Abstract : This study identified and compared the frequency of homology and recombination hotspots in sexual diploid and tetraploid species, apomictic tetraploids and hybrids of Brachiaria, by immunolocalization of the radiation sensitive 51 (RAD51). Roots were fixed in 4% paraformaldehyde. We applied primary antibody (Rat anti-Arabidopsis thaliana RAD51) and detected with a secondary antibody (Goat anti-rat IgG FITC). The analysis of this protein showed diffuse signals at leptotene. In zygotene, signals of RAD51 were identified in unpaired chromosomes or partially paired and adjacent signals of RAD51 in paired chromosomes, confirming its close involvement with the search for chromosomal homology. During pachytene, RAD51 signals were located between... |

The genus Urochloa P. Beauv. [syn. Brachiaria (Trin.) Griseb.] comprises species of great economic relevance as forages. The genomic constitution for the allotetraploid species Urochloa brizantha (cv. Marandu) and Urochloa decumbens (cv. Basilisk) and the diploid Urochloa ruziziensis was previously proposed as BBB1B1, B1B1B2B2 and B2B2, respectively. Evidence indicates U. ruziziensis as the ancestral donor of genome B2 in U. decumbens allotetraploidy, but the origin of the genomes B and B1 is still unknown. There are diploid genotypes of U. brizantha and U. decumbens that ay be potential ancestors of the tetraploids. The aim of this study was to determine the genomic constitution and relationships between genotypes of U. brizantha (2x and 4x), U. decumbens... |

The common bean has great social and economic importance in Brazil and is the subject of a high number of publications, especially in the fields of genetics and breeding. Breeding programs aim to increase grain yield; however, mitosis and meiosis represent under explored research areas that have a direct impact on grain yield. Therefore, the study of cell division could be another tool available to bean geneticists and breeders. The aim of this study was to investigate irregularities occurring during the cell cycle and meiosis in common bean. The common bean cultivar used was BRSMG Talismã, which owing to its high yield and grain quality is recommended for cultivation in Brazil. We classified the interphase nuclei, estimated the mitotic and meiotic index,... |

Digitaria abyssinica (African couchgrass) has economic potential as forage. This species is taxonomically complex, with a high number of synonyms, basic chromosome number x = 9, and diploid and tetraploid cytotypes. Anatomical, histological, and cytotaxonomic studies may contribute to the taxonomic description of this species and reveal its polyploidization and hybridization events. To this end, this study aimed to characterize the karyotype and leaf epidermis histology of two D. abyssinica genotypes. Chromosome characterization was performed based on chromosome banding using chromomycin A (CMA) and 4,6-diamidino-2-phenylindole (DAPI) and identification of 35S and 5S rDNA sites by fluorescent in situ hybridization. Nuclear DNA quantification was performed... |

Abstract Cynodon is a genus of plants with forage potential that has attracted the interest of breeders. These species have high morphological variability in a large number of varieties and cytotypes, hampering identification. This study aimed to determine the karyotype asymmetry index among accessions of Cynodon to discriminate between them. Karyotype symmetry was based on three estimates, which were compared. The basic number for the genus is x = 9. The results of the chromosome count and DNA quantification, respectively, were as follows: two diploid accessions (2n = 2x = 18 and 1.08 ± 0.094 to 1.17 ± 0.036 pg DNA and ± standard deviation), one triploid accession (2n = 3x = 27 and 1.63 ± 0.017 pg DNA), four tetraploid accessions (2n = 4x = 36 and 1.88 ±... |
