15N-labed glycine synthesis Anais da ABC (AABC)
Tavares,Claudinéia R.O.; Bendassolli,José A.; Coelho,Fernando; Sant'ana Filho,Carlos R.; Prestes,Clelber V..
This work describes a method for 15N-isotope-labeled glycine synthesis, as well as details about a recovery line for nitrogen residues. To that effect, amination of alpha-haloacids was performed, using carboxylic chloroacetic acid and labeled aqueous ammonia (15NH3). Special care was taken to avoid possible 15NH3 losses, since its production cost is high. In that respect, although the purchase cost of the 13N-labeled compound (radioactive) is lower, the stable tracer produced constitutes an important tool for N cycling studies in living organisms, also minimizing labor and environmental hazards, as well as time limitation problems in field studies. The tests were carried out with three replications, and variable 15NH3(aq) volumes in the reaction were used...

2,3,8-Trisubstituted Quinolines with Antimalarial Activity Anais da ABC (AABC)
MARTINEZ,PABLO D.G.; KRAKE,SUSANN H.; POGGI,MAITIA L.; CAMPBELL,SIMON F.; WILLIS,PAUL A.; DIAS,LUIZ C..
ABSTRACT Combination therapy drugs are considered a fundamental way to control malaria as it mimimizes the risk of emergence of resistance to the individual partner drugs. Consequently, this type of therapy constitutes a driving force for the discovery of new drugs with different modes of action, since this will provide options for combining different drugs to achieve the optimum antimalarial treatment. In this context, a 2,3,8-trisubstitued quinoline compound was found in a high throughput screen (HTS) to show an excellent inhibition of P. falciparum NF54 (IC50 = 22 nM) and low cytotoxicity. We performed a detailed evaluation of the substituents to improve the metabolic stability and solubility liabilities of the original hit and identified derivatives...

Gomes,Urubatã E.; Oliveira,Diogo L. de; Berti,Luciana C.; Amaral,Olavo; Souza,Diogo O.; Wofchuk,Susana T..
Scientific activity in Brazil has experienced an accelerated growth in the past decades, with an increase in productivity that greatly surpasses the international average. This growth has occurred mostly at the expense of centers of excellence in public universities, which account for the vast majority of the country's scientific output. The aim of this study was to evaluate the production of the Department of Biochemistry of a large public university in southern Brazil (Universidade Federal do Rio Grande do Sul), as well as to identify internal and external policies that have influenced this growing production profile. We have performed a historical analysis of the scientific output of this Department of Biochemistry, which accounts for a considerable...

40Ar/39Ar ages (600-570 Ma) of the Serra do Azeite transtensional shear zone: evidence for syncontractional extension in the Cajati area, southern Ribeira belt Anais da ABC (AABC)
Machado,Rômulo; Dehler,Nolan M.; Vasconcelos,Paulo.
This paper presents 40Ar/39Ar ages of the rocks from the Serra do Azeite transtensional shear zone in the southern part of the Ribeira belt, between the States of São Paulo and Paraná, and also discusses the regional correlations and the tectonic implications for other parts of the belt. The geochronological data suggest that transtensional deformation was active between 600 and 580 Ma (hornblende and muscovite apparent ages, respectively). This time span is considerably older than previous proposals for the period of activity of these structures (520-480 Ma) in the northern segment of this belt and in the Araçuaí belt. Kinematic analysis of the dated mylonites shows extensional structures with top-down movement to ESE compatible with structures found in...

This study aimed to investigate how 6-bromoindirubin-3’-oxime (BIO) increases the osteogenic differentiation of canine bone mesenchymal stem cells (BMSCs) and the role of the Wnt/β-catenin signaling pathway in this process. We mimicked the effect of Wnt by adding BIO to the culture medium of BMSCs and examined whether canonical Wnt signaling positively affects the differentiation of these cells into osteoblasts. Canine BMSCs were cultured with 0.5 and 1.0 μM BIO under osteogenic conditions and then differentiation markers were investigated. It was found that BIO significantly increased the activity of alkaline phosphatase (ALP), the number of ALP-positive cells, the mineralization level and calcium deposits. Moreover, cells cultured with 0.5 and 1.0 μM...

A brief report on some health aspects of rats fed with crescent levels of recombinant chagasin, a potential plant defense protein Anais da ABC (AABC)
Oliveira Neto,Osmundo B.; Farias,Davi F.; Vasconcelos,Ilka M.; Paes,Norma S.; Monteiro,Ana C. S.; Silva,Maria C. M. da; Guimarães,Luciane M.; Carvalho,Ana F.U.; Grossi-de-Sá,Maria F..
Chagasin may be considered a potential plant-incorporated protectant (PIP) protein due to its deleterious effects on insect pests. However, extensive safety studies with PIP's are necessary before introducing them into the target plant. Thus, a short-term feeding trial in rats with high doses of r-chagasin was conducted to provide evidences about its safety. Three test diets containing casein + r-chagasin (0.25, 0.5 and 1% of total protein) were offered to rats (10 days). The test diets did not show adverse effects upon the development, organ weight, hematological parameters and serum protein profiles of rats, providing preliminary information on the safety of r-chagasin.

A close look at above ground biomass of a large and heterogeneous Seasonally Dry Tropical Forest - Caatinga in North East of Brazil Anais da ABC (AABC)
CASTANHO,ANDREA D.A.; COE,MICHAEL; ANDRADE,EUNICE M.; WALKER,WAYNE; BACCINI,ALESSANDRO; CAMPOS,DIEGO A.; FARINA,MARY.
Abstract This work is focused on characterizing and understanding the aboveground biomass of Caatinga in a semiarid region in northeastern Brazil. The quantification of Caatinga biomass is limited by the small number of field plots, which are inadequate for addressing the biome’s extreme heterogeneity. Satellite-derived biomass products can address spatial and temporal changes but they have not been validated for seasonally dry tropical forests. Here we combine a compilation of published field phytosociological observations with a new 30m spatial resolution satellite biomass product. Both data were significantly correlated, satellite estimates consistently captured the wide variability of the biomass across the different physiognomies (2-272 Mg/ha). Based...

A comparative expression analysis of gene transcripts in brain tissue of non-transgenic and GH-transgenic zebrafish (Danio rerio) using a DDRT-PCR approach Anais da ABC (AABC)
Alves-Costa,Fernanda A.; Figueiredo,Márcio A.; Lanes,Carlos F.C.; Almeida,Daniela V.; Marins,Luis F.; Wasko,Adriane P..
The presence of higher level of exogenous growth hormone (GH) in transgenic animals could lead to several physiological alterations. A GH transgenic zebrafish (Danio rerio) line was compared to nontransgenic (NT) samples of the species through a DDRT-PCR approach, with the goal of identifying candidate differentially expressed transcripts in brain tissues that could be involved in GH overexpression. Densitometric analyses of two selected amplification products, p300 and ADCY2, pointed to a significant lower gene expression in the transgenic zebrafish (104.02 ± 57.71; 224.10 ± 91.73) when compared to NT samples (249.75 ± 30.08; 342.95 ± 65.19).
