland, K..
Polycyclic aromatic hydrocarbons (PAH) are environmental contaminants that pose significant risk to health of fish. The International Workshop on Integrated Assessment of Contaminant Impacts on the North Sea (ICON) provided the framework to investigate biomarker responses as well as contaminant concentrations side by side in marine ecosystems. Concentrations of the main PAH metabolites 1-hydroxypyrene, 1-hydroxyphenanthren and 3-hydroxybenzo(a)pyrene were determined in bile by HPLC with fluorescence detection. Fish species under investigation were dab (Limanda limanda), flounder (Platichthys flesus) and haddock (Melanogrammus aeglefinus). A contamination gradient was demonstrated from the low contaminated waters of Iceland and off-shore regions of the...

Relationship between PAH biotransformation as measured by biliary metabolites and EROD activity, and genotoxicity in juveniles of sole (Solea solea) ArchiMer
Wessel, Nathalie; Santos, Raphael; Menard, Dominique; Le Menach, K.; Buchet, Vincent; Le Bayon, Nicolas; Loizeau, Veronique; Burgeot, Thierry; Budzinski, H.; Akcha, Farida.
Polycylic aromatic hydrocarbons (PAHs) are ubiquitous contaminants in the marine environment Their toxicity is mainly linked to the ability of marine species to biotransform them into reactive metabolites PAHs are thus often detected at trace levels in animal tissues For biomonitoring purposes this findings have two main consequences (i) the determination of the PAH tissue concentration is not suitable for the evaluation of individual exposure to PAHs (ii) It can explain sometimes the lack of correlations obtained with relevant markers of toxicity such as genotoxicity biomarkers The aim of the present study was to better investigate the link between PAH exposure and genotoxicity in marine flatfish During a laboratory experiment juvenile soles were exposed...
