Amylase production by solid-state fermentation of agro-industrial wastes using Bacillus sp. BJM

Solid state fermentation was carried out using various agro- industrial wastes with the best amylase producing strain isolated from soil. Different physicochemical conditions were varied for maximum enzyme production. The strain produced about 5400 units/g of amylase at 1:3 moisture content, 20% inoculum, after 72 h of incubation with Mustard Oil seed cake as the substrate. The optimum temperature and pH of the enzyme activity were found to be 50ºC and 6 respectively. The enzyme was found to be thermostable at 70ºC for about 2 h without any salt. It showed stability at pH range 5-7. The metal ions as Na+, Ca++, Mg++ and Co++ enhanced the enzyme activity.

Application of bacterial and yeast biosurfactants for enhanced removal and biodegradation of motor oil from contaminated sand Electron. J. Biotechnol.

Background This study investigated the potential application of two biosurfactants for enhanced removal capability and biodegradation of motor oil contaminated sand under laboratory conditions. The biosurfactants were produced by the yeast Candida sphaerica and by the bacterium Bacillus sp. cultivated in low-cost substrates. The ability of removing motor oil from soil by the two biosurfactants was identified and compared with that of the synthetic surfactants Tween 80 and Triton X-100. Results Both crude and isolated biosurfactants showed excellent effectiveness on motor oil removal from contaminated sand under kinetic conditions (70-90%), while the synthetic surfactants removed between 55 and 80% of the oil. Bacterial treatment of alkaline cement kiln dust using Bacillus halodurans strain KG1 BJM

Abstract This study was conducted to isolate an acid-producing, alkaliphilic bacterium to reduce the alkalinity of cement industry waste (cement kiln dust). Gram-positive isolate KG1 grew well at pH values of 6–12, temperatures of 28–50 °C, and NaCl concentrations of 0–16% and thus was further screened for its potential to reduce the pH of an alkaline medium. Phenotypic characteristics of the KG1 isolate were consistent with those of the genus Bacillus, and the highest level of 16S rRNA gene sequence similarity was found with Bacillus halodurans strain DSM 497 (94.7%). Characterization of a novel lipase from Bacillus sp. isolated from tannery wastes BJM

Kinetics of a lipase isolated from Bacillus sp. was studied. The enzyme showed maximum activity at pH 9 and temperature 60ºC. The Michaelis constant (K M 0.31 µM) obtained from three different plots i.e., Lineweaver-Burk, Hanes-Wolf and Hofstee, was found to be lower than already reported lipases that confirmed higher affinity of the enzyme for its substrate p-NPL (p-nitrophenyl laurate). Vmax of the enzyme was found to be 7.6 µM/mL/min. Energy of activation calculated from Arrhenius plot was found to be 20.607 kJmol-1. Characterization of extracellular polymers synthesized by tropical intertidal biofilm bacteria ArchiMer

Aims: This study was performed to determine the potential of tropical intertidal biofilm bacteria as a source of novel exopolymers (EPS). Methods and Results: A screening procedure was implemented to detect EPS-producing biofilm bacteria. Isolates MC3B-10 and MC6B-22, identified respectively as a Microbacterium species and Bacillus species by 16S rDNA and cellular fatty acids analyses, produced different EPS, as evidenced by colorimetric and gas chromatographic analyses. The polymer produced by isolate MC3B-10 displays significant surfactant activity, and may chelate calcium as evidenced by spectroscopic analysis. Controle da mancha foliar de Eucalyptus grandis e Eucalyptus urophylla causada por Cylindrocladium sp. com Bacillus sp. Repositório Alice

Folhas destacadas, de plantas sadias de E. grandis e E. urophilla foram mantidas em placas de Petri de plastico contendo um disco de papel de filtro previamente umedecido com agua destilada, para estudar o controle de Cylindrocladium sp. usando um isolado de Bacillus sp., o qual apresentou grande eficiencia do crescimento micelial deste fungo in vitro. Culture conditions for the production of thermostable amylase by Bacillus sp BJM

Studies on the alpha-amylase production were carried out with a bacterial strain isolated from a soil sample. The cells were cultivated in a mineral medium containing soluble starch as sole carbon source. The addition of calcium (10 mM) or peptone (1%) and yeast extract (0.5%) to the mineral medium shortened the lag period and improved the growth and alpha-amylase synthesis. The addition of glucose to the culture diminished greatly the synthesis of alpha-amylase, demonstrating that a classical glucose effect is operative in this organism. Effect of the culture conditions on the production of an extracellular protease by thermophilic Bacillus sp and some properties of the enzymatic activity BJM

Protease production by thermophilic Bacillus sp strain SMIA-2 cultivated in liquid cultures containing 1% maltose as a carbon source and supplemented with whey protein (0.1%) and corn steep liquor (0.3%) reached a maximum at 14 h, with levels of 42 U/mg protein. The microorganism was capable of utilizing a wide range of carbon sources, but protease activity varied according the carbon source. Starch and maltose were the best carbon sources in the present study for protease secretion, while lactose and sucrose were less effective. Effect of the medium composition on formation of amylase by Bacillus sp BABT

Studies on the alpha -amylase synthesis was carried out with a moderately thermophilic, facultatively anaerobic Bacillus sp, isolated from soil samples. The cells were cultivated in a complex medium containing soluble starch or maltose as carbon source. The levels of the alpha -amylaseactivity detected in culture supernatants varied greatly with the type of carbon source used. Maltose, soluble starch and citrate stimulated alpha -amylaseformation. Addition of exogenous glucose repressed formation of alpha -amylase, demonstrating that a classical glucose effect was operative in this organism. A produção de poligalacturonase pelo termofílico Bacillus sp. SMIA-2, cultivado em meio líquido contendo pectina cítrica como única fonte de carbono, alcançou a sua máxima atividade enzimática em 30 horas, com níveis de 42 IU.mL-1. Entre as várias fontes orgânicas e inorgânicas de nitrogênio testadas, o sulfato de amônio foi a que proporcionou maior atividade da poligalacturonase. O aumento da concentração da pectina cítrica, no meio de cultura, acima de 0,5% não proporcionou um aumento da atividade da enzima. Produção de proteases por Bacillus sp SMIA-2 crescido em soro de leite e água de maceração de milho e compatibilidade das enzimas com detergentes comerciais Ciênc. Tecnol. Aliment.

A produção de proteases por Bacillus sp. SMIA-2 cultivado em um meio de cultura contendo soro de leite e água de maceração de milho foi estudada. Além disso, a compatibilidade da enzima com detergentes comerciais foi também avaliada. A atividade máxima da enzima (70 U/mg proteína) foi observada na fase estacionária de crescimento, com 32 h de incubação. Estudos sobre a caracterização da protease revelaram que a temperatura ótima para atividade desta enzima foi 70 °C e que a mesma manteve 91% de sua atividade quando incubada a 70 °C na presença do cálcio. O valor ótimo de pH encontrado para a protease foi 8,0, sendo que a enzima manteve 85% e 46% de sua atividade quando incubada por 1 h em pH 9 e pH 10 respectivamente. Production and properties of Xylanase from thermophilic Bacillus sp. BABT

An aerobic, thermophilic, xylanolytic bacterium was isolated from local soil. The results of 16S rRNA sequence comparisons indicated that the isolate was closely related to Bacillus caldoxylolyticus and Bacillus sp strain AK1. These organisms exhibited 94% levels of ribossomal DNA sequence homology. Studies on the xylanase characterisation from liquid cultures grown on beechwood xylan revealed that the enzyme retained 100% of activity for 2 hours at temperatures ranging from 30 to 50º C, while at 60, 70 and 100º C, 10%, 11% and 29% of the original activities were lost, respectively. Properties of an amylase from thermophilic Bacillus SP. BJM

alpha-Amylase production by thermophilic Bacillus sp strain SMIA-2 cultivated in liquid cultures containing soluble starch as a carbon source and supplemented with 0.05% whey protein and 0.2% peptone reached a maximum activity at 32 h, with levels of 37 U/mL. Studies on the amylase characterization revealed that the optimum temperature of this enzyme was 90ºC. The enzyme was stable for 1 h at temperatures ranging from 40-50ºC while at 90ºC, 66% of its maximum activity was lost. Protease with collagenolytic activity produced by Bacillus sp. DPUA 1728 from Amazonian soil BJM

Qualitative analyses were carried out on solid medium with insoluble collagen 0.25% (w/v) to detect proteases with collagenolytic activity produced by Bacillus sp. In cultures incubated for 24 h, a 23 full factorial design with four repetitions at the center point was developed to analyze the effects and interactions between initial pH, temperature and the concentration of gelatin. Based on the results of the first 23 full factorial design, a successive 23 full factorial design was performed. The most favorable production conditions were found to be 1.5% (w/v) gelatin, pH 9.0 and 37 °C with enzymatic activity of 86.27 U/mL. Screening of bacterial strains for pectinolytic activity: characterization of the polygalacturonase produced by Bacillus sp Rev. Microbiol.

One hundred sixty eight bacterial strains, isolated from soil and samples of vegetable in decomposition, were screened for the use of citrus pectin as the sole carbon source. 102 were positive for pectinase depolymerization in assay plates as evidenced by clear hydrolization halos. Among them, 30% presented considerable pectinolytic activity. The cultivation of these strains by submerged and semi-solid fermentation for polygalacturonase production indicated that five strains of Bacillus sp produced high quantities of the enzyme. The physico-chemical characteristics, such as optimum pH of 6.0 - 7.0, optimum temperatures between 45oC and 55oC, stability at temperatures above 40oC and in neutral and alkaline pH, were determined.
