Exposure to aflatoxins (AFs) in the diet may favour the development of hepatocellular carcinoma (HCC) and the acute exacerbation of hepatitis in chronic hepatitis B virus (HBV) carriers. Measurement of biomarkers such as aflatoxin M1 (AFM1), a metabolite of aflatoxin B1 (AFB1), in urine allows for the assessment of populations exposed to aflatoxins. The aim of this study was to investigate the occurrence of aflatoxin M1 in the urine of HBV carrier and non-carrier patients. ABSTRACT: The effects of mycotoxins on the productive performance of growing pigs and broilers were evaluated using meta-analytical approach. Two databases were constructed: (1) Broilers, with information collected from 51,497 birds and published in 158 scientific papers from 1980 to 2016; and (2) Pigs, with information collected from 7,743 animals and published in 72 scientific papers from 1980 to 2015. The meta-analyses were performed independently for each specie, following three sequential analyses: graphical, correlation, and variance-covariance. The biflavonoids 6,6"-bigenkwanin, amenthoflavone, 7,7"-dimethoxyagastisflavone and tetradimethoxybigenkwanin isolated from Ouratea species were tested for inhibitory activity on Aspergillus flavus cultures. Suspensions of Aspergillus flavus spores were inoculated into 50 ml of YES medium at different biflavonoid concentrations: 5 and 10 µg/ml for 6,6"-bigenkwanin, amenthoflavone and 7,7"-dimethoxyagastisflavone, and 5, 10, 15 and 20 µg/ml for tetradimethoxybigenkwanin. The four biflavonoids showed inhibitory activity on aflatoxin B1 and B2 production (P<0.001), but did not inhibit fungal growth at the concentration tested (P>0.05). These results show that biflavonoids can be used for the development of agents to control aflatoxin production. |

O amendoim é uma cultura oleaginosa que constitui importante fonte de óleos e proteínas para populações do Mali e do Brasil. Em ambos os países, é produzido, principalmente, por pequenos produtores, em áreas que variam de 0,3 ha a 5 ha, com espaçamentos entrelinhas de, aproximadamente, 0,50 m, utilizando-se de 10 a 20 sementes por metro e gastando-se em torno de 90 kg de sementes/ha. A contaminação por aflatoxinas é o principal problema associado à cultura e está intimamente ligada ao crescimento de fungos (bolores) que se desenvolvem, favorecidos pelas condições climáticas nos dois países. |

In this study the bright greenish-yellow fluorescence test, widely used by the corn milling industry, was compared to the thin-layer chromatography (TLC) and spectrofluorimetry methods for aflatoxin detection in 40 corn samples naturally contaminated by the Aspergillus section Flavi. According to the corn processing industry criteria, all the samples were adequate for human and animal consumption by the bright greenish-yellow fluorescence test, but TLC and spectrofluorimetry analysis detected aflatoxins above the maximum tolerated limit (20 µg/kg) in 7 and 8 samples, respectively. A method for determining aflatoxins B1 (AFB1), B2 (AFB2),G1 (AFG1) andG2 (AFG2) in maize with florisil clean up was optimised aiming at one-dimensional thin layer chromatography (TLC) analysis with visual and densitometric quantification. Aflatoxins were extracted with chloroform: water (30:1, v/v), purified through florisil cartridges, separated on TLC plate, detected and quantified by visual and densitometric analysis. The in-house method performance characteristics were determined by using spiked, naturally contaminated maize samples, and certified reference material. The aflatoxin distribution in corn fractions obtained after visual segregation for defects in 30 samples, known to be contaminated, was studied. Each sample was passed through a 5.0 mm round holes sieve, graded for defects and then segregated in sound kernels (regular kernels) and non-sound kernels (injured, germinated, fermented, moldy, heated, insect damaged, immature, broken, hollow, fermented up to ¼, discolored, extraneous materials, and injured by other causes), as defined by the Brazilian Official Grading rules for corn. Macrophages exposed to 10 mug/mL citrinin (CTR) or 0.01 mug CTR mixed with 0.04 mug aflatoxin B1 (AFB1) for a period of 2 h at 37ºC, were infected with 10(6) Toxoplasma gondii tachyzoites/muL. The parasites were treated with mycotoxins (2 h at 37ºC) before being added to the macrophage culture. The number of tachyzoites was quantified 2, 24, 48, 72 and 96 h after infection. During the first 2 hours, 59% infectivity was observed in the control. After exposure to CTR or the mixture of toxins (CTR-AFB1), macrophages were infected with 77.5% and 75% of the inoculated tachyzoites, respectively. The effect of ten powdered spice plants was evaluated at the concentration of 1, 2, 3 and 4% to observe the mycelial growth and sporulation of Aspergillus niger and Eurotium repens. The spices were added to the culture media PDA and CYA20S. Clove completely inhibited the mycelial growth of the tested fungi. The other spices: cinnamon, garlic, thyme, mint, anis, oregano and onion were, in a decreasing order, promising antifungals. Bay leaf and basil did not show a pronounced fungistatic effect.
