A physiological and biochemical approach to selecting the ideal diet for Ostrea edulis (L.) broodstock conditioning (part A) ArchiMer
Gonzalez-araya, Ricardo; Queau, Isabelle; Quere, Claudie; Moal, Jeanne; Robert, Rene.
To select the best microalgae for Ostrea edulis conditioning, four single species diets were fed to batches of broodstock, which were then compared using physiological and biochemical approaches. Ingestion and absorption were first studied, showing that Chaetoceros gracilis and Skeletonema marinoï were efficiently ingested (4.9–5.3 mg g−1 h−1) and absorbed (1.9–2.5 mg g−1 h−1) while Tetraselmis suecica led to the lowest physiological responses (0.36 and 0.12 mg g−1 h−1 respectively). When O. edulis were fed any microalgae except T-ISO with only 79 mg g−1, gonads accumulated carbohydrates from 116 to 141 mg g−1 and the extent of accumulation depended on the species supplied in the order T. suecica<C. gracilis<S. marinoï. When oysters were fed either...

Anaesthesia and gonad sampling in the European flat oyster (Ostrea edulis) ArchiMer
Suquet, Marc; Gonzalez, Ricardo; Lebrun, Luc; Queau, Isabelle; Mingant, Christian; Robert, Rene.
Controlled reproduction of the European flat oyster requires the development of tools adapted to this species, including a practical anaesthesia and gonad sampling protocol to facilitate sex determination and the verification of gametogenesis. Three replicate groups of 10 oysters (mean weight +/- SD: 29.9 +/- 8.5 g) were anaesthetised in 5 L containers using magnesium chloride designed either for laboratory (Flucka (R)) or agricultural (Dead Sea Work (R): DSW) use, at two concentrations (50 or 72 g L-1). No significant differences were observed in the percentages of oysters anaesthetised or subsequent oyster mortality with the different anaesthetics or concentrations, but increasing water temperature from 14.9 to 18.8 degrees C significantly increased the...

Anesthesia in Pacific oyster, Crassostrea gigas ArchiMer
Suquet, Marc; De Kermoysan, Goulwen; Araya, Ricardo Gonzalez; Queau, Isabelle; Lebrun, Luc; Le Souchu, Pierrick; Mingant, Christian.
A reliable anaesthesia and sampling protocol for Pacific oysters will enable experiments to be conducted without sacrificing animals and will facilitate successive sampling of individuals for gametogenesis studies. As no such techniques were available for Crassostrea gigas, the present study aimed to define suitable anaesthetic conditions for use with this species. Three groups of ten oysters (mean weight +/- SD, 32.1 +/- 9.0 g) were anaesthetised in 5 L containers. Among different chemicals: benzocaine, eugenol and three different types of magnesium chloride (a laboratory one Flucka (R) - and two designed for agriculture - DEUSA International (R) and Dead SeaWorks (R)) and concentrations tested, one laboratory (concentration: 72 g L-1) and one...

Biological characteristics of sperm in European flat oyster ( Ostrea edulis ) ArchiMer
Suquet, Marc; Pouvreau, Stephane; Queau, Isabelle; Boulais, Myrina; Le Grand, Jacqueline; Ratiskol, Dominique; Cosson, Jacky.
n brooding mollusc species that incubate larvae before their release in seawater, the biology of gametes remains poorly explored. The present study describes some biological characteristics of sperm of the European flat oyster, a native species that has been over-exploited in the past and is nowadays classified as an endangered species in Europe. Flat oysters were collected by divers in the Rade de Brest (Finistère, France), during its natural reproduction period. Gonadal pH is acidic (6.31 ± 0.10). Spermatozoa are clustered in spermatozeugmata, an acellular structure in which the sperm heads are embedded. After their transfer in seawater, spermatozeugmata have a mean diameter of 64 ± 3 μm and they release free spermatozoa for a mean duration of...

Caractéristiques biologiques des gamètes et larves d'huître plate européenne (Ostrea edulis): données préliminaires ArchiMer
Suquet, Marc; Queau, Isabelle; Le Grand, Jacqueline; Ratiskol, Dominique; Pouvreau, Stephane.
The biology of gametes and larvae in brooding mollusc species remains little explored. The aim of this study is to present the results obtained in 2016, on biological characteristics of gametes and larvae of European flat oyster.       Measurements were carried out on 50 oysters, collected by divers in the Rade de Brest (Finistère, France), during the spawning period of this species. Gonad pH is acidic (6.58±0.54). After the dissociation of spermatozeugmata, an acellular structure to which the spermatozoa are clustered, the percentage of motile spermatozoa ranged from 20 to 34% and the Velocity of the Average Path from 70 to 85µm/s. Spermatozeugmata were shed after serotonin injection. Oocytes (mean diameter: 129.4±19.1µm) were rarely observed (2...

Effect of age and environmental conditions on the movement characteristics of Pacific oyster (Crassostrea gigas) trochophores ArchiMer
Suquet, Marc; Rimond, Flore; Cosson, J.; Wilson-leedy, J.; Lebrun, Luc; Queau, Isabelle; Mingant, Christian; Fauvel, Christian.
The movement characteristics of Pacific oyster trochophores have received very little coverage in the scientific literature. Described here are not only changes in the swimming characteristics of Crassostrea gigas trochophores (size: 53-77 mu m) in relation to time after fertilization and to inter-female variation but also the effects of salinity and pH on movement characteristics. The percentage of motile trochophores was measured on images obtained through a dissecting microscope and the Velocity Average Path (VAP) was assessed using a Computer Assisted Sperm Analysis (CASA) system. At 20 degrees C, the first movements of the trochophores were observed at 6.5 h post-fertilization. The mean (+/- SD) percentage of swimming trochophores and the VAP observed...

Metabolic adjustments in the oyster Crassostrea gigas according to oxygen level and temperature ArchiMer
Le Moullac, Gilles; Queau, Isabelle; Le Souchu, Pierrick; Pouvreau, Stephane; Moal, Jeanne; Le Coz, Jean-rene; Samain, Jean-francois.
The purpose of this study was to examine the responses of the oyster C. gigas to oxygen levels at subcellular and whole-organism levels. Two experiments were carried out. The first experiment was designed to measure at 15°C, 20°C and 25°C for 20 hours the clearance (CR) and oxygen consumption (OC) rates of oysters exposed at different concentrations of oxygen. The goal of this first part was to estimate the hypoxic threshold for oysters below which their metabolism shifts toward anaerobiosis, by estimating the oxygen critical point (PcO2) at 15, 20 and 25°C. The second experiment was carried out to evaluate the metabolic adaptations to hypoxia for 20 days at three temperatures: 12°C, 15°C and 20°C. The metabolic pathways were characterized by the...

Optimizing sperm cryopreservation in great scallop (Pecten maximus) ArchiMer
Suquet, Marc; Malo, Florent; Gourtay, Clemence; Ratiskol, Dominique; Riobe, Marion; Le Grand, Jacqueline; Queau, Isabelle; Mingant, Christian.
A basic cryopreservation protocol for great scallop sperm was recently published, with which thawed sperm survival remained limited to 25% of control values (Suquet et al. 2014). We designed here a set of five experiments to test ways of improving thawed sperm survival and storage by studying the effects of (1) cryoprotectant concentration; (2) sugar incorporation in the extender; (3) egg yolk addition to the extender; (4) sperm dilution rate; and (5) storage capacity of thawed sperm at 4 degrees C. A sixth experiment was then performed to compare rearing performances of larvae produced using thawed sperm from the optimal protocol defined in the present study with those produced using fresh sperm. During the first experiment, a significantly higher...

pH controls spermatozoa motility in the Pacific oyster ( Crassostrea gigas ) ArchiMer
Boulais, Myrina; Suquet, Marc; Arsenault-pernet, Marie Eve Julie; Malo, Florent; Queau, Isabelle; Pignet, Patricia; Ratiskol, Dominique; Le Grand, Jacqueline; Huber, Matthias; Cosson, Jacky.
Investigating the roles of chemical factors stimulating and inhibiting sperm motility is required to understand the mechanisms of spermatozoa movement. In this study, we described the composition of the seminal fluid (osmotic pressure, pH, and ions) and investigated the roles of these factors and salinity in initiating spermatozoa movement in the Pacific oyster. The acidic pH of the gonad (5.82 ± 0.22) maintained sperm in the quiescent stage and initiation of flagellar movement was triggered by a sudden increase of spermatozoa external pH (pHe) when released in seawater (SW). At pH 6.4, percentage of motile spermatozoa was three times higher when they were activated in SW containing 30 mM NH4Cl, which alkalinizes internal pH (pHi) of spermatozoa, compared...

Seasonal variation of sperm quality in Pacific oyster (Crassostrea gigas) ArchiMer
Suquet, Marc; Malo, Florent; Queau, Isabelle; Ratiskol, Dominique; Quere, Claudie; Le Grand, Jacqueline; Fauvel, Christian.
Seasonal changes of sperm quality which can affect sperm biological parameters throughout the breeding period, have been little studied in mollusc species. Controlling gamete quality would aid the management of gametes in hatcheries and the development of selection programs. The aim of the present study was to describe the changes in sperm quality of wild Pacific oysters through the spawning season by comparing sperm parameters at the beginning (May), middle (July) and end (October) of this period using a panel of bio-descriptors. These parameters were studied over the 2014 breeding season based on shed sperm collected after serotonin injection of wild breeders. A significantly higher percentage of motile sperm was observed at the end of the spawning...

The quality of oyster spermatozoa has become an issue for modern aquaculture. However, reliable protocols used to assess gamete quality are lacking. The aim of this work is to define a standardised experimental fertilisation protocol for Pacific oyster oocytes. Six experiments have been carried out in this study. The optimal conditions for Pacific oyster experimental fertilisation have been defined: (1) oocytes can be conserved in seawater for at least 4 h before fertilisation. (2) oocyte concentration: 100-1000 ml(-1) seawater, (3) fertilisation volume: 10-100 nil, (4) spermatozoa: oocyte ratio: 400 to maintain discriminating conditions, (5) gamete contact time: longer than 10 mill and (6) fertilisation and incubation temperatures: both 19 degrees C. Setting of a procedure for experimental incubation of Pacific oyster (Crassostrea gigas) embryos ArchiMer
Suquet, Marc; Amourda, Christopher; Mingant, Christian; Queau, Isabelle; Lebrun, Luc; Brizard, Raphael.
Standardized experimental protocols designed to study Pacific oyster (Crassostrea gigas) gamete quality have not been previously published. Gamete sampling variations and confounding effect of interacting factors result in large variations between replicates, decreasing the effects of the studied factors. This work aims at defining a standardized procedure for incubation of oyster embryos designed for experimental purposes. In a first phase, four experiments were developed to improve embryo sampling and handling. They showed that a minimum of 50 embryos must be counted to decrease the variation between counting replicates. For reliable results, sampling must be carried out from 0 to 7 min after a careful agitation of seawater containing embryos...

Suquet, Marc; Labbe, Catherine; Puyo, Sophie; Mingant, Christian; Quittet, Benjamin; Boulais, Myrina; Queau, Isabelle; Ratiskol, Dominique; Diss, Blandine; Haffray, Pierrick.
This study is the first demonstration of successful post-thawing development to reproduction stage of diploid cryopreserved larvae in an aquatic invertebrate. Survival, growth and reproductive performances were studied in juvenile and adult Pacific oysters grown from cryopreserved embryos. Cryopreservation was performed at three early stages: trochophore (13 +/- 2 hours post fertilization: hpf), early D-larvae (24 +/- 2 hpf) and late D-larvae (43 +/- 2 hpf). From the beginning (88 days) at the end of the ongrowing phase (195 days), no mortality was recorded and mean body weights did not differ between the thawed oysters and the control. At the end of the growing-out phase (982 days), survival of the oysters cryopreserved at 13 +/- 2 hpf and at 43 +/- 2 hpf...

Harmful algal blooms are a threat to aquatic organisms and coastal ecosystems. Among harmful species, the widespread distributed genus Alexandrium is of global importance. This genus is well-known for the synthesis of paralytic shellfish toxins which are toxic for humans through the consumption of contaminated shellfish. While the effects of Alexandrium species upon the physiology of bivalves are now well documented, consequences on reproduction remain poorly studied. In France, Alexandrium minutum blooms have been recurrent for the last decades, generally appearing during the reproduction season of most bivalves including the oyster Crassostrea gigas. These blooms could not only affect gametogenesis but also spawning, larval development or juvenile...
