Resumo: O trigo (Triticum aestivum L) constitui uma das principais culturas de importância agrícola para o Brasil. Contudo, a ocorrência de doenças como a brusone, tem limitado a produtividade dessa cultura nas regiões produtoras. Desse modo, o objetivo desse trabalho foi avaliar o potencial de uma linhagem Streptomyces sp. Antifungal activity of bacterial strains from the rhizosphere of Stachytarpheta crassifolia. Repositório Alice

This study evaluated the antifungal action of biomolecules produced from the secondary metabolism of bacterial strains found in the rhizosphere of semi arid plants against human pathogenic Candida albicans. Crude extracts were obtained using ethyl acetate as an organic solvent and the bioactivity was assessed with a bioautography technique. The results showed that bacterial strains, Alcaligenes faecalis MRbS12 and Bacillus cereus MRbS26, had compounds with antifungal bioactivity. The largest inhibition zones for both compounds were located on spots with Rf values below 0.500, indicating that the molecules possibly had polar characteristics. These results suggested that microorganisms found in the environment could have bioprospecting potential.

Abstract: Aims: Epiphytic bacteria, isolated from Deschampsia antarctica, were screened for their potential to inhibit the plant pathogen Botrytis cinerea, the causal agent of gray mold disease of strawberry pseudofruits. This phytopathogenic fungus is more active and the disease is more serious in temperate climate where the temperatures are lower.

Abstract The current increment of invasive fungal infections and the availability of new broad-spectrum antifungal agents has increased the use of these agents by non-expert practitioners, without an impact on mortality. To improve efficacy while minimizing prescription errors and to reduce the high monetary cost to the health systems, the principles of pharmacokinetics (PK) and pharmacodynamics (PD) are necessary. Antifungal suscepitibility profile of candida spp. oral isolates obtained from denture wearers BJM

Denture stomatitis is an inflammatory condition that occurs in denture wearers and is frequently associated with Candida yeasts. Antifungal susceptibility profiles have been extensively evaluated for candidiasis patients or immunosupressed individuals, but not for healthy Candida carriers. In the present study, fluconazole, itraconazole, voriconazole, terbinafine and 5-flucytosin were tested against 109 oral Candida spp. isolates. All antifungal agents were effective against the samples tested except for terbinafine. This work might provide epidemiological information about Candida spp. drug susceptibility in oral healthy individuals.

Antifungal susceptibilities of Cryptococcus species complex isolates from AIDS and non-AIDS patients in Southeast China BJID

Cryptococcus spp. are common causes of mycoses in immunocompromised patients. To determine the drug susceptibilities of clinical Cryptococcus spp. isolates, the characteristics of 61 clinical Cryptococcus spp. complex isolates and their antifungal susceptibilities were investigated, including 52 C. neoformans and 9 C. gattii isolates collected at Shanghai between 1993 and 2009. Antifungal susceptibility of clinical isolates to amphotericin B, fluconazole, itraconazole, and flucytosine were determined by the microdilution method M27-A2 and the ATB FUNGUS 3 kit. This study investigated the prevalence of C. dubliniensis in a Brazilian family with an HIV - infected child. A total of 42 oral isolates were obtained from eight family members. The identification of C. dubliniensis was performed by polymerase chain reactions (PCR) using primers against a specific sequence of the C. dubliniensis cytochrome b gene. Only the HIV-infected child and his grandmother were colonized by C. dubliniensis. In this study C. dubliniensis isolated from the HIV-infected child exhibited high susceptibility for azoles tested with MICs of 0.125 and 0.5 µg/mL for voriconazole and fluconazole, respectively. Changes in hyphal morphology due to chitosan treatment in some fungal species. Repositório Alice

In this work, changes in the hyphal morphology due to chitosan treatment in some fungal species were studied. Scanning electron microscope (SEM) observations revealed that chitosans with molar fraction of acetyl groups (FA 0.16 and 0.18) and degree of polymerization (DP 1,089 and 1,242) had a direct effect on the morphology of the chitosan-treated fungi, reflecting its potential for causing a delay in the growth of Alternaria alternata (500 µg × mL-1), Botrytis cinerea (1,000 µg × mL-1), Penicillium expansum (1,000 µg × mL-1) and Rhizopus stolonifer (500 µg × mL-1). Mycelial aggregation and structural changes such as excessive branching, swelling of the cell wall and hyphae size reduction were observed in the micrographs.

Development of mutants of Coniothyrium minitans with improved efficiency for control of Sclerotinia sclerotiorum. Repositório Alice

Coniothurium minitans (CM) is hyperparasitic to Sclerotinia sderotiorum (55), a palhogen of many economically important crops. In this paper, we describe lhe isolation of improved mutants of CM, using a UV - irradiation regime, with altered chitinase production and tolerance to high concentration of iprodione, which are effect:iveagainst Ss. Three out of the 59 mutants obtained inhibited the mycelial growth of CM. Infectivity of sclerotia by the new mutants was assayed by the plant-tissue-based system using carrot segments. Different culture media applied to the study of Cryptococcus neoformans susceptibility to amphotericin B and fluconazole BJM

This study compared the performance of three culture media used for the determination of minimal inhibitory concentration (MIC) of amphotericin B and fluconazole against 48 Cryptococcus neoformans strains. For amphotericin B the media employed were RPMI 1640 medium, as indicated by NCCLS as reference, Yeast Nitrogen Base (YNB) as proposed by Ghannoum et al. (YNB-1), Antibiotic medium 3 (AM3) indicated by others, and YNB as proposed by us (YNB-2). In YNB-1, amphotericin B showed broader MIC ranges, varying from 0.125 µg/ml to 1.0 µg/ml; in RPMI 1640 medium and YNB-2 the MICs were similar and in AM3 the MIC ranges were narrower. The in vitro activity of antifungal and antiseptic agents were evaluated against dermatophytes isolated from patients with tinea pedis. The antifungals studied were: ciclopirox olamine, cetoconazole, tolciclate and terbinafine, and the antiseptics were: povidine iodine (PVPI), propolis, Fungol®, Andriodermol®, and boric acid. Several studies have reported the occurrence of infections caused by Candida yeasts as well as the increasing prevalence of non albicans species. The aim of the present work is focused on the obtaining of heteroresistance to amphotericin B and fluconazole in Candida species using two distinct methodologies: selection and induction. In this work we studied the susceptibility to heat and antifungal agents of the first strains of environmental Cryptococcus neoformans var neoformans (serotype D) isolated in the state of Rio Grande do Sul, Brazil. In order to achieve a rigorous analysis, we employed the methodology recommended by NCCLS, Yeast Nitrogen Base (YNB) proposed by Ghannoum et al (YNB-1), Antibiotic medium 3 (AM3) indicated by others, YNB adjusted to the NCCLS methodology (YNB-2) and Etest. Our results indicate that all strains were susceptible to amphotericin B (0.0625-0.5 µg/mL), fluconazole (0.125-8.0 µg/mL), itraconazole (0.031-0.25 µg/mL) and flucytosine (0.125-4.0 µg/mL). Malassezia pachydermatisis associated with dermatomycoses and otomycosis in dogs and cats. This study compared the susceptibility of M. pachydermatis isolates from sick (G1) and healthy (G2) animals to azole and polyene antifungals using the M27-A3 protocol. Isolates from G1 animals were less sensitive to amphotericin B, nystatin, fluconazole, clotrimazole and miconazole.

Use of the VITEK 2 system to identify and test the antifungal susceptibility of clinically relevant yeast species BJM

Eleven quality control isolates (Candida albicans ATCC 64548, C. tropicalis ATCC 200956, C. glabrata ATCC 90030, C. lusitaniae ATCC 200951, C. parapsilosis ATCC 22019, C. krusei ATCC 6258, C. dubliniensis ATCC 6330, Saccharomyces cerevisiae ATCC 9763, Cryptococcus neoformans ATCC 90012, C. gattii FIOCRUZ-CPF 60, and Trichosporon mucoides ATCC 204094) and 32 bloodstream isolates, including C. albicans, C. tropicalis, C. parapsilosis, C. glabrata, C. krusei, C. guilliermondii, C. pelliculosa (Pichia anomala), C. haemulonii, C. lusitaniae, and C. kefyr were identified at the species level by the VITEK 2 system.
