We examined the effects of Amylomyces rouxii, which is a mold found in some fermented foods in Indonesia, on serum cholesterol and hepatic LDL receptor mRNA in rats. Rats were fed a 0.5% cholesterol-enriched diet with (A. rouxii group) or without (control group) 30g/kg A. rouxii for 4 wk. There were no significant differences in the body weight, food intake or liver weight among the groups. However, the weight of the cecum in the A. rouxii-fed group was significantly higher than that in the control group. The cecal pH in the A. rouxii-fed group was significantly lower than that in the control group. Cecal acetic acid, propionic acid and total SCFA concentrations in the A. rouxii-fed group were significantly higher than those in the control group. The serum...

Atherogenic lipid profile of Brazilian near-term newborns BJMBR
Pardo,I.M.C.G.; Geloneze,B.; Tambascia,M.A.; Barros-Filho,A.A..
Cardiovascular disease is the primary cause of death in Brazil. Recent studies have shown that low birth weight and preterm birth are linked to a higher prevalence of cardiovascular disease. The aim of the present study was to compare the levels of lipids and apolipoproteins and atherogenic indexes between term and near-term newborn infants. A sample of umbilical cord blood was obtained from 135 newborns (66 males) divided into two groups: 25 near-term neonates (35-36.6 weeks of gestational age) and 110 term neonates (37-42 weeks of gestational age). The total cholesterol concentrations were higher in the near-term neonates than in the term group (94.04 ± 8.02 vs 70.42 ± 1.63 mg/dl, P < 0.01), due to an increase in the LDL-cholesterol fraction in the...

Australian consumers' willingness to pay and willingness to purchase a hypothetical lower cholesterol pork product AgEcon
Bellhouse, Amy; Malcolm, Bill; Griffith, Garry R.; Dunshea, Frank.
This study investigated whether there would be an increase in consumer willingness to pay and purchase if reduced cholesterol pork was introduced to the Australian market. A stated choice analysis was used, with the following questions addressed. How are current purchases of fresh pork affected by concerns about cholesterol content? What financial premium, if any, would consumers place on reduced cholesterol pork? Would consumers buy more pork if a low cholesterol option were available? Is there a group of consumers, such as those with high cholesterol, who have an increased willingness to pay for or purchase reduced cholesterol pork when compared to consumers without this health problem? Results from this study indicated that at present the majority of...

Body measurements and serum lipid profile of overweight adult dogs fed diet with containing conjugated linoleic acid Ciência Rural
Rivera,Nancy Lorena Montaño; Félix,Ananda Portela; Ferreira,Fabiano Montiani; Silva,Ana Vitória Fisher da; Maiorka,Alex.
Studies with human beings and animals have shown that dietary conjugated linoleic acid (CLA) promotes changes in body structure, markedly, body fat reduction. This research aimed to assess the effect of CLA (60% of conjugated linoleic acid methyl ester, at the proportion of 1:1 of the 9, 11: 10, 12 isomers) on changes in body weight, subcutaneous tissue thickness, body fat mass and concentration of serum lipids (triacylglycerols, total cholesterol, LDL and HDL) in overweight dogs. Dogs of different breeds weighting 16.85±7.13 assigned in a randomized assay. Two types of diets were offered for 120 days: control (-CLA) and test (+CLA, 0.3%). There was no difference between control and test groups considering body weight and serum levels of triacylglycerols,...

Cholesterol changes the fatty acid composition of rat enterocytes BJMBR
Gazzola,J.; Martins,E.F.; Miyasaka,C.K.; Palanch,A.C.; Vecchia,M.G.; Curi,R..
The effect of free cholesterol on the fatty acid composition and growth of rat fetal enterocytes was investigated in the absence and presence of 10% (v/v) fetal calf serum. Cholesterol caused a significant reduction of cell number after 6 and 12 h in culture. The fatty acid composition of enterocytes cultured in the presence of serum was also changed by the presence of 20 µM cholesterol. The fatty acid profile was determined by HPLC using fluorescence detection (325 nm excitation and 395 nm emission). Cholesterol (20 µM) increased the proportion (given in percentage of the total fatty acids) of the following fatty acids in cultured cells: lauric (by 42%), oleic (by 34%), linoleic (by 44%) and gamma-linolenic (by 20%) acids and reduced the proportion of...

Cholesterol induces fetal rat enterocyte death in culture BJMBR
Gazzola,J.; Silva,É.P.P.; Kanunfre,C.C.; Verlengia,R.; Vecchia,M.G.; Curi,R..
The effect of cholesterol on fetal rat enterocytes and IEC-6 cells (line originated from normal rat small intestine) was examined. Both cells were cultured in the presence of 20 to 80 µM cholesterol for up to 72 h. Apoptosis was determined by flow cytometric analysis and fluorescence microscopy. The expression of HMG-CoA reductase and peroxisome proliferator-activated receptor gamma (PPARgamma) was measured by RT-PCR. The addition of 20 µM cholesterol reduced enterocyte proliferation as early as 6 h of culture. Reduction of enterocyte proliferation by 28 and 41% was observed after 24 h of culture in the presence and absence of 10% fetal calf serum, respectively, with the effect lasting up to 72 h. Treatment of IEC-6 cells with cholesterol for 24 h raised...

Cholesterol oxidation: Health hazard and the role of antioxidants in prevention Biol. Res.
VALENZUELA,ALFONSO; SANHUEZA,JULIO; NIETO,SUSANA.
Cholesterol is a molecule with a double bond in its structure and is therefore susceptible to oxidation leading to the formation of oxysterols. These oxidation products are found in many commonly-consumed foods and are formed during their manufacture and/or processing. Concern about oxysterols consumption arises from the potential cytotoxic, mutagenic, atherogenic, and possibly carcinogenic effects of some oxysterols. Eggs and egg-derived products are the main dietary sources of oxysterols. Thermally-processed milk and milk-derived products are another source of oxysterols in our diet. Foods fried in vegetable/animal oil, such as meats and French-fried potatoes, are major sources of oxysterols in the Western diet. Efforts to prevent or to reduce...

Coronary fat content evaluated by morphometry in patients with severe atherosclerosis has no relation with serum lipid levels BJMBR
Braz Jr.,D.J.; Gutierrez,P.S.; Luz,P.L. da.
The relationship between lipid serum levels and coronary atherosclerotic plaque fat content was studied in 51 necropsy patients. Serum lipids were measured by standard techniques, during life, in the absence of lipid-lowering drugs. Intima, intimal fat and media areas were measured using a computerized system in cryosections of the odd segments of the right, anterior descending and circumflex coronary arteries stained with Sudan-IV. Mean intimal and lipid areas were 5.74 ± 1.98 and 1.22 ± 0.55 mm² (22.12 ± 8.48%) in 26 cases with high cholesterol (³200 mg/dL) and 4.98 ± 1.94 and 1.16 ± 0.66 mm² (22.75 ± 9.06%) in 25 cases with normal cholesterol (<200 mg/dL; P > 0.05). Patients with high levels of low-density lipoprotein (³130 mg/dL, N = 15) had a...

Critical review: cholesterol and fat-soluble vitamins on shrimp feeding Sci. Agrar.Paran. / SAP
Lima, Rodrigo Barbosa; Figueiredo-Lima, Denise Fontana.
Four types of poly unsaturated fatty acids (PUFA) are essential for all shrimp species, linoleic (LOA, 18:2n-6), alfa-linolenic (ALA, 18:3n-3), eicosapentaenoic (EPA, 20:5n-3) and docosahexaenoic (DHA, 22:6n-3) acids. Moreover, the arachidonic acid (ARA, 20:4n-6), even when present, seems to be, in crustaceans, not so important or yet dispensable. It is relevant knowing that cholesterol as much as PUFA are not well absorbed when not followed by a substantial amount of lecithin, reaching 5% of the ration. The sum of PUFA reaches around 3% of the diet, even varying according to the species, stage and ration type. For Penaeus monodon a complex mathematic model was formulated in order to describe the relationships between the requirements of the cited PUFA....

In this work, we examined the impact of polymorphism in the cytochrome P450 (CYP) 3A5 gene, CYP3A5*1 (6986A > G, rs 776746), on the reduction in the lipid levels caused by simvastatin and atorvastatin. We studied 350 hyperlipidemic patients who received 10-40 mg of atorvastatin (n = 175) or simvastatin (n = 175) daily. Genotyping for CYP3A5 was done by PCR-RFLP analysis. Differences in the lipid profile before and after treatment were expressed as the % difference. The frequency of CYP3A5 polymorphism was 13.4% for heterozygotes and 86.6% for homozygotes. Comparison of the responses to same dose of each drug showed that the highest % difference was associated with total cholesterol (TC) in subjects receiving atorvastatin 40 mg compared with simvastatin...

Diagnosis of Smith-Lemli-Opitz syndrome by ultraviolet spectrophotometry BJMBR
Scalco,F.B.; Cruzes,V.M.; Vendramini,R.C.; Brunetti,I.L.; Moretti-Ferreira,D..
Smith-Lemli-Opitz syndrome (SLOS) is an autosomal recessive disorder due to an inborn error of cholesterol metabolism, characterized by congenital malformations, dysmorphism of multiple organs, mental retardation and delayed neuropsychomotor development resulting from cholesterol biosynthesis deficiency. A defect in 3ß-hydroxysteroid-delta7-reductase (delta7-sterol-reductase), responsible for the conversion of 7-dehydrocholesterol (7-DHC) to cholesterol, causes an increase in 7-DHC and frequently reduces plasma cholesterol levels. The clinical diagnosis of SLOS cannot always be conclusive because of the remarkable variability of clinical expression of the disorder. Thus, confirmation by the measurement of plasma 7-DHC levels is needed. In the present...

Dietary models for inducing hypercholesterolemia in rats BABT
Matos,Sheyla Leite; Paula,Heberth de; Pedrosa,Maria Lúcia; Santos,Rinaldo Cardoso dos; Oliveira,Eduardo Luiz de; Chianca Júnior,Deoclécio Alves; Silva,Marcelo Eustáquio.
The present work aimed at finding a dietetical model capable of promoting the highest hypercholesterolemia without affecting the development of the rats. Sixty female Fisher rats were divided into five groups. The first one was fed a control diet; the remaining four were fed hypercholesterolemic diets with cholesterol and different contents of soybean oil, starch, casein, micronutrients and fiber and, consequently, different caloric values. After eight weeks animals were evaluated in relation to growth, fecal excretion, liver weight and fat, cholesterol and its fractions, serum biochemical parameters and sistolic pressure and compared with controls. The best result was obtained with the diet containing 25 % soybean oil, 1.0 % cholesterol, 13 % fiber and...
