A study of chlorophyll-like and phycobilin pigments in the C endosymbiont of the apple- snail Pomacea canaliculata Biocell
Vega,Israel A.; Dellagnola,Federico A.; Hurst,Jorge A.; Godoy,Martín S.; Castro-Vazquez,Alfredo.
Pigments present in the brown-greenish C morph of an intracellular endosymbiont of Pomacea canaliculata were investigated. Acetone extracts of the endosymbiotic corpuscles showed an absorption spectrum similar to that of chlorophylls. Three fractions obtained from silica gel column chromatography of the acetone extracts (C I, C II and C III), were studied by positive ion fast atom bombardment-mass spectrometry (FAB-MS) and hydrogen-nuclear magnetic resonance (H-NMR). Results indicated the presence of (1) a sterol in the yellow colored C I fraction; (2) a mixture of pheophorbides a and b in the major green fraction, C II; and (3) a modified pheophorbide a in the smaller green fraction, C III. Aqueous extracts of the C endosymbiont did not show evidence of...

Analysis of chlorophyll fluorescence reveals stage specific patterns of chloroplast-containing cells during Arabidopsis embryogenesis Biol. Res.
TEJOS,RICARDO I; MERCADO,ANA V; MEISEL,LEE A.
The basic body plan of a plant is established early in embryogenesis when cells differentiate, giving rise to the apical and basal regions of the embryo. Using chlorophyll fluorescence as a marker for chloroplasts, we have detected specific patterns of chloroplast-containing cells at specific stages of embryogenesis. Non-randomly distributed chloroplast-containing cells are seen as early as the globular stage of embryogenesis in Arabidopsis. In the heart stage of embryogenesis, chloroplast containing cells are detected in epidermal cells as well as a central region of the heart stage embryo, forming a triangular septum of chloroplast-containing cells that divides the embryo into three equal sectors. Torpedo stage embryos have chloroplast-containing...

Changes in phosphate content and phosphatase activities in rice seedlings exposed to arsenite Braz. J. Plant Physiol.
Mishra,Shruti; Dubey,R.S..
The effect of arsenite (As2O3) in situ on the level of the phosphate pool and activities of phosphohydrolytic enzymes was examined in rice (Oryza sativa L.) seedlings grown for 5-20 d in sand cultures. The effects were manifested via a decline in phosphate content and inhibition of the activities of key phosphatases. Application of 50 µM As2O3 in situ resulted in 34 to 77% inhibition of acid phosphatase activity in roots and about 38 to 50% inhibition of activity in shoots of 15-20-d-old seedlings. Similarly, alkaline phosphatase activity was inhibited in shoots under in situ As (III) toxicity. Varietal as well as organ specific differences were observed in the response of inorganic pyrophosphatase activity to in situ As (III) treatment. A moderately toxic...

Changes in the ultrastructure of soybean cultivars in response to manganese supply in solution culture Scientia Agricola
Lavres Junior,José; Reis,André Rodrigues; Rossi,Mônica Lanzoni; Cabral,Cleusa Pereira; Nogueira,Neusa de Lima; Malavolta,Eurípedes.
The deleterious effects of Mn stress on many species have been studied, mainly concerning biochemical, physiological and growth parameters of plants. However, there are few studies relating the anatomical and ultrastructural changes in response to manganese (Mn) nutritional disorders, This study examined the leaf ultrastructure of Mn-inefficient (IAC-15, Santa Rosa) and Mn-efficient (IAC-Foscarin 31) soybean (Glycine max L.) genotypes in response to three rates of Mn (0.5, 2 and 200 µmol L-1) in the nutrient solution. Symptoms of Mn deficiency developed 12 days after transplanting in IAC-15 and Santa Rosa, followed by IAC-Foscarin 31 on the 15th day. Only IAC-15 and Santa Rosa leaves showed symptoms of Mn toxicity. The Mn concentration in leaves ranged...

Gene pool sharing and genetic bottleneck effects in subpopulations of Eschweilera ovata (Cambess.) Mart. ex Miers (Lecythidaceae) in the Atlantic Forest of southern Bahia, Brazil Genet. Mol. Biol.
Santos,Alesandro S.; Borges,Daniela B.; Vivas,Caio V.; Berg,Cassio Van Den; Rodrigues,Polliana S.; Tarazi,Roberto; Gaiotto,Fernanda Amato.
Abstract Forest loss and fragmentation are the main threats to the maintenance of the Atlantic Forest, an important global biodiversity hotspot. Because of the current critical level of deforestation, ecological corridors are needed to facilitate species dispersion and gene flow among fragments. This study was conducted to investigate the genetic variability and gene pool sharing of Eschweilera ovata in five forest remnants in southern Bahia, Brazil using nuclear simple sequence repeat (nSSR) and plastid simple sequence repeat (cpSSR) microsatellite markers. cpSSR marker analysis revealed the domains of four haplotypes, showing that 80% of the individuals had only four maternal origins, reflecting a founder effect and/or genetic bottleneck. The results of...

Isolation and intracellular localization of insulin-like proteins from leaves of Bauhinia variegata BJMBR
Azevedo,C.R.; Maciel,F.M.; Silva,L.B.; Ferreira,A.T.S.; da Cunha,M.; Machado,O.L.T.; Fernandes,K.V.S.; Oliveira,A.E.A.; Xavier-Filho,J..
Evidence based on immunological cross-reactivity and anti-diabetic properties has suggested the presence of insulin-like peptides in plants. The objective of the present study was to investigate the presence of insulin-like proteins in the leaves of Bauhinia variegata ("pata-de-vaca", "mororó"), a plant widely utilized in popular medicine as an anti-diabetic agent. We show that an insulin-like protein was present in the leaves of this plant. A chloroplast protein with a molecular mass similar to that of bovine insulin was extracted from 2-mm thick 15% SDS-PAGE gels and fractionated with a 2 x 24 cm Sephadex G-50 column. The activity of this insulin-like protein (0.48 mg/mL) on serum glucose levels of four-week-old Swiss albino (CF1) diabetic mice was...

Salt stress affects mRNA editing in soybean chloroplasts Genet. Mol. Biol.
Rodrigues,Nureyev F.; Fonseca,Guilherme C. da; Kulcheski,Franceli R.; Margis,Rogério.
Abstract Soybean, a crop known by its economic and nutritional importance, has been the subject of several studies that assess the impact and the effective plant responses to abiotic stresses. Salt stress is one of the main environmental stresses and negatively impacts crop growth and yield. In this work, the RNA editing process in the chloroplast of soybean plants was evaluated in response to a salt stress. Bioinformatics approach using sRNA and mRNA libraries were employed to detect specific sites showing differences in editing efficiency. RT-qPCR was used to measure editing efficiency at selected sites. We observed that transcripts of NDHA, NDHB, RPS14 and RPS16 genes presented differences in coverage and editing rates between control and salt-treated...

The chloroplast genome of the desiccation-tolerant moss Pseudocrossidium replicatum (Taylor) R.H. Zander Genet. Mol. Biol.
Cevallos,Miguel A.; Guerrero,Gabriela; Ríos,Selma; Arroyo,Analilia; Villalobos,Miguel Angel; Porta,Helena.
Abstract Mosses in conjunction with hornworts and liverworts are collectively referred to as bryophytes. These seedless, nonvascular plants are the closest extant relatives of early terrestrial plants and their study is essential to understand the evolutionary first steps of land plants. Here we report the complete chloroplast (cp) genome sequence of Pseudocrossidium replicatum, a moss belonging to the Pottiaceae family that is common in the central highlands of Mexico, in South America, in southern USA, and in Kenia. The cp genome (plastome) of P. replicatum is 123,512 bp in size, comprising inverted repeats of 9,886 bp and single-copy regions of 85,146 bp (LSC) and 18,594 bp (SSC). The plastome encodes 82 different proteins, 31 different tRNAs, and 4...
