Asymptotic viscoacoustic diffraction tomography of ultrasonic laboratory data: a tool for rock properties analysis ArchiMer
Ribodetti, A; Operto, S; Virieux, J; Lambare, G; Valero, Hp; Gibert, D.
This paper presents an application of 2.5-D asymptotic viscoacoustic diffraction tomography to ultrasonic data recorded during a physically scaled laboratory experiment. This scaled experiment was used to test the reliability of our method when applied to a real data set to estimate the attenuation factor Q. Diffraction tomography relies on ray theory to compute the Green functions in a smooth background medium and on the Born approximation to linearize the relation between the scattered wavefield and the velocity and Q perturbations. The perturbations are inferred from the data by an iterative (linear) quasi-Newtonian algorithm. The inversion formula was specifically developed to account for the acquisition geometry designed in this study. The derivation...

Attenuation and immunogenicity of recombinant yellow fever 17D-dengue type 2 virus for rhesus monkeys BJMBR
Galler,R.; Marchevsky,R.S.; Caride,E.; Almeida,L.F.C.; Yamamura,A.M.Y.; Jabor,A.V.; Motta,M.C.A.; Bonaldo,M.C.; Coutinho,E.S.F.; Freire,M.S..
A chimeric yellow fever (YF)-dengue serotype 2 (dengue 2) virus was constructed by replacing the premembrane and envelope genes of the YF 17D virus with those from dengue 2 virus strains of Southeast Asian genotype. The virus grew to high titers in Vero cells and, after passage 2, was used for immunogenicity and attenuation studies in rhesus monkeys. Subcutaneous immunization of naive rhesus monkeys with the 17D-D2 chimeric virus induced a neutralizing antibody response associated with the protection of 6 of 7 monkeys against viremia by wild-type dengue 2 virus. Neutralizing antibody titers to dengue 2 were significantly lower in YF-immune animals than in YF-naive monkeys and protection against challenge with wild-type dengue 2 virus was observed in only 2...

Experimental infection of commercial layers with wild or attenuated Salmonella Gallinarum mutant strains: anatomic pathology, total blood cell count and serum protein levels Rev. Bras. Ciênc. Avic.
Garcia,KO; Berchieri Jr.,A; Santana,AM; Alarcon,MFF; Freitas Neto,OC; Fagliari,JJ.
The aim of the present study was to comparatively evaluate hemogram, blood serum components and anatomopathologic alterations in commercial layers experimentally challenged with an attenuated vaccine candidate strain (SG∆cobS∆cbiA) and other two pathogenic strains (SGDcobS and SGNalr) of Gallinarum (SG). In total, 280 commercial layers were randomly divided into 4 groups (G1, G2, G3 and G4). At five days of age, birds from groups G1 received approximately 107 colony forming units (CFU) of SGDcobS; meanwhile birds from group G2 and G3 received the same dose of SGNalr and SG∆cobS∆cbiA, respectively. Birds from G4 were not infected. At 24 hours before (DBI) and 24 hours after (1 DAI), and three (3 DAI), five (5 DAI), seven (7 DAI) ten (10 DAI), and fifteen...

GPM-Derived Climatology of Attenuation Due to Clouds and Precipitation at Ka-Band ArchiMer
Battaglia, Alessandro; Mroz, Kamil; Watters, Daniel; Ardhuin, Fabrice.
Attenuation from clouds and precipitation hinders the use of Ka-band in SARs, radar altimeters and in satellite link communications. The NASA-JAXA Global Precipitation Measurement (GPM) mission, with its core satellite payload including a dual-frequency (13.6 and 35.5 GHz) radar and a multifrequency passive microwave radiometer, offers an unprecedented opportunity for better quantifying such attenuation effects. Based on four years of GPM products, this article presents a global climatology of Ka-band attenuation caused by clouds and precipitation and analyses the impact of the precipitation diurnal cycle. As expected, regions of high attenuation mirror precipitation patterns. Clouds and precipitation cause two-way attenuation at 35.5 GHz in excess of 3 dB...

Limited replication of yellow fever 17DD and 17D-Dengue recombinant viruses in rhesus monkeys Anais da ABC (AABC)
Trindade,Gisela F.; Marchevsky,Renato S.; Fillipis,Ana M.B. de; Nogueira,Rita M.R.; Bonaldo,Myrna C.; Acero,Pedro C.; Caride,Elena; Freire,Marcos S.; Galler,Ricardo.
For the development of safe live attenuated flavivirus vaccines one of the main properties to be established is viral replication. We have used real-time reverse transcriptase-polymerase chain reaction and virus titration by plaque assay to determine the replication of yellow fever 17DD virus (YFV 17DD) and recombinant yellow fever 17D viruses expressing envelope proteins of dengue virus serotypes 2 and 4 (17D-DENV-2 and 17D-DENV-4). Serum samples from rhesus monkeys inoculated with YFV 17DD and 17D-DENV chimeras by intracerebral or subcutaneous route were used to determine and compare the viremia induced by these viruses. Viral load quantification in samples from monkeys inoculated by either route with YFV 17DD virus suggested a restricted capability of...

Safety and efficacy of a salmonella gallinarum ΔcobSΔcbiA strain with potential to prevent chicken infections by salmonella gallinarum and salmonella enteritidis Rev. Bras. Ciênc. Avic.
Paiva,JB; Penha Filho,RAC; Moura,BS; Berchieri Junior,A.
New vaccine design techniques have allowed the development of effective vaccine strains against Salmonella infections inwhich the risks of reversion to the wild type and virulence is null. The mutant strain Salmonella Gallinarum ΔcobSΔcbiA was previously shown to be avirulent in chickens. In this study, this strain was tested as a vaccine against Salmonella Gallinarum (SG) and S. Enteritidis (SE) infections, and its protection levels, safety and possible risks of reversion to virulence after vaccination of layers were evaluated. Birds were vaccinated at five days of age or at five and 25 days of age. At 45 days of age, brown and white layers were challenged with SG and SE wild strains, respectively. Two assays to test the possibility of reversion to...

The yellow fever 17D vaccine virus: molecular basis of viral attenuation and its use as an expression vector BJMBR
The yellow fever (YF) virus is the prototype flavivirus. The use of molecular techniques has unraveled the basic mechanisms of viral genome structure and expression. Recent trends in flavivirus research include the use of infectious clone technology with which it is possible to recover virus from cloned cDNA. Using this technique, mutations can be introduced at any point of the viral genome and their resulting effect on virus phenotype can be assessed. This approach has opened new possibilities to study several biological viral features with special emphasis on the issue of virulence/attenuation of the YF virus. The feasibility of using YF virus 17D vaccine strain, for which infectious cDNA is available, as a vector for the expression of heterologous...

Use of molecular markers can help to understand the genetic diversity of Babesia bovis. Repositório Alice
SIMAS, P. V. M; BASSETTO, C. C.; GIGLIOTI, R.; OKINO, C. H.; OLIVEIRA, H. N. de; OLIVEIRA, M. C. de S..
Cattle babesiosis is a tick-borne disease responsible for significant losses for the livestock industries in tropical areas of the world. These piroplasms are under constant control of the host immune system, which lead to a strong selective pressure for arising more virulent or attenuated phenotypes. Aiming to better understand the most critical genetic modifications in Babesia bovis genome, related to virulence, an in silico analysis was performed using DNA sequences from GenBank. Fourteen genes (sbp-2, sbp-4, trap, msa-1, msa-2b, msa-2c, Bv80 (or Bb-1), 18S rRNA, acs-1, ama-1, ?-tub, cp-2, p0, rap-1a) related to parasite infection and immunogenicity and ITS region were selected for alignment and comparison of several isolates of Babesia bovis from...

Validation of the TOPEX rain algorithm: Comparison with ground-based radar - art. no. 4038 ArchiMer
Mcmillan, A; Quartly, G; Srokosz, M; Tournadre, Jean.
[1] Recently developed algorithms have shown the potential recovery of rainfall information from spaceborne dual-frequency altimeters. Given the long mission achieved with TOPEX and the prospect of several other dual-frequency altimeters, we need to validate the altimetrically derived values so as to foster their integration with rain information from different sensors. Comparison with some alternative climatologies shows the bimonthly means for TOPEX to be low. Rather than apply a bulk correction we investigate individual rain events to understand the cause of TOPEX's underestimation. In this paper we compare TOPEX with near-simultaneous ground-based rain radars based at a number of locations, examining both the detection of rain and the quantitative...
