Large yellow croaker is an important marine fish species for aquaculture in China. Larval rearing of this species is still based on live prey feeding. The aim of this study was to describe the onset and development of the main digestive enzymes during larval development of this species, in order to obtain essential data for the formulation of a compound diet adapted to larvae. Enzymatic assays were conducted from day I post hatching to day 40 in larvae fed the following live prey sequence: rotifers, Artemia nauplii, copepods, and then frozen copepods. Assays were done on whole larvae until day 19, then on dissected larvae and on purified brush border membranes of intestine. Pancreatic enzymes, amylase, and trypsin, showed high activities from early stages,... |

The aim of this study was to compare the effect of fluoride (F) on alkaline phosphatase activity in the liver and plasma of the rats. Four groups of male Wistar rats (n=6), which received drinking water containing 5, 15 or 50 ppm F or deionized water (control) throughout the experiment were included in the study. The animals were euthanized and had their tissues and blood plasma collected for the analysis of fluoride and alkaline phosphatase. There was an increase in F concentration in most tissues in the animals treated with higher F concentrations, except for the heart. The alkaline phosphatase assay showed an increase in the activity in the liver and blood plasma of the animals treated with fluoride concentrations of 15 and 50 ppm (p<0.05). This... |

The effects of sublethal concentrations of cadmium (0.64 µg/L), iron (0.043 mg/L) and zinc (0.31 mg/L) and a mixture of these metals on succinate dehydrogenase (SD) and alkaline phosphatase (AP) activity and on structural changes in the mitochondria of epithelium cells of the digestive tract were examined in the oligochaete Limnodrillus hoffmeisteri after 96 h of exposure in artificial sediments. SD activity was significantly inhibited, particularly in treatments with Cd alone (92.57%), while AP increased its activity with Cd alone (73.23%). However, when this metal was mixed with Fe and Zn, the inhibition of SD activity was lower (67.82%) than with Cd alone, showing an antagonistic effect and AP increased its activity (73.26%). Mitochondria were... |

The effect of breeder age on long bone development was studied in chicken embryos from 12 days of incubation until hatching. Fertile eggs were incubated and randomly assigned in a 2 x 6 factorial arrangement (two breeder ages - 38 and 60 weeks and six incubation days - 12, 14, 16, 18, 20, and 21). Enzymatic activity of acid and alkaline phosphatases in tibial epiphyses and weights as well as length and width in tibias and femurs of the embryos were determined. Tartrate-resistant acid and alkaline phosphatases activity in epiphyses was not affected by breeder age. Absolute weight and width of femur and tibia were larger in 60-week-old embryos compared to 38-week-old. Enzymatic activity and morphometric measurements increased with incubation day,... |

This work reports the first isolation and characterization of an alkaline phosphatase (AP) from a hyperthermophilic archaeon. An AP gene from Pyrococcus abyssi, a euryarchaeon isolated from a deep-sea hydrothermal vent, was cloned and the enzyme expressed in Escherichia coli. Analysis of the sequence showed conservation of the active site and structural elements of the E. coli AP. The recombinant AP was purified and characterized. Monomeric and homodimeric active forms were detected, with a monomer molecular mass of 54 kDa. Apparent optimum pH and temperature were estimated at 11.0 and 70 degreesC, respectively. Thus far, P. abyssi AP has been demonstrated to be the most thermostable AP, with half-lives at 100 and 105 degreesC of 18 and 5 h, respectively.... |

Alkaline phosphatase (ALP) is important in calcification and its expression seems to be associated with the inflammatory process. We investigated the in vitro acute effects of compounds used for the prevention or treatment of cardiovascular diseases on total ALP activity from male Wistar rat heart homogenate. ALP activity was determined by quantifying, at 410 nm, the p-nitrophenol released from p-nitrophenylphosphate (substrate in Tris buffer, pH 10.4). Using specific inhibitors of ALP activity and the reverse transcription-polymerase chain reaction, we showed that the rat heart had high ALP activity (31.73 ± 3.43 nmol p-nitrophenol·mg protein-1·min-1): mainly tissue-nonspecific ALP but also tissue-specific intestinal ALP type II. Both ALP isoenzymes... |

Given the potential significance of enzyme activities as a link between internal metabolic pathways and environmental nutrients, we investigated the relationships of nitrate reductase (NR) and alkaline phosphatase (AP) with primary production and inorganic nutrients in South Shetland Islands, Antarctica. Enzymatic activities of the phytoplankton (0.7-210 µm), primary productivity, autotrophic biomass and inorganic nutrients were studied in the upper 100 m depth at nine stations during a cruise in the northwestern area of South Shetland Islands (Antarctica), during late austral spring (December 2000). NR activities fluctuated between 0 and 42.8 nmol L-1 h-1 (mean = 10.08 nmol L-1 h-1, SD = 10.42 nmol L-1 h-1), AP activities between 0.81 and 5.67 nmol L-1... |

The role of nickel in soils of tropical areas under the application of sewage sludge is still not very well known. This study was carried out under greenhouse conditions in Jaboticabal, São Paulo State, Brazil, with the objective of evaluating the impact of the application of sewage sludge previously contaminated with rates of nickel (329, 502, 746 and 1119 mg kg-1, dry basis) on a soil cropped with sorghum, in relation to soil enzyme activity and soil microbial biomass. Soil samples were collected at the beginning and the end of the experiment. The experimental design was completely randomized, with five treatments (control and four rates of Ni in the sewage sludge) and four replications. C and N of the soil microbial biomass and enzyme activities (acid... |

ABSTRACT The current study was conducted to elucidate the clinical effect of ochratoxin A in Gallus gallusdomesticus and to ameliorate its toxic effects by the development and characterization of highly porous carbon-based adsorbent derived from coconut shell. A series of experiments were performed on one day-old chicks (Group A to F). Clinical signs of the positive control (group B fed on 400 ppb ochratoxin A contaminated feed) included depression, diarrhea, increased water intake, low body weight, high degree of genotoxicity, swollen and hemorrhagic kidneys and liver. Serum alanine amino transferase (ALT), alkaline phosphatase (ALP), creatinine and urea concentration significantly increased while albumin, globulin and total proteins were found... |

The sustainable composition of diets of high nutritional value is of the utmost importance for intensive aquaculture. Digestion and absorption of nutrients depends on the activity of the digestive enzymes, in particular those located in the brush border membrane of enterocytes, which are responsible for the final stages of breaking down and absorption of nutrients. In the present study, the substitution of fish meal by lupin or rapeseed meal in the diet was evaluated on gilthead sea bream (Sparus aurata) and goldfish (Carassius auratus). The objectives were to compare the activities of intestinal brush border enzymes in both species fed the control and experimental diets. When gilthead sea bream were fed the vegetable diets, significantly lower activities... |

ABSTRACT To describe potential intraspecific variation in phosphorus incorporation in two strains of Phaeodactylum tricornutum (Bohlin), Ub3 and Ub7, alkaline phosphatase (AP) activity was evaluated via enzyme-labeled fluorescence assay. Analysis using the probe ELF-97(r) provides individual evaluation, and therefore can determine the nutritional status of inorganic phosphorus in phytoplanktonic cells. Bioassays compared the control treatment to both phosphate-enriched and phosphate-depleted treatments by varying only the phosphate concentration in the media. The P. tricornutum strains exhibited differences in their development when incubated in the phosphate-enriched media. The development of the Ub7 strain differed by exhibiting "luxury uptake" and... |
