A polyphasic approach to the identification of aflatoxigenic and non-aflatoxigenic strains of Aspergillus section flavi isolated from portuguese almonds IPB - Escola Superior Agrária
Rodrigues, Paula; Venâncio, Armando; Kozakiewicz, Zofia; Lima, Nelson.
A polyphasic approach consisting of morphological, chemical and molecular characterization was applied to 31 isolates of Aspergillus Section Flavi originating from Portuguese almonds, with the aim of characterizing and identifying aflatoxigenic and non-aflatoxigenic strains. On the basis of morphological characters (mainly colony color on Czapek-Dox agar and conidia morphology), we found two distinct groups among the population under study: 18 isolates (58%) had dark-green colonies and rough conidia, and were classified as A. parasiticus; the remaining 13 isolates (42%) had yellow-green colonies and smooth to finely rough globose conidia, and were classified as A. flavus. Chemical characterization involved the screening of the isolates for aflatoxins B...

Aflatoxin M1 in the urine of non-carriers and chronic carriers of hepatitis B virus in Maringa, Brazil BJPS
Giolo,Marcel Padovani; Oliveira,Christiane Minervino de; Bertolini,Dennis Armando; Lonardoni,Maria Valdrinez Campana; Gouveia,Matheus Sampaio; Netto,Daisy Pontes; Nixdorf,Suzana Lucy; Machinski Junior,Miguel.
Exposure to aflatoxins (AFs) in the diet may favour the development of hepatocellular carcinoma (HCC) and the acute exacerbation of hepatitis in chronic hepatitis B virus (HBV) carriers. Measurement of biomarkers such as aflatoxin M1 (AFM1), a metabolite of aflatoxin B1 (AFB1), in urine allows for the assessment of populations exposed to aflatoxins. The aim of this study was to investigate the occurrence of aflatoxin M1 in the urine of HBV carrier and non-carrier patients. One group included 43 randomly selected HBV carriers treated at two hospitals in the city of Maringa, Brazil, from March to June 2008. Control group consisted of 29 healthy adult volunteers with anti-HBs positive and HBsAg negative test results. Detection of AFM1 was performed by...

Assessing the implications of mycotoxins on productive efficiency of broilers and growing pigs Scientia Agricola
Kipper,Marcos; Andretta,Ines; Ribeiro,Andréa Machado Leal; Pires,Paula Gabriela da Silva; Franceschina,Carolina Schell; Cardinal,Kátia Maria; Moraes,Priscila de Oliveira; Schroeder,Bruna.
ABSTRACT: The effects of mycotoxins on the productive performance of growing pigs and broilers were evaluated using meta-analytical approach. Two databases were constructed: (1) Broilers, with information collected from 51,497 birds and published in 158 scientific papers from 1980 to 2016; and (2) Pigs, with information collected from 7,743 animals and published in 72 scientific papers from 1980 to 2015. The meta-analyses were performed independently for each specie, following three sequential analyses: graphical, correlation, and variance-covariance. Broilers and pigs challenged by mycotoxins reduced (p < 0.05) feed intake by 9 and 6 %, weight gain by 15 and 11 %, and feed efficiency by 6 and 4 % compared with non-challenged animals, respectively....

Biflavonoids inhibit the production of aflatoxin by Aspergillus flavus BJMBR
Gonçalez,E.; Felicio,J.D.; Pinto,M.M..
The biflavonoids 6,6"-bigenkwanin, amenthoflavone, 7,7"-dimethoxyagastisflavone and tetradimethoxybigenkwanin isolated from Ouratea species were tested for inhibitory activity on Aspergillus flavus cultures. Suspensions of Aspergillus flavus spores were inoculated into 50 ml of YES medium at different biflavonoid concentrations: 5 and 10 µg/ml for 6,6"-bigenkwanin, amenthoflavone and 7,7"-dimethoxyagastisflavone, and 5, 10, 15 and 20 µg/ml for tetradimethoxybigenkwanin. The four biflavonoids showed inhibitory activity on aflatoxin B1 and B2 production (P<0.001), but did not inhibit fungal growth at the concentration tested (P>0.05). These results show that biflavonoids can be used for the development of agents to control aflatoxin production.

Characterisation of mycoflora and aflatoxigenic fungi from portuguese almonds: from production to commercialization. IPB - Escola Superior Agrária
Rodrigues, Paula; Venâncio, Armando; Lima, Nelson.
Aflatoxins are among the most carcinogenic natural compounds known, and have been frequently detected in numerous agricultural commodities, nut fruits being among the most contaminated ones. These toxins are a result of the secondary metabolism of fungi belonging to the genus Aspergillus Section Flavi, namelly Aspergillus flavus, A. parasiticus and A. nomius. The early detection of aflatoxigenic fungal contamination and the preventive control of aflatoxin production seems to be the only viable way to respond to the rigorous limits imposed by EU legislation. In Portugal, knowledge on mycoflora and mycotoxigenic profiles of almonds is scarce. Our objective was to characterise portuguese almonds in respect to mycoflora, with strong emphasis on aflatoxigenic...

Comparison of thin-layer chromatography, spectrofluorimetry and bright greenish-yellow fluorescence test for aflatoxin detection in corn BABT
Ono,Elisabete Yurie Sataque; Silva,Marcelo da; Ribeiro,Ricardo Marcelo Reche; Ono,Mario Augusto; Hayashi,Luciana; Garcia,Glauco Tironi; Hirooka,Elisa Yoko.
In this study the bright greenish-yellow fluorescence test, widely used by the corn milling industry, was compared to the thin-layer chromatography (TLC) and spectrofluorimetry methods for aflatoxin detection in 40 corn samples naturally contaminated by the Aspergillus section Flavi. According to the corn processing industry criteria, all the samples were adequate for human and animal consumption by the bright greenish-yellow fluorescence test, but TLC and spectrofluorimetry analysis detected aflatoxins above the maximum tolerated limit (20 µg/kg) in 7 and 8 samples, respectively. Aflatoxins were detected in 16 (40%) corn samples by TLC, with levels ranging from 4.0 to 54.0 µg/kg (mean 19.97 ± 15.97 µg/kg), and in 25 (62.5%) samples by spectrofluorimetry,...

Abstract Aspergillus and Fusarium are the fungi genera most frequently isolated from cereal grains and other commodities. They are capable of producing mycotoxins, which can affect the human and animal health. Synthetic fungicides have been used to control these fungi, nevertheless, they have acquired resistance and other alternatives are necessary since they now need higher amounts. Therefore, the aim of this study was to evaluate the efficacy of cinnamon, clove and thyme essential oils (EOs) alone and encapsulated in chitosan on the radial growth, spore germination and mycotoxin production by Fusarium verticillioides and Aspergillus parasiticus. The composition of the EOs was determined by gas chromatography mass spectrometry (GC-MS). EOs inhibited...

Determining aflatoxins B1, B2, G1 and G2 in maize using florisil clean up with thin layer chromatography and visual and densitometric quantification Ciênc. Tecnol. Aliment.
CASTRO,Luciana de; VARGAS,Eugenia Azevedo.
A method for determining aflatoxins B1 (AFB1), B2 (AFB2),G1 (AFG1) andG2 (AFG2) in maize with florisil clean up was optimised aiming at one-dimensional thin layer chromatography (TLC) analysis with visual and densitometric quantification. Aflatoxins were extracted with chloroform: water (30:1, v/v), purified through florisil cartridges, separated on TLC plate, detected and quantified by visual and densitometric analysis. The in-house method performance characteristics were determined by using spiked, naturally contaminated maize samples, and certified reference material. The mean recoveries for aflatoxins were 94.2, 81.9, 93.5 and 97.3% in the range of 1.0 to 242 µg/kg for AFB1, 0.3 to 85mg/kg for AFB2, 0.6 to 148mg/kg for AFG1 and 0.6 to 140mg/kg for...

Distribution of aflatoxins in corn fractions visually segregated for defects BJM
Piedade,Fabiana Segatti; Fonseca,Homero; Gloria,Eduardo Micotti da; Calori-Domingues,Maria Antonia; Piedade,Sônia Maria Stefano; Barbin,Décio.
The aflatoxin distribution in corn fractions obtained after visual segregation for defects in 30 samples, known to be contaminated, was studied. Each sample was passed through a 5.0 mm round holes sieve, graded for defects and then segregated in sound kernels (regular kernels) and non-sound kernels (injured, germinated, fermented, moldy, heated, insect damaged, immature, broken, hollow, fermented up to ¼, discolored, extraneous materials, and injured by other causes), as defined by the Brazilian Official Grading rules for corn. The non-sound kernels showed the highest contamination levels in all samples. The contamination levels of non-sound kernels (20% of total weight) ranged from 23 to 1,365 µg/kg of aflatoxins (B1, B2, G1 and G2) and were higher than...

Effect of citrinin and in association with aflatoxin B1 on the infectivity and proliferation of Toxoplasma gondii in vitro BJID
Herzog-Soares,Joanna D'Arc A; Freire,Ronald Bastos.
Macrophages exposed to 10 mug/mL citrinin (CTR) or 0.01 mug CTR mixed with 0.04 mug aflatoxin B1 (AFB1) for a period of 2 h at 37ºC, were infected with 10(6) Toxoplasma gondii tachyzoites/muL. The parasites were treated with mycotoxins (2 h at 37ºC) before being added to the macrophage culture. The number of tachyzoites was quantified 2, 24, 48, 72 and 96 h after infection. During the first 2 hours, 59% infectivity was observed in the control. After exposure to CTR or the mixture of toxins (CTR-AFB1), macrophages were infected with 77.5% and 75% of the inoculated tachyzoites, respectively. Similarly, 72.3% of the cells were infected when cultured together with previously treated parasites. The treatment with CTR-AFB1 gave rise to 2.9 times more tachyzoites...

Effect of powdered spice treatments on mycelial growth, sporulation and production of aflatoxins by toxigenic fungi Ciência e Agrotecnologia
Chalfoun,Sára Maria; Pereira,Marcelo Cláudio; Resende,Mario Lúcio V.; Angélico,Caroline Lima; Silva,Rozane Aparecida da.
The effect of ten powdered spice plants was evaluated at the concentration of 1, 2, 3 and 4% to observe the mycelial growth and sporulation of Aspergillus niger and Eurotium repens. The spices were added to the culture media PDA and CYA20S. Clove completely inhibited the mycelial growth of the tested fungi. The other spices: cinnamon, garlic, thyme, mint, anis, oregano and onion were, in a decreasing order, promising antifungals. Bay leaf and basil did not show a pronounced fungistatic effect. The antitoxigenic potential of the spices was tested against one aflatoxin-producing strain of AspergiIIus flavus. The spices were tested at the same concentrations previously mentioned and were added to the culture medium YES, appropriate for the production of those...

Identification and characterization of Aspergillus flavus and aflatoxins IPB - Escola Superior Agrária
Rodrigues, Paula; Soares, Célia; Kozakiewicz, Zofia; Paterson, Russell; Lima, Nelson; Venâncio, Armando.
Aspergillus flavus is the main producer of the well known carcinogenic aflatoxins. The presence of this fungus and aflatoxins is of huge concern in terms of food safety. The identification of A. flavus is not straightforward due to similarities with closely related species (e.g. A. parasiticus and A. nomius). Also, from the biochemical point of view the closely-related species are able to produce different mycotoxins. In order to clarify the differentiation between species the identification schemes is revisited. Selective media, data from mycotoxins production and molecular biology tools are discussed in order to clarify the concept of A. flavus species.
