tThe regular water supply during life cycle is essential to determine the yield in the legumes. In semiaridenvironments, the irregular rainfall and high temperature influence the phenology of crops impairing theyield. Peanut is an oleaginous crop with broad adaptation to tropical and semiarid climates, but yield isoften harmed when plants face water irregularities during reproductive phase. Essential plant oils contain biopesticides that could be used to control many crop pests. Tetranychus spp. are mites that cause damage to several crops and are primarily controlled by synthetic pesticides. Literature showed that mites can be controlled with essential oils of plants containing eugenol. In this work, we evaluated the bioactivity of basil (Ocimum basicilum) accessions for peanut-spider mites control based on molecular, biochemical and agronomic assays. Zygotic and embryonic cotton structures were used in in vitro procedures to obtain stable plants. Zygotes (24, 48, and 72 h) and immature embryos (10 and 25 days after fertilization) were grown in MS medium supplemented with glucose, indoleacetic acid, kinetin, and activated charcoal. Regenerated seedlings were acclimatized and grown in 10-kg pots for further phenological evaluation, based on agronomic traits and possible occurrence of somaclonal effects. We found that zygotes and immature embryos at 10 days did not develop physiological competence to allow regeneration of plants, based on adopted procedures. Gene sequences previously identified in Arabidopsis buds were used as references in order to estimate temporal and tissue expression in buds, leaves, stem, and root tissues in cotton plants. Buds were evaluated during 3 phases: 2-8, 10-12, and 14-20 mm. Primers were designed for the ARF6, ATFY, and SEUSS genes for use in semi-quantitative reverse transcription-polymerase chain reaction and quantitative reverse transcription-polymerase chain reaction. Different levels of expression of the 3 genes were confirmed in cotton buds as well as in other tissues.
