The phytopathogenic fungus Moniliophthora perniciosa (Stahel) Aime & Philips-Mora, causal agent of witches' broom disease of cocoa, causes countless damage to cocoa production in Brazil. Molecular studies have attempted to identify genes that play important roles in fungal survival and virulence. In this study, sequences deposited in the M. perniciosa Genome Sequencing Project database were analyzed to identify potential biological targets. ABSTRACT Twenty-seven Tritirachium strains were present in the URM culture collection originally founded in Brazil by Augusto Chaves Batista. Fifteen freshly-prepared cultures were obtained from these original strains preserved under mineral oil. DNA was extracted for analysing phylogenetic relationships using the sequence information available from Tritirachium type materials and reference strains. Phylogenetic analysis using the internal transcribed spacer (ITS) ribosomal DNA sequences revealed that eight of these strains belong to the same clade as of T. oryzae and that the four other strains belong to the same clade as of T. candoliense. We analyzed specimens identified as Amanita muscaria, some recently collected and others already deposited in herbaria, in Brazil. We concluded that two subspecies of A. muscaria occur in Brazil: A. muscaria var. muscaria; and A.muscaria var. flavivolvata. The first taxon was found in association with Castanea sativa, and the second (one specimen only) was found in association with Pinus and Eucalyptus spp. Morphologically, A. muscaria var. flavivolvata is distinguished by a shallower subhymenium and by basidiospores that are more elongated than are those of A. muscaria var. muscaria, which is the more widely known subspecies. The basidiospores of Pisolithus sp. contain large amounts of lipids, indicating provision for future germination in the host rhizosphere. However, the accumulation, composition, and mobilization of lipids during formation and germination of these spores are largely unknown. In this study, lipid storage and fatty acid composition during basidiosporogenesis were analyzed in fresh basidiocarps using bright-field microscopy and gas chromatography. Abundant lipid bodies are found in the hyphae, basidia, and basidiospores of fungal basidiocarps. This evidences a considerable C transport in the basidiocarp to meet the C demand during basidiospore formation.
