1a,25(OH)2D3 induces capacitative calcium entry involving a trpc3 protein in skeletal muscle and osteoblastic cells Biol. Res.
SANTILLÁN,G; KATZ,S; BUITRAGO,C; BOLAND,R.
This work describes the involvement of TRPC proteins in capacitative calcium entry (CCE) induced by 1a,25-dihydroxy-vitamin-D3 [1a,25(OH)2D3] in chick skeletal muscle and in rat osteoblast-like cells (ROS 17/2.8) and the role of the vitamin D receptor (VDR) in this non-genomic rapid response mediated by the hormone. We propose that an endogenous TRPC3 protein mediates 1a,25(OH)2D3 modulation of CCE in these cells, which seems to implicate VDR-TRPC3 association and the participation of an INAD-like scaffold protein.

50 years of amino acid hydrophobicity scales: revisiting the capacity for peptide classification Biol. Res.
Simm,Stefan; Einloft,Jens; Mirus,Oliver; Schleiff,Enrico.
BACKGROUND: Physicochemical properties are frequently analyzed to characterize protein-sequences of known and unknown function. Especially the hydrophobicity of amino acids is often used for structural prediction or for the detection of membrane associated or embedded β-sheets and α-helices. For this purpose many scales classifying amino acids according to their physicochemical properties have been defined over the past decades. In parallel, several hydrophobicity parameters have been defined for calculation of peptide properties. We analyzed the performance of separating sequence pools using 98 hydrophobicity scales and five different hydrophobicity parameters, namely the overall hydrophobicity, the hydrophobic moment for detection of...

Most cells of the developing mammalian brain derive from the ventricular (VZ) and the subventricular (SVZ) zones. The VZ is formed by the multipotent radial glia/neural stem cells (NSCs) while the SVZ harbors the rapidly proliferative neural precursor cells (NPCs). Evidence from human and animal models indicates that the common history of hydrocephalus and brain maldevelopment starts early in embryonic life with disruption of the VZ and SVZ. We propose that a "cell junction pathology" involving adherent and gap junctions is a final common outcome of a wide range of gene mutations resulting in proteins abnormally expressed by the VZ cells undergoing disruption. Disruption of the VZ during fetal development implies the loss of NSCs whereas VZ disruption...

A comparison of methods employed to evaluate antioxidant capabilities Biol. Res.
PEREZ,DRUSO D; LEIGHTON,FEDERICO; ASPEE,ALEXIS; ALIAGA,CAROLINA; LISSI,EDUARDO.
Thrce ditferent methodologies frequently employed to evaluate the indexes that report the antioxidant capabilities of pure compounds and/or complex mixtures of antioxidants are applied to a series of mono- and polyphenols, as well as to two wine (red and white) samples. These methodologies are based on the bleaching of a stable radical, the effect of the additive upon luminol chemiluminescence induced by peroxyl radicals, and the effect of the additive upon the bleaching of the fluorescence from a dye molecule. Widely ditferent responses are obtained from the different methodologies. These differences are interpreted in terms of the different factors (stoichiametric factors and/or reactivities) that determines the indexes evaluated by these different...

A high-fat diet decreases GABA concentration in the frontal cortex and hippocampus of rats Biol. Res.
Sandoval-Salazar,Cuauhtemoc; Ramírez-Emiliano,Joel; Trejo-Bahena,Aurora; Oviedo-Solís,Cecilia I.; Solís-Ortiz,Martha Silvia.
BACKGROUND: It has been proposed that the γ-aminobutyric acid (GABA) plays a key role in the regulation of food intake and body weight by controlling the excitability, plasticity and the synchronization of neuronal activity in the frontal cortex (FC). It has been also proposed that the high-fat diet (HFD) could disturb the metabolism of glutamate and consequently the GABA levels, but the mechanism is not yet clearly understood. Therefore, the aim of this study was to investigate the effect of a HFD on the GABA levels in the FC and hippocampus of rats RESULTS: The HFD significantly increased weight gain and blood glucose levels, whereas decreased the GABA levels in the FC and hippocampus compared with standard diet-fed rats CONCLUSIONS: HFD...

A light- and electron microscopic study of primordial germ cells in the zebra fish (Danio rerio) Biol. Res.
Deniz Koç,Nazan; Yüce,Rikap.
In sexually reproducing organisms, primordial germ cells (PGCs) give rise to the cells of the germ line, the gametes. In many animals, PGCs are set apart from somatic cells early during embryogenesis. This study explores the origin of primordial germ cells (PGCs) of the zebra fish and examines their morphology during early development (1st day-15th day). PGCs were selectively stained by the alkaline phosphatase histochemical reaction and viewed by light and electron microscopy from the time they are first detectable in the yolk sac endoderm. PGCs occurred in the subendodermal space on the syncytial periblast; differing from the surrounding endodermal cells. Later the PGCs moved to between the blastoderm and yolk sac and transferred to the dorsal mesentery...

A Lux-like Quorum Sensing System in the Extreme Acidophile Acidithiobacillus ferrooxidans Biol. Res.
RIVAS,MARIELLA; SEEGER,MICHAEL; HOLMES,DAVID S; JEDLICKI,EUGENIA.
The genome of the acidophilic, proteobacterium Acidithiobacillus ferrooxidans, contains linked but divergently oriented genes, termed afeI and afeR, whose predicted protein products are significantly similar to the LuxI and LuxR families of proteins. A possible promoter and Lux box are predicted upstream of afeI. A cloned copy of afeI, expressed in E. coli, encodes an enzyme that catalyzes the production of a diffusible compound identified by gas chromatography and mass spectrometry as an unsubstituted N-acyl homoserine lactone (AHL) of chain length C14. This AHL can be detected by a reporter strain of Sinorhizobium meliloti Rm41 suggesting that it is biologically active. The reporter strain also responds to extracts of the supernatant of A. ferrooxidans...

A micromethod for high throughput RNA extraction in forest trees Biol. Res.
LE PROVOST,GREGOIRE; HERRERA,RAÚL; PAIVA,JORGE AP; CHAUMEIL,PHILIPPE; SALIN,FRANCK; PLOMION,CHRISTOPHE.
A large quantity of high quality RNA is often required in the analysis of gene expression. However, RNA extraction from samples taken from woody plants is generally complex, and represents the main limitation to study gene expression, particularly in refractory species like conifers. Standard RNA extraction protocols are available but they are highly time consuming, and not adapted to large scale extraction. Here we present a high-throughput RNA extraction protocol. This protocol was adapted to a micro-scale by modifying the classical cetyltrimethylammonium (CTAB) protocol developed for pine: (i) quantity of material used (100-200 mg of sample), (ii) disruption of samples in microtube using a mechanical tissue disrupter, and (iii) the use of SSTE buffer....

A neutral cue facilitates detection of a visual target by modulating attention Biol. Res.
HAMAME,CARLOS M; DELANO,PAÚL H; ROBLES,LUIS.
Twelve rats were trained to perform a two-choice visual detection task in which a right or left light was presented and the animáis were required to press the lever located under the illuminated light for a food reward. In seventy percent of the triáis the target light was preceded by presentation of a neutral cue (a central light). Relevance of the neutral cue for detection of the target was analyzed by comparing behavioral Índices of attention in its presence and absence. Accuracy was significantly higher in presence than in absence of the neutral cue, while mean response latencies were lower in presence than in absence of the neutral cue. These results indicate that the animáis allocated attentional resources on the target detection during a high...

A pilot study on genetic variation in purine-rich elements in the nephrin gene promoter in type 2 diabetic patients Biol. Res.
GONZÁLEZ,RODRIGO; TIRADO,ARES; BALANDA,MONSERRAT; ALVO,MIRIAM; BARQUÍN,INÉS; DURRUTY,PILAR; LOBOS,SERGIO; SEELENFREUND,DANIELA.
Diabetic nephropathy (DN) is one of the major complications of type 2 diabetes and is associated with coronary disease. Nephrin, a protein mainly expressed in glomeruli, is decreased in DN and other kidney diseases. Since insulin levels are misregulated in type 2 diabetes, a possible connection between DN and its decreased nephrin expression could be the presence of regulatory elements responsive to insulin in the nephrin gene (NPHS1) promoter region. In this work, using bioinformatic tools, we identified a purine-rich GAGA element in the nephrin gene promoter and conducted a genomic study in search of the presence of polymorphisms in this element and its possible association with DN in type 2 diabetic patients. We amplified and sequenced a 514 bp promoter...

A possible role of Drosophila CTCF in mitotic bookmarking and maintaining chromatin domains during the cell cycle Biol. Res.
Shen,Wenlong; Wang,Dong; Ye,Bingyu; Shi,Minglei; Zhang,Yan; Zhao,Zhihu.
BACKGROUND: The CCCTC-binding factor (CTCF) is a highly conserved insulator protein that plays various roles in many cellular processes. CTCF is one of the main architecture proteins in higher eukaryotes, and in combination with other architecture proteins and regulators, also shapes the three-dimensional organization of a genome. Experiments show CTCF partially remains associated with chromatin during mitosis. However, the role of CTCF in the maintenance and propagation of genome architectures throughout the cell cycle remains elusive. RESULTS: We performed a comprehensive bioinformatics analysis on public datasets of Drosophila CTCF (dCTCF). We characterized dCTCF-binding sites according to their occupancy status during the cell cycle, and identified...

A Pyranine based Procedure for Evaluation of the Total Antioxidant Potential (TRAP) of Polyphenols: A Comparison with closely related Methodologies Biol. Res.
CAMPOS,ANA MARÍA; SOTOMAYOR,CARLOS PATRICIO; PINO,EDUARDO; LISSI,EDUARDO.
A novel procedure for the evaluation of total reactive antioxidant potentials (TRAP) is described. The method is based on the measurement of the bleaching of pyranine by peroxyl radicals. The addition of the antioxidants produces a clear induction time whose magnitude is directly related to the antioxidant concentration. A comparison of the values obtained with those reported employing closely related methodologies shows that the results are significantly affected by the substrate employed to monitor the steady state free radical concentration. Possible sources of this dependence are discussed

A radiographic method to estimate lung volume and its use in small mammals Biol. Res.
CANALS,MAURICIO; OLIVARES,RICARDO; ROSENMANN,Mario.
In this paper we develop a method to estimate lung volume using chest x-rays of small mammals. We applied this method to assess the lung volume of several rodents. We showed that a good estimator of the lung volume is: V*L = 0.496 · V RX ≈ 1/2·V RX , where V RX is a measurement obtained from the x-ray that represents the volume of a rectangular box containing the lungs and mediastinum organs. The proposed formula may be interpreted as the volume of an ellipsoid formed by both lungs joined at their bases. When that relationship was used to estimate lung volume, values similar to those expected from allometric relationship were found in four rodents. In two others, M. musculus and R. norvegicus, lung volume was similar to reported data, although...

Prunus persica has been proposed as a genomic model for deciduous trees and the Rosaceae family. Optimized protocols for RNA isolation are necessary to further advance studies in this model species such that functional genomics analyses may be performed. Here we present an optimized protocol to rapidly and efficiently purify high quality total RNA from peach fruits (Prunus persica). Isolating high-quality RNA from fruit tissue is often difficult due to large quantities of polysaccharides and polyphenolic compounds that accumulate in this tissue and co-purify with the RNA. Here we demonstrate that a modified version of the method used to isolate RNA from pine trees and the woody plant Cinnamomun tenuipilum is ideal for isolating high quality RNA from the...

A review on plant importance, biotechnological aspects, and cultivation challenges of jojoba plant Biol. Res.
Al-Obaidi,Jameel R.; Halabi,Mohammed Farouq; AlKhalifah,Nasser S.; Asanar,Shanavaskhan; Al-Soqeer,Abdulrahman A.; Attia,M. F..
Abstract Jojoba is considered a promising oil crop and is cultivated for diverse purposes in many countries. The jojoba seed produces unique high-quality oil with a wide range of applications such as medical and industrial-related products. The plant also has potential value in combatting desertification and land degradation in dry and semi-dry areas. Although the plant is known for its high-temperature and high-salinity tolerance growth ability, issues such as its male-biased ratio, relatively late flowering and seed production time hamper the cultivation of this plant. The development of efficient biotechnological platforms for better cultivation and an improved production cycle is a necessity for farmers cultivating the plant. In the last 20 years, many...

A roadmap to directed enzyme evolution and screening systems for biotechnological applications Biol. Res.
Martínez,Ronny; Schwaneberg,Ulrich.
Enzymes have been long used in man-made biochemical processes, from brewing and fermentation to current industrial production of fine chemicals. The ever-growing demand for enzymes in increasingly specific applications requires tailoring naturally occurring enzymes to the non-natural conditions found in industrial processes. Relationships between enzyme sequence, structure and activity are far from understood, thus hindering the capacity to design tailored biocatalysts. In the field of protein engineering, directed enzyme evolution is a powerful algorithm to generate and identify novel and improved enzymes through iterative rounds of mutagenesis and screening applying a specific evolutive pressure. In practice, critical checkpoints in directed evolution...
