A novel expression vector for the secretion of abaecin in Bacillus subtilis BJM

ABSTRACT This study aimed to describe a Bacillus subtilis expression system based on genetically modified B. subtilis. Abaecin, an antimicrobial peptide obtained from Apis mellifera, can enhance the effect of pore-forming peptides from other species on the inhibition of bacterial growth. For the exogenous expression, the abaecin gene was fused with a tobacco etch virus protease cleavage site, a promoter Pglv, and a mature beta-glucanase signal peptide. Also, a B. subtilis expression system was constructed. The recombinant abaecin gene was expressed and purified as a recombinant protein in the culture supernatant. Actividad antifúngica de extractos crudos de Bacillus subtilis contra fitopatógenos de soja (Glycine max) y efecto de su coinoculación con Bradyrhizobium japonicum Agrociencia

Entre los microorganismos causantes de enfermedades pre y post cosecha en los cultivos de soja (Glycine max) están las especies fúngicas Fusarium solani y Pythium sp. Cuatro cepas del género Bacillus (B. subtilis ATCC6633, B. amylolyticus, B. subtilis var. natto, B. subtilis var. natto domesticado) se probaron para evaluar su respuesta inhibitoria en los hongos fitopatógenos mencionados. Los tratamientos de inoculación fueron: 1) semillas inoculadas con Bradyrhizobium japonicum y 2) semillas coinoculadas con B. japonicum y B. subtilis. Se evaluó el control in vitro de Burkholderia cepacia (cepa CCIBP-E-556), Serratia plymuthica (cepa ATCC 1270) y Bacillus subtilis (cepa ATCC 6051), sobre Macrophomina phaseolina (Tassi) Goid, utilizando el método de cultivo dual.Se midió el radio de la colonia del hongo y se determinó el porcentaje de inhibición del crecimiento radial. B. cepacia produjo el mayor efecto sobre el aislado de M. phaseolina, provocando un porcentaje de inhibición del 43,5%, y ocasionando cambios en la apariencia del micelio, que se observaron en la zona de inhibición con el uso del microscópico óptico. Antibiosis and dark-pigments secretion by the phytopathogenic and environmental fungal species after interaction in vitro with a Bacillus subtilis isolate BABT

In this work, different reactions in vitro between an environmental bacterial isolate and fungal species were related. The Gram-positive bacteria had terminal and subterminal endospores, presented metabolic characteristics of mesophilic and acidophilic growth, halotolerance, positive to nitrate reduction and enzyme production, as caseinase and catalase. The analysis of partial sequences containing 400 to 700 bases of the 16S ribosomal RNA gene showed identity with the genus Bacillus. Lipopeptides represent a class of microbial surfactants with increasing scientific, therapeutic and biotechnological interests. The genus Bacillus is a producer of these active compounds, and among them B. subtilis produces surfactin, the most potent biosurfactant known. These compounds can act as antibiotics, antivirals, antitumorals, immunomodulators and enzyme inhibitors. Avaliação da biodegradação de parafinas e da produção de biosurfactante por Bacillus subtilis na presença de petróleo BJM

Os experimentos com Bacillus subtilis para avaliação da tensão superficial foram realizados com meio de cultivo contendo como nutrientes básicos 0,4% de ions nitrato e 4% de glicose, na presença de petróleo. A produção de surfactina foi observada pela redução da tensão superficial do meio de cultura fermentado. Surfactina foi isolada a partir do meio de cultura fermentado por B. subtilis, por precipitação ácida seguida de extração com clorofórmio-metanol. The aim of this work was to examine the changes that occur in cells of the fungus Fonsecaea pedrosoi after in vitro co-culturing with Bacillus subtilis and to explore the results of this interaction in vivo in an experimental murine infection. The aim of the present study was to isolate autochthonous microorganisms with biosurfactants capacities from the rhizospere of the cotton (Gossypium spp.), cachanilla (Pluchea sericea) and salicornia (Salicornia bigelovii) in the Mexicali valley. The biosurfactant activity and biosurfactant productions by the strains isolated from the rhizosphere of the cotton (Bs-Alg), cachanilla (Bs-Cach) and salicornia (Bs-Cach01) were determined using oil spreading technique and emulsifcation activity with corn, olive, soybean and diesel oils, respectively. Mediante experimentos de laboratorio e invernadero se analizó la bioeficacia de cinco cepas de Bacillus spp.; de la mezcla de los bioproductos quitosan y extracto de Larrea tridentata, así como de cinco funguicidas sintéticos contra el hongo Alternaria dauci Kühn. Además, se determinó el efecto de estos compuestos en el crecimiento del cultivo de zanahoria. The objective of this work was to evaluate the addition of Bacillus subtilis probiotic to the feed of common snook (Centropomus undecimalis) fingerlings, in alternate and continuous regimens. Six hundred and sixty fish, with average length of 5.90±0.88 cm and weight of 1.92±0.28 g, were stocked in 12 cages of 1.0 m3, with 55 fish each. Comparison of plant growth-promotion with Pseudomonas aeruginosa and Bacillus subtilis in three vegetables BJM

Our objective was to compare some plant growth promoting rhizobacteria (PGPR) properties of Bacillus subtilis and Pseudomonas aeruginosa as representatives of their two genera. Solanum lycopersicum L. (tomato), Abelmoschus esculentus (okra), and Amaranthus sp. (African spinach) were inoculated with the bacterial cultures. At 60 days after planting, dry biomass for plants treated with B. subtilis and P. aeruginosa increased 31% for tomato, 36% and 29% for okra, and 83% and 40% for African spinach respectively over the non-bacterized control. Considering all the parameters tested, there were similarities but no significant difference at P < 0.05 between the overall performances of the two organisms.

As a precursor, pimelic acid plays an important role in biotin biosynthesis pathway of Bacillus subtilis. Fermentations supplemented with pimelic acid could improve the production of biotin, however, with a disadvantage-high cost. So it is necessary to improve the biosynthesis of pimelic acid via genetic engineering in B. subtilis. Objetivou-se, com este trabalho, avaliar o efeito de probiótico sobre o desempenho e qualidade de ovos de poedeiras. A suplementação de probiótico foi iniciada na fase de recria e os efeitos foram avaliados de 50 a 66 semanas. O delineamento experimental adotado foi inteiramente ao acaso com quatro tratamentos (suplementação de probiótico na fase de recria e postura, suplementação de probiótico somente na fase de recria, suplementação de probiótico somente na fase de postura e ausência de suplementação) e seis repetições de oito aves cada. This work examines the economic advantages of probiotic use in the diet of Nile tilapia broodstock during the reproductive period. For this purpose, Bacillus subtilis was applied as a feed additive. The experimental design was completely randomized with three treatment groups: the T0 control (without probiotic), the T1 continuous probiotic intake, and the T2 alternate probiotic intake at a dose of 0.50 g kg-1 of feed (1010 CFU g-1) with four replicates. For the reproduction assay, 118 females and 48 males of Nile tilapia (proportion 4 males:9 females. hapa-1) (weight 527.65 g ± 185.98 g and length 30.16 cm ± 3.57 cm) were distributed into 12 hapas (3.5 × 2.0 × 1.5 m).
