Abstract The Myrtaceae family has representatives spread worldwide, and Jabuticaba tree is an important species native to Brazil, which is grown not only in the backyards and domestic orchards, but also in commercial orchards, landscaping projects and in several tropical and subtropical countries. The propagation of this species in a sexual form or by seeds is simple and efficient, being able to reach 100%. However, long juvenile phase and genetic segregation have led to the search for methods that allow the propagation of selected trees and guarantee through their standardized and productive clones, fruit quality and fruiting precocity. Protocols for asexual propagation are still scarce, but they show efficient methods, making possible the commercial use... |

Abstract In Brazil, there is a growing interest in the crop of the olive tree, especially in those regions considered suitable for cultivation, where low temperatures occur between 8ºC and 10ºC in the period before flowering. The areas with these characteristics are located mainly in the South and Southeast regions of the country, characterized by a temperate climate in function of altitude, with the occurrence of natural vernalization. The propagation is an integral part of the production chain and the first step towards the implantation of the olive tree crop or renovation of the existing orchards. Obtaining quality seedlings, besides guaranteeing uniformity and reliable varietal origin, is a factor that influences the whole life of the orchard,... |

AbstractHyaluronidases are enzymes that mainly degrade hyaluronan, the major glycosaminoglycan of the interstitial matrix. They are involved in several pathological and physiological activities including fertilization, wound healing, embryogenesis, angiogenesis, diffusion of toxins and drugs, metastasis, pneumonia, sepsis, bacteremia, meningitis, inflammation and allergy, among others. Hyaluronidases are widely distributed in nature and the enzymes from mammalian spermatozoa, lysosomes and animal venoms belong to the subclass EC 3.2.1.35. To date, only five three-dimensional structures for arthropod venom hyaluronidases (Apis mellifera and Vespula vulgaris) were determined. Additionally, there are four molecular models for hyaluronidases fromMesobuthus... |

The present reflection refers to data obtained about the social representations of genomic research and its applications through interviews with legislators and lawyers, biomedical researchers and civilians and the review of scientific and legal literature in four Latin American countries: Argentine, Chile, Mexico and Peru. Several issues are addressed: little access to prevention and therapeutic methods, lack of equity in health benefits, commercialization of gene sequences through patents which leads to commercial exploitation of underdeveloped countries, the possibility of physical or psychological damage or genetic discrimination, the possibility of genetic modifications or abortion for eugenic reasons, the necessity of safeguarding confidentiality,... |

The objective of the present study was to report the molecular cloning and determination of the sequences N-methyl-D-aspartate subunit-1 receptor (NMDAR-1) in the hypothalamus of sheep. The re-amplified DNA template for NMDA -1 from the hypothalamus of male sheep was cloned using pBluescript-Sk-plasmids (pBSK, 2958 bp). Purified plasmids containing the NMDA receptor c-DNA were sequenced using the dye-terminator chemistry at the University of Nebraska-Lincoln, Nebraska, USA. Results were entered into the National Center for Biotechnology Web site as accession number AY434689. The sequence of Ovis aries (sheep) NMDAR-1 mRNA from hypothalamus of male sheep has 94-97% homology with Homo sapiens, 94-100% homology with Sus scrofa, and 88-90% homology with Rattus... |

Crude extracts of house dust mites are used clinically for diagnosis and immunotherapy of allergic diseases, including bronchial asthma, perennial rhinitis, and atopic dermatitis. However, crude extracts are complexes with non-allergenic antigens and lack effective concentrations of important allergens, resulting in several side effects. Dermatophagoides farinae (Hughes; Acari: Pyroglyphidae) is one of the predominant sources of dust mite allergens, which has more than 30 groups of allergen. The cDNA coding for the group 5 allergen of D. farinae from China was cloned, sequenced and expressed. According to alignment using the VECTOR NTI 9.0 software, there were eight mismatched nucleotides in five cDNA clones resulting in seven incompatible amino acid... |

Our objective was to clone, express and characterize adult Dermatophagoides farinae group 1 (Der f 1) allergens to further produce recombinant allergens for future clinical applications in order to eliminate side reactions from crude extracts of mites. Based on GenBank data, we designed primers and amplified the cDNA fragment coding for Der f 1 by nested-PCR. After purification and recovery, the cDNA fragment was cloned into the pMD19-T vector. The fragment was then sequenced, subcloned into the plasmid pET28a(+), expressed in Escherichia coli BL21 and identified by Western blotting. The cDNA coding for Der f 1 was cloned, sequenced and expressed successfully. Sequence analysis showed the presence of an open reading frame containing 966 bp that encodes a... |

For promoter studies the cloning, subcloning and transfer to different plasmid vectors usually requires use of restriction enzymes and ligation reactions. One obstacle is the nucleotide polymorphisms of eukaryotic genomic DNA, which has the consequence that a sequence often differs from published sequences. Therefore sequencing, rigorous restriction enzyme analysis or introduction of suitable sites has to be performed prior to cloning and subcloning. In addition, conventional methods using restriction enzymes, insert purifications and ligations is expensive and labour demanding. We have developed a fast, efficient and inexpensive Cre recombinase-loxP based method, which allows cloning of promoter regions and subcloning of these into a variety of vectors in... |

In Chile, most avocado orchards have been planted using 'Hass' as the scion variety on seedling rootstocks. This has led to the presence of a large variety of individuals, some of which show, among other characteristics, high productivity and a low degree of biennial bearing cycle in the same growing conditions. The most common method used in other countries to propagate such rootstocks from trees involves severe topping; however, this technique is notably destructive, making it difficult to employ in commercial orchards. With the goal of developing alternate methods for forcing shoot production from the rootstock, we tested two different procedures in July 2002 and March 2003, respectively: wounding in conjunction with the application of 1.1 M or 2.2 M... |
