in mollusc hatcheries. Consequently, we assessed themost commonly used species to establish their growth and size profiles, their cytomorphological, biochemical, biomolecular identification as well as their cytometer signs, and also their ability to grow under extreme conditions of temperature and salinity. This work aimed at matching the conformity of the strains used in Argenton (dispatched to most of the French commercial hatcheries) to those of a referenced culture collection. A previous screening preceded this study, because of the 45 species and/or clones held originally, Argenton kept no more than 15 species. As most of the latter are also represented in referenced collection, our strains are therefore correctly identified. On the other hand, the... |

This study investigated the non-neutrality of genetic polymorphism in two alpha-amylase genes (AMYA and AMYB) in the oyster Crassostrea gigas. Bi-parental oyster families, bred to be polymorphic for markers in these genes, were monitored for growth and survival for 1 year under standard culture conditions in two French production sites. Within-family genotype frequencies indicated that the two amylase genes were closely linked (c. 1.7 cM). Within two of three families, significant differences in growth were observed between genotypes at one of the two production sites, suggesting that this polymorphism is not neutral and might be under selection because of its role in digestive function. Estimated daily yields were different between amylase genotypes,... |

Pacific oyster hatchery production remains highly variable, partially due to inconsistent gamete quality. However, little attention has been paid to oocyte quality in Pacific oyster compared to important vertebrate aquaculture species such as fishes. The aims of the present study were to identify criteria of oocyte and trochophore quality used as predictive tools for embryo developmental success at the D-larval stage. Twenty-six potential proxies of oocyte and trochophore quality were studied on 25 mature females, as determined using histology. Some morphological and biochemical characteristics of mature oocytes were described. Protein appeared to be the main constituent of fully mature Pacific oyster oocytes. Neutral lipids accounted for 61% of the total... |

Fertilization of the oocyte is a critical process of sexual reproduction depending among other factors on spermatozoa functionality. Mitochondria participate in many crucial processes for spermatozoa motility and fertilizing ability. The Pacific oyster, Crassostrea gigas, is characterized by a high inter-individual variability of its reproductive success which has been shown to be partially due to the variability of gamete quality. The present study explored spermatozoa characteristics such as: i) relationships among mitochondrial functionality (ATP content, mitochondrial membrane potential (MMP), reactive oxygen species production); sperm viability (percentages of live, dying, and dead); and motility; and ii) their involvement in inter-oyster variability... |

Effects of simultaneous exposure of Pacific oyster, Crassostrea gigas, to both a harmful dinoflagellate that produces Paralytic Shellfish Toxins (PST), Alexandrium minutum, and cadmium (Cd) and copper (Cu), were assessed. Oysters were exposed to a mix of Cd-Cu with two different diets (i.e. A. minutum or Tisochrysis lutea) and compared to control oysters fed A. minutum or T. lutea, respectively, without metal addition. Metals and PST accumulations, digestive gland lipid composition, and cellular and biochemical hemolymph variables were measured after 4 days of exposure. Oysters exposed to Cd-Cu accumulated about thirty-six times less PSTs than oysters exposed to A. minutum alone. Exposure to Cd-Cu induced significant changes in neutral lipids (increase in... |

Changes in sperm features during the movement phase are especially interesting to study in external fertilization species whose sperm duration movement is long because this implies a significant adaptation of moving cells to the external medium. This study describes the changes in tetraploid Pacific oyster sperm characteristics in relation to time post activation. Sperm individually collected on three tetraploid males were activated in seawater. Their features were analysed over a 24 h period and compared to a sperm pool collected on three diploid males as a reference. The percentage of motile spermatozoa, the intracellular ATP content, and the fine structure of spermatozoa were studied in relation to time post activation. Furthermore, the fertilisation... |

The impact of diets upon the fatty acid composition of haemocyte polar lipids and consequently upon immune parameters has been tested in the oyster Crassostrea gigas and the clam Ruditapes philippinarum. Oysters and clams were fed each of three cultured algae: Chaetoceros calcitrans, which is rich in 20:5(n-3) and 20:4(n-6) and poor in 22:6(n-3) fatty acids; T-Iso (Isochrysis sp.), which is rich in 22:6(n-3) and deficient in 20:5(n-3) and 20:4(n-6); and Tetraselmis suecica, which is deficient in 22:6(n-3) and contains only small amounts of 20:5(n-3) and 20:4(n-6). Fatty acid composition of haemocyte polar lipids was greatly affected by the diet. Oysters and clams fed C. calcitrans maintained a higher proportion of 20:5(n-3) and 20:4(n-6) in their haemocyte... |

In the natural environment bivalve larvae are exposed to variable conditions and can therefore face periods of food shortage. To understand the resistance of early life stages of Pacific oyster (Crassostrea gigas), larvae were experimentally starved for 4 days at different periods of their development, and the resulting variations in their lipid reserves then analysed using image analysis and biochemical techniques. Faced with a temporary lack of food, the larvae halted their growth in size and weight and started to live off their lipid reserves; this effect was highly marked when the period of food deprivation was at the beginning of larval rearing, but was less marked when food deprivation was after day 14. The larvae conserved their developmental... |

The main objective of this study was to identify algal diets that maximize the survival and growth and alter the biochemical content of Pecten maximus larvae with the aim of improving metamorphosis. We also evaluated the potential of the cryptophyceae Rhodomonas salina as a food source for these larvae. Two flagellates, Isochrysis aff. galbana (T) and Pavlova lutheri (P), and two diatoms, Chaetoceros gracilis (C) or Skeletonema costatum (S), were tested as two ternary diets, namely PTC and PTS. PTC and PTS were compared with diets that also included R. salina (R). The addition of R. salina and the replacement of C. gracilis by S. costatum in the traditional hatchery diet seem to be two interesting alternatives for increasing the productivity of larval... |

Sperm characteristics of scallops have not been well described in the scientific literature. The effects of sperm release technique (thermal shock versus serotonin injection), of sperm collection technique (testis sampling versus serotonin injection), of sperm sampling location along the genital tract, of in vitro sperm maturation, and of time post activation on scallop sperm characteristics were assessed in the present work. Whatever sperm release technique used, no significant differences were observed regarding the percentage of motile spermatozoa and the velocity of the average path (YAP). Compared to testicular sperm, a higher percentage of motile spermatozoa, YAP and intracellular adenosine triphosphate (ATP) content were observed for sperm shed... |

Standard and mutant Tisochrysis lutea strains were grown in batch culture for 10 days under nitrogen (N)-replete and N-reduced conditions to determine the effects of N supply in culture medium and growth phase on microalgal physiology and biochemistry. These two T. lutea strains were compared in terms of growth, morphology, associated free-living bacterial community, viability, intracellular lipid content (as measured by Bodipy staining), chlorophyll autofluorescence, and biochemical composition, with a focus on lipid class and fatty acid compositions. The standard strain (T) reached higher cell counts regardless of N supply in culture medium. In both T. lutea strains, microalgal final cell density was significantly lower in N-reduced medium. Here, we assess the physiological effects induced by environmental concentrations of pesticides in Pacific oyster Crassostrea gigas. Oysters were exposed for 14 d to trace levels of metconazole (0.2 and 2 mu g/L), isoproturon (0.1 and 1 mu g/L), or both in a mixture (0.2 and 0.1 mu g/L, respectively). Exposure to trace levels of pesticides had no effect on the filtration rate, growth, and energy reserves of oysters. However, oysters exposed to metconazole and isoproturon showed an overactivation of the sensing-kinase AMP-activated protein kinase alpha (AMPK alpha), a key enzyme involved in energy metabolism and more particularly glycolysis. In the meantime, these exposed oysters showed a decrease in hexokinase and pyruvate kinase activities, whereas 2-DE... |

Mass mortality outbreaks (90-100%) in both juvenile and adult blue mussels were reported in 2014 along the Pertuis Charentais area of the French Atlantic coast. A series of studies were carried out to learn more about the phenomenon through detecting pathogens, describing environmental conditions and establishing the physiological status of the animals. Bacteria identif ied as belonging to the V. splendidus species were detected in dying mussels and seem able to cause mortality in blue mussels in the laboratory. Unusual environmental conditions (large amounts of fresh water, re-suspension of sediments during a series of storms, and a high renewal rate of specific water masses in Brittany’s Pertuis area) may be partially responsible for sparking the phenomen |

Essential fatty acids (EFA) are important for bivalve larval survival and growth. The purpose of this study was to quantitatively assess for the first time through a mass-balance approach dietary EFA incorporation and synthesis within Crassostrea gigas larvae. A first experiment was carried out using two microalgae, Tisochrysis lutea (T) and Chaetoceros neogracile (Cg), as mono- and bi-specific diets. A second experiment using a similar design was performed to confirm and extend the results obtained in the first. Flow-through larval rearing was used for accurate control of food supply and measurement of ingestion. Non-methylene-interrupted fatty acids were synthetized from precursors supplied in the diet: 16:1n-7 and 18:1n-9, mediated by Delta 5... |

Sessile animals that live on the foreshore undergo tidal cycles, and have to face variations in physical and chemical parameters such as oxygen concentration. During emersion, availability of dissolved oxygen can be lowered for bivalves, which have only a small reserve of seawater inside their closed shell. Differences in oxygen concentration are thus expected to lead to modifications of the metabolism, including changes in mitochondrial activity. Previous studies investigated air exposure under extreme conditions, which do not always reflect environmental conditions these invertebrates have to cope with. In this study, oxidative capacities of gill mitochondria of the oyster Crassostrea gigas were studied during a tidal cycle period, by comparing oysters... |

Triploidy induction in shellfish aims to obtain faster growth and sterility of reared individuals. Triploid Pacific oysters are most often not totally sterile, but have greatly reduced reproductive capacities compared to those recorded in diploid individuals. The description of gamete output in triploid aquatic animals is most often confined to the assessment of fecundity in females. The present work aims at further describing gamete quality of triploid Pacific oysters and comparing it to values observed in diploid individuals. Diploid and triploid oysters (produced by mating tetraploid males × diploid females) were reared in standard conditions and then transferred to the wild to allow the offspring to attain reproductive maturity. At the end of... |

Effects of a remarkably high overall lipid Tisochrysis lutea strain (T+) upon gross biochemical composition, fatty acid (FA), sterol and lipid class composition of Crassostrea gigas larvae were evaluated and compared with a normal strain of Tisochrysis lutea (T) and the diatom Chaetoceros neogracile (Cg). In a first experiment, the influence of different single diets (T, T+ and Cg) and a bispecific diet (TCg) was studied, whereas, effects of monospecific diets (T and T+) and bispecific diets (TCg and T+Cg) were evaluated in a second experiment. The strain T+ was very rich in triglycerides (TAG: 93–95% of total neutral lipids), saturated FA (45%), monounsaturated FA (31–33%) and total fatty acids (4.0–4.7 pg cell−1).
