Bradyrhizobium elkanii is successfully used in the formulation of commercial inoculants and, together with B. japonicum, it fully supplies the plant nitrogen demands. Despite the similarity between B. japonicum and B. elkanii species, several works demonstrated genetic and physiological differences between them. In this work Representational Difference Analysis (RDA) was used for genomic comparison between B. elkanii SEMIA 587, a crop inoculant strain, and B. japonicum USDA 110, a reference strain. Two hundred sequences were obtained. From these, 46 sequences belonged exclusively to the genome of B. elkanii strain, and 154 showed similarity to sequences from B. japonicum genome. From the 46 sequences with no similarity to sequences from B. japonicum, 39... |

In order to contribute for the optimization of biological nitrogen fixation (BNF) associated with cowpea in Cerrado areas in the Northeast region of Brazil, this work aimed to analyze the diversity of rhizobial populations in eight areas of Cerrado, during a soybean and rice-cowpea rotation. Morphological traits (mucous production and colony morphology), genotypic analyzes (ARDRA 16S) and intrinsic resistance to antibiotics were determined for a collection of isolates captured using cowpea as a host-plant. The morphological data showed a inverse correlation (p < 0.05) between the number of legume (soybean and cowpea) crops, according to the history of each area, and rhizobium diversity, estimated by the Shannon-Weaver index. ARDRA data showed that... |

Abstract: The objective of this work was to assess the effects of 1-methylcyclopropene (1-MCP) on traits related with biological nitrogen fixation on 'BRS 268' soybean (Glycine max) subjected to water restriction. Plants were either exposed to drought between 32 (R2) and 47 (R3) days after sowing (DAS) or kept well-watered, in combination with exposure or not to 1-MCP. On the second day under drought (34 DAS), plants from both water conditions were exposed to 1-MCP in a hermetically sealed chamber for 15 hours. Control plants, dry or well-watered, that were not exposed to 1-MCP were kept in a separate chamber. At 36 (R2) and 47 (R3) DAS, shoot and root dry weights, leaf area index, number and dry weight of nodules, total ureides in sap, and N concentration... |

Abstract A successful symbiotic relationship between soybean [Glycine max (L.) Merr.] and Bradyrhizobium species requires expression of the bacterial structural nod genes that encode for the synthesis of lipochitooligosaccharide nodulation signal molecules, known as Nod factors (NFs). Bradyrhizobium diazoefficiens USDA 110 possesses a wide nodulation gene repertoire that allows NF assembly and modification, with transcription of the nodYABCSUIJnolMNOnodZ operon depending upon specific activators, i.e., products of regulatory nod genes that are responsive to signaling molecules such as flavonoid compounds exuded by host plant roots. Central to this regulatory circuit of nod gene expression are NodD proteins, members of the LysR-type regulator family. In... |

To evaluate the interaction between foliar application of salicylic acid and Bradyrhizobium inoculation on the morphophysiology of cowpea under water stress conditions, four genotypes (BRS Rouxinol, BRS Marataoã, BRS Aracê and BR 17 Gurguéia) were subjected to five combinations of water availability: 100% replacement of crop evapotranspiration (control); 50% replacement of crop evapotranspiration (water stress); water stress + salicylic acid; water stress + Bradyrhizobium inoculation; and water stress + salicylic acid + Bradyrhizobium inoculation. The experiment was set up in a 4 × 5 factorial randomized block design, with four replicates and four plants per plot. Water stress negatively affected the leaf water potential, growth, proline contents and... |

The peanut plant has high plasticity and great adaptability to adverse conditions, including drought. To mitigate the negative effects of drought on legumes, nitrogen-fixing microorganisms have been investigated in some plant species, such as soybeans and beans. We analyzed the role Bradyrhizobium strains in peanut genotypes subjected to water deficit by means of plant growth, physiological and gene expression analysis. The research was conducted under greenhouse conditions with the runner peanut genotypes (IAC Runner 886, 2012-33 and 2012-47) and two Bradyrhizobium strains (ESA 123 and SEMIA 6144). After 20 days of germination, the water supply was completely interrupted and gas exchange analysis were carried out using an infrared gas analyzer, up to the... |

Drought is one of the environmental factors that most affects peanut cultivation in semi-arid regions, resulting in economic losses to growers. However, growth promoting bacteria are able to reduce water deficit damage in some plant species. In this context, this study aimed to evaluate the interaction of Bradyrhizobium strains reducing water stress effects on peanut genotypes by antioxidant enzymes activities, leaf gas exchanges and vegetative growth, as well as to determine the taxonomic positioning of strain ESA 123. The 16S rRNA gene of ESA 123 was amplified by PCR and sequenced by dideoxy Sanger sequencing method. An experiment was performed in greenhouse with three peanut genotypes (BRS Havana, CNPA 76 AM and 2012-4), two Bradyrhizobium strains... |

The co-inoculation of Bradyrhizobium with other non-bradyrhizobial strains was already assessed on cowpea, but the co-inoculation of two Bradyrhizobium strains was not tested up to now. This study aimed to evaluate the cowpea growth, N accumulation, and Bradyrhizobium competitiveness of the elite strain B. pachyrhizi BR 3262 when co-inoculated with other efcient Bradyrhizobium from the Brazilian semiarid region. Three potted-plant experiments were carried out. In the frst assay, 35 efcient Bradyrhizobium isolates obtained from the semiarid region of Brazil were co-inoculated with the elite strains B. pachyrhizi BR 3262. The experiment was conducted in gnotobiotic conditions. The plant growth, nodulation, N nutritional variables, and nodular occupation were... |
