A method for DNA sequencing of ribosomal RNA (rRNA) genes, amplified by polymerase chain reaction (PCR), using internal primers, designed on the basis of conserved regions of rRNA genes for determining a near complete sequence (99%) of the gene using an automated DNA sequencer (Applied Biosystem Incorporation, USA) is described. The procedure is extremely rapid as cloning of the gene is not required for sequence determination. In addition time consuming steps such as ethanol precipitation and hazardous steps such as phenol/chloroform extractions are excluded from the protocol for the purification of extension products after Taq cycle sequencing using the ABI dye terminator chemistry. A novel anaerobic bacterial strain, ST07-YET, was isolated from a carbonate chimney of the Prony Hydrothermal Field (PHF) in New Caledonia. Cells were Gram-stain-positive, straight rods (0.7-0.8 x 3.0-5.0 mu m) and motile by means of lateral flagella. Strain ST07-YET was mesophilic (optimum 35 degrees C), moderately alkaliphilic and halotolerant (optimum pH 8.7 and 5 gr l(-1) NaCl). Elemental sulfur, sulfate, thiosulfate, sulfite, nitrate and nitrite were not used as terminal electron acceptors. A strictly anaerobic, slightly halophilic and moderately thermophilic, sporulating rod designated strain DVird3(T) was isolated from deep-sea hydrothermal vent samples collected at a depth of approximately 800 m on the Atlantic Ocean Ridge. Strain DVird3(T) possessed a few laterally inserted flagella, had a DNA G+C content of 33.1 mol% and grew optimally at pH 6.6 and at 45 degreesC. Growth was observed at temperatures between 20 and 58 degreesC and at pH values between 5.8 and 8.2. The microflora developing during a continuous enrichment culture from a hydrothermal chimney sample was investigated by molecular methods. The culture was performed in a gas-lift bioreactor under anaerobic conditions, at 90 degrees C and pH 6.5, on a complex medium containing sulfur as the terminal electron acceptor. Archaeal and bacterial diversity was studied. Microorganisms affiliated with the genera Pyrococcus, Marinitoga, and Bacillus were detected through DGGE analysis of 16S rDNA. A strictly anaerobic, thermophilic, gram-positive, spore-forming eubacterium designated strain SEBR 5268 was isolated from an oil field at a depth of 2,100 m, where the temperature was 92°C. The cells of this organism were gram-positive, straight, motile rods (0,5 by 2 to 3 micrometers) with peritrichous flagella. The cells occurred singly or in pairs during the logarithmic growth phase, but were pleomorphic and filamentous (length, 15 micrometers) in old cultures. A novel Gram-negative, non-spore-forming, vibrio-shaped, anaerobic, alkaliphilic, sulfate-reducing bacterium, designated strain PAR22NT, was isolated from sediment samples collected at an alkaline crater lake in Guanajuato (Mexico). Strain PAR22NT grew at temperatures between 15 and 37 °C (optimum, 32 °C), at pH between pH 8.3 and 10.1 (optimum, pH 9.0–9.6), and in the presence of NaCl up to 10 %. #Thermoanaerobacter finnii$ fermente le glucose et le xylose en lactate, acétate, H2, CO2 et éthanol qui est le produit majeur du métabolisme. Une autre bactérie du genre #Thermoanaerobacter$ isolée d'une eau de gisement pétrolier produit des quantités aussi importantes d'éthanol que de lactate à partir de ces sucres. We found that #Thermosipho africanus$ was able to ferment D-glucose, D-ribose, maltose, and starch, while D-galactose, fructose, and sucrose were utilized poorly. Acetate, H2, and CO2, as well as small amounts of ethanol and lactate, were end products of glucose metabolism in this organism. The presence of thiosulfate as an electron acceptor greatly improved growth and increased acetate production from the sugars. The genus #Thermosipho$ is the only genus in the order #Thermotogales$ that has been described as a non-carbohydrate fermenter. We propose that the description of the genus #Thermosipho$ be emended because the only species in this genus, #T. africanus$, is a carbohydrate fermenter that is able to utilize thiosulfate as an electron acceptor.... |
