Comparative QTL mapping for adaptive traits among populations and species can lead to the identification of genomic regions conserved over a long evolutionary period and therefore potentially harboring genes of great importance for adaptive processes. Genome-wide analysis of LIM gene family in Populus trichocarpa, Arabidopsis thaliana and Oryza sativa Inra

In Eukaryotes, LIM proteins act as developmental regulators in basic cellular processes such as regulating the transcription or organizing the cytoskeleton. The LIM domain protein family in plants has mainly been studied in sunflower and tobacco plants, where several of its members exhibit a specific pattern of expression in pollen. In this paper, wefinely characterized in poplar six transcripts encoding these proteins. In Populus trichocarpa genome, the 12 LIMgene models identified all appear to be duplicated genes. Identification of the structure and origin of thioacidolysis marker compounds for cinnamyl alcohol dehydrogenase deficiency in angiosperms Inra

Molecular marker compounds, derived from lignin by the thioacidolysis degradative method, for cinnamyl alcohol dehydrogenase (CAD) deficiency in angiosperms have been structurally identified as indene derivatives. They are shown to derive from hydroxycinnamyl aldehydes that have undergone 8-O-4-cross-coupling during lignification. As such, they are valuable markers for ascertaining plant responses to various levels of CAD down-regulation. Their derivation illustrates that hydroxycinnamyl aldehydes incorporate into angiosperm lignins by endwise coupling reactions in much the same way as normal monolignols do, suggesting that the hydroxycinnamyl aldehydes should be considered authentic lignin precursors.

Les biotechnologies chez les arbres forestiers Inra

Le développement actuel des biotechnologies ouvre des potentialités importantes en matière d'amélioration génétique des arbres forestiers. Notamment, celles-ci devraient permettre de diffuser rapidement les nouvelles variétés aux performances accrues nécessaires au maintien de la compétitivité de la filière-bois. Cette revue fait le point sur les possibilités et les limitations de l'utilisation des biotechnologies chez les arbres forestiers:multiplication végétative in vitro par embryogénèse somatique et par microbouturage, transformation génétique et sélection assistée par marqueurs.

Quantitative 31P NMR spectroscopy of lignins from transgenic poplars Inra

Milled wood lignins isolated from genetically modified poplar trees were studied by quantitative 31P NMR spectroscopy, in combination with thioacidolysis and mild alkaline hydrolysis. The genetictransformation of the trees included down regulation of CAD and COMT enzyme activities.These analyses confirmed that moderate CAD down-regulation does not substantially alter ligninstructure. In contrast, severe CAD deficiency alters the lignin structure more profoundly by decreasingthe syringyl/guaiacyl ratio and increasing the degree of condensation of the lignin. Themost severe alterations were observed in the lignins from the COMT-transformed lines.

Stability of transgene expression in poplar: A model forest tree species Inra

Notre étude a pour but d'évaluer la stabilité de l'expression d'un transgène chez un peuplier hybride (Populus tremula x P. alba) transformé avec le gène reporter uidA sous le contrôle soit du promoteur 35S, soit du promoteur du gène cad2 de l'eucalyptus (EuCAD) à spécificité vasculaire. L'expression du transgène a été suivie quantitativement par fluorimétrie et qualitativement par histochimie à partir de matériel collecté in vitro, en serre ou en champ sur quelques centaines d'arbres, issus de 44 lignées transgéniques. Wood formation in Angiosperms Inra

La formation du bois est un processus de développement complexe, impliquant cinqétapes majeures : (1) la division des cellules à partir d’un méristème secondaire, appelé le cambium; (2) l’élongation des cellules ; (3) le dépôt d’une paroi secondaire épaisse ; (4) la mort programmée des cellules ; et (5) leur imprégnation par des composés phénoliques conduisant à la formation du bois de coeur. Grâce au développement des techniques de génomique et aux travaux menés sur la caractérisation d’arbres transgéniques, certains mécanismes moléculaires impliqués dans la formation du bois commencent à être mieux connus. Dans cette revue, deux aspects importants pour la qualité du bois seront approfondis : la lignification et le contrôle de l’angle des microfibrilles...
