An RNA extraction method with high integrity and purity as well as the selection of adequate reference genes are prerequisites for gene expression analysis. For common bean seeds, there is no well-defined protocol that can be used in a laboratory routine for gene expression analysis. In this study, an extraction protocol for RNA from common bean seeds, which produced material with good integrity for qPCR (RIN ≥ 6.5), was optimized. In addition, 10 reference genes were evaluated under qPCR standard conditions using different tissue samples of common beans. Carioca beans are subjected to the hard-to-cook phenomena (HTC), which results in loss of technological and sensorial qualities. To better elucidate this postharvest hardening process, changes in technological and biochemical properties of grains during storage at 40 °C and 75% relative humidity were investigated, and the results were analyzed using Principal Component Analysis (PCA). Results showed that the hardening process occurs at different intensity for each genotype, and those more susceptible to HTC presented higher water absorption index (WAI) and peroxidase activity. O fenômeno hard-to-cook promove a diminuição no amaciamento por cocção, alteração na palatabilidade e diminuição na disponibilidade de proteínas e carboidratos, prejudicando a comercialização dos grãos. Drought stress, particularly during the flowering and grain-filling stages of growth, contributes to serious yield loss in common bean (Phaseolus vulgaris L.). The aim of this study was to identify genes induced in response to drought stress using transcriptome analysis of contrasting genotypes. O objetivo desse trabalho foi quantificar a expressão de genes de arroz homólogos de Arabidopsis previamente relacionados ao aumento do potencial produtivo (RUBIRICE, RUBICORN, AVP1, DA1 e TOR) por meio da análise de PCR quantitativo (qPCR). The identification and validation of candidate genes related to traits of interest is a time consuming and expensive process and the homology among genes from different species can facilitate the identification of genes of the target species from the genomic information of a model species. This study aimed to quantify the expression of homologous rice genes previously related to drought tolerance in Arabidopsis. Genes related to the response to drought stress in leaf and root tissue of drought-susceptible (DS) and tolerant (DT) genotypes were characterized by RNA-Seq. In total, 54,750 transcripts, representative of 28,590 genes, were identified; of these, 1,648 were of high-fidelity (merge of 12 libraries) and described for the first time in the Andean germplasm. From the 1,239 differentially expressed genes (DEGs), 458 were identified in DT, with a predominance of genes in categories of oxidative stress, response to stimulus and kinase activity. New carioca bean cultivars are being introduced into the market necessitating their evaluation under trade conditions, which often require storage under ambient conditions. We therefore evaluated the darkening and hardening processes of six carioca bean genotypes each representing regular and slow darkening trait during storage under ambient conditions for five months to elucidate their relationship as a breeding strategy.
