A novel hot-plate test sensitive to hyperalgesic stimuli and non-opioid analgesics BJMBR
Lavich,T.R.; Cordeiro,R.S.B.; Silva,P.M.R.; Martins,M.A..
It is widely accepted that the classical constant-temperature hot-plate test is insensitive to cyclooxygenase inhibitors. In the current study, we developed a variant of the hot-plate test procedure (modified hot-plate (MHP) test) to measure inflammatory nociception in freely moving rats and mice. Following left and right hind paw stimulation with a phlogogen and vehicle, respectively, the animals were placed individually on a hot-plate surface at 51ºC and the withdrawal latency for each paw was determined simultaneously in measurements performed at 15, 60, 180, and 360 min post-challenge. Plantar stimulation of rats (250 and 500 µg/paw) and mice (125-500 µg/paw) with carrageenan led to a rapid hyperalgesic response of the ipsilateral paw that reached a...

Rheumatoid arthritis is a chronic inflammatory joint disease associated with the development of oxidative stress and inflammation. The safety and efficacy profile of 85% methanolic extract of Corallocarpus epigaeus (CE) was evaluated in the present study. In safety profile LD50 value was determined by carrying out an acute toxicity study. In efficacy profile, the analgesic activity was evaluated by both hot plate and tail immersion tests. The anti-inflammatory activity was assessed by carrageenan-induced paw edema and anti-arthritic effect by complete Freund's adjuvant induced arthritis. Phytochemical screening of different CE extracts and quantitative analysis of both raw herb and 85% methanolic extract have been also carried out. The methanolic extract...

Development and optimization of a mixed beverage made of whey and water-soluble soybean extract flavored with chocolate using a simplex-centroid design Ciênc. Tecnol. Aliment.
OLIVEIRA,Dóris Faria de; GRANATO,Daniel; BARANA,Ana Cláudia.
Abstract This study aimed to combine the nutritional advantages of whey and soybean by developing a type of chocolate beverage with water-soluble soybean extract dissolved in whey. Different concentrations of thickeners (carrageenan, pectin and starch – maximum level of 500 mg.100 mL-1) were tested by a simplex-centroid design. Several physicochemical, rheological, and sensory properties of the beverages were measured and a multi-response optimization was conducted aiming to obtain a whey and soybean beverage with increased overall sensory impression and maximum purchase intention. Beverages presented mean protein levels higher than 3.1 g.100 mL-1, a low content of lipids (< 2 g.100 mL-1) and total soluble solids ≥20 g.100 mL-1. Response surface...

Effect of acupuncture on TNF-alpha, IL-1b and IL-10 concentrations in the peritoneal exudates of carrageenan-induced peritonitis in rats Ciência Rural
Scognamillo-Szabó,Márcia Valéria Rizzo; Bechara,Gervásio Henrique; Cunha,Fernando de Queiroz.
Acupuncture is an ancient and empirical therapeutic procedure known by its efficacy in the treatment of pain. However, the influence of acupuncture on inflammatory process is still poorly understood and additional research is needed. In this work, we investigated the mechanism of action of manual acupuncture on the inhibition of neutrophil migration to the peritoneal cavity induced by the inflammatory stimulus carrageenan in Wistar rats. Previous results from our laboratory showed that this anti-inflammatory effect is not due to endogenous corticoid release. Furthermore, the concentration of IL-1b, but not of TNF-alpha or IL-10 in the carrageenan-induced exudates was reduced in the acupuncture group. Further research...

Effect of addition of different hydrocolloids on pasting, thermal, and rheological properties of cassava starch Ciênc. Tecnol. Aliment.
Leite,Tatiana Dias; Nicoleti,Joel Fernando; Penna,Ana Lúcia Barretto; Franco,Célia Maria Landi.
Starches and gums are hydrocolloids frequently used in food systems to provide proper texture, moisture, and water mobility. Starch-gum interaction in food systems can change the starch granule swelling and its gelatinization and rheological properties. In this study, the effect of the addition of xanthan gum (XG), sodium carboxymethyl cellulose (SCMC), and carrageenan (CAR) at the concentrations of the 0.15, 0.25, 0.35 and 0.45% (w/v) on the pasting, thermal, and rheological properties of cassava starch was studied. The swelling power (SP) and the scanning electron microscopy (SEM) of the starch gels were also evaluated. The results obtained showed that xanthan gum (XG) had a strong interaction with the cassava starch penetrating between starch granules...

Evolutionary Evidence of Algal Polysaccharide Degradation Acquisition by Pseudoalteromonas carrageenovora 9(T) to Adapt to Macroalgal Niches ArchiMer
Gobet, Angelique; Barbeyron, Tristan; Matard-mann, Maria; Magdelenat, Ghislaine; Vallenet, David; Duchaud, Eric; Michel, Gurvan.
About half of seaweed biomass is composed of polysaccharides. Most of these complex polymers have a marked polyanionic character. For instance, the red algal cell wall is mainly composed of sulfated galactans, agars and carrageenans, while brown algae contain alginate and fucose-containing sulfated polysaccharides (FCSP) as cell wall polysaccharides. Some marine heterotrophic bacteria have developed abilities to grow on such macroalgal polysaccharides. This is the case of Pseudoalteromonas carrageenovora 9(T) (ATCC 43555(T)), a marine gammaproteobacterium isolated in 1955 and which was an early model organism for studying carrageenan catabolism. We present here the genomic analysis of P. carrageenovora. Its genome is composed of two chromosomes and of a...

Hind limb ischemic preconditioning induces an anti-inflammatory response by remote organs in rats BJMBR
Souza Filho,M.V.P.; Loiola,R.T.; Rocha,E.L.; Simão,A.F.L.; Gomes,A.S.; Souza,M.H.L.P.; Ribeiro,R.A..
Ischemic preconditioning (IPC), a strategy used to attenuate ischemia-reperfusion injury, consists of brief ischemic periods, each followed by reperfusion, prior to a sustained ischemic insult. The purpose of the present study was to evaluate the local and systemic anti-inflammatory effects of hind limb IPC in male Wistar rat (200-250 g) models of acute inflammation. IPC was induced with right hind limb ischemia for 10 min by placing an elastic rubber band tourniquet on the proximal part of the limb followed by 30 min of reperfusion. Groups (N = 6-8) were submitted to right or left paw edema (PE) with carrageenan (100 µg) or Dextran (200 µg), hemorrhagic cystitis with ifosfamide (200 mg/kg, ip) or gastric injury (GI) with indomethacin (20 mg/kg, vo)....

Long-term ethanol intoxication reduces inflammatory responses in rats BJMBR
Carvalho,E.M.; Brito,G.A.C.; Pessoa,B.B.G.P.; Ribeiro,R.A.; Capaz,F.R..
The anti-inflammatory effects of long-term ethanol intoxication were determined during ethanol treatment and withdrawal on the basis of neutrophil and eosinophil migration, hind paw edema and mast cell degranulation. Male Wistar rats (180-200 g, around 2 months of age) were exposed to increasing concentrations of ethanol vapor over a 10-day period. One group was evaluated immediately after exposure (treated group - intoxicated), and another was studied 7 h later (withdrawal group). Ethanol inhalation treatment significantly inhibited carrageenan- (62% for the intoxicated group, N = 5, and 35% for the withdrawal group, N = 6) and dextran-induced paw edema (32% for intoxicated rats and 26% for withdrawal rats, N = 5 per group). Ethanol inhalation...

Participation of the NO/cGMP/K+ATP pathway in the antinociception induced by Walker tumor bearing in rats BJMBR
Barbosa,A.L.R.; Pinheiro,C.A.; Oliveira,G.J.; Torres,J.N.L.; Moraes,M.O.; Ribeiro,R.A.; Vale,M.L.; Souza,M.H.L.P..
Implantation of Walker 256 tumor decreases acute systemic inflammation in rats. Inflammatory hyperalgesia is one of the most important events of acute inflammation. The L-arginine/NO/cGMP/K+ATP pathway has been proposed as the mechanism of peripheral antinociception mediated by several drugs and physical exercise. The objective of this study was to investigate a possible involvement of the NO/cGMP/K+ATP pathway in antinociception induced in Walker 256 tumor-bearing male Wistar rats (180-220 g). The groups consisted of 5-6 animals. Mechanical inflammatory hypernociception was evaluated using an electronic version of the von Frey test. Walker tumor (4th and 7th day post-implantation) reduced prostaglandin E2- (PGE2, 400 ng/paw; 50 µL; intraplantar injection)...

Restorative Effects of A Newly Synthesized Peptide, Obiopeptide-1, in Cyclophosphamide- or Carrageenan-pretreated Mice Infected with Opportunistic Bacteria OAK
Fujii, Yasuhiro; Maki, Yoshiyuki; Ito, Masanari; Saito, Atsushi; Igarashi, Ikuo; Ono, Kenichiro; Itoh, Kikuji; Suzuki, Naoyoshi.
Large numbers of cyclophosphamide- or carrageenan-pretreated immunosuppressed adult ddY mice died within 10 days after inoculation with several species of opportunistic bacteria, including P. aeruginosa, K. pneumoniae, E. coli, S. aureus and methicillin resistant S. aureus. When immunosuppressed mice were administered bacteria in combination with two 100 μg/mouse intramuscular doses of a newly synthesized peptide, Obiopeptide-1 (OP-1), survival rates increased significantly. At 24 and 48 hours after intraperitoneal inoculation with P. aeruginosa, counts of viable organisms from the livers, spleens, lungs, hearts, and kidneys of mice that were administered cyclophosphamide in combination with OP-1 were significantly lower than counts from mice that were...
