Effect of pirimicarb application on field ppulations of Myzus persicae (Sulz.) (Homoptera: Aphididae) with different esterase levels Anais da SEB
Furiatti,Rui S.; M. N. Lázzari,Sonia.
The frequency and intensity of Myzus persicae (Sulz.) resistance were studied using bioassays and biochemical tests in two potato crops in Ibicoara, Bahia. Both areas received several insecticide applications which failed to control M. persicae. By the end of the crop season, one of the areas received two applications of pirimicarb and the other was not sprayed. Six samples of 700 aphids each were randomly collected before and after the pirimicarb applications, with an interval of four days. Specimens of M. persicae were also collected from isolated potato plants. At the laboratory, the samples were characterized by the CL50 based on insecticide bioassays and by total esterase activity using colorimetric assays. After the pirimicarb applications, the...

Soybean seed galactinol synthase activity as determined by a novel colorimetric assay Rev. Bras. Fisiol. Veg.
RIBEIRO,MARLUCI; FELIX,CARLOS R.; LOZZI,SILENE DE PAULINO.
Galactinol synthase (GS) is a key enzyme for the biosynthesis of raffinose oligosaccharides (RO) which are the flatulence factors present in soybean seeds and several other legumes. Understanding of soybean seed GS properties is, therefore, of biotechnological interest. The GS enzyme catalyses formation of galactinol and UDP from UDP-gal and myo-inositol. This enzyme is currently assayed by an isotopic method. We have then idealized a more convenient method for GS assay based on the indirect colorimetric determination of the UDP formed which is then hydrolyzed by exogenous apyrase and the resulting Pi quantified by a modification of the colorimetric method of Fiske & SubbaRow. The color developed is stable, and the method is suitable for detection...

Dourine is caused by Trypanosoma equiperdum via coitus with an infected horse. Although dourine is distributed in Equidae worldwide and is listed as an internationally important animal disease by the World Organization for Animal Health (OIE), no effective treatment strategies have been established. In addition, there are no reports on drug discovery, because no drug screening system exists for this parasite. A new T.?equiperdum strain was recently isolated from the genital organ of a stallion that showed typical symptoms of dourine. In the present study, we adapted T.?equiperdum IVM-t1 from soft agarose media to HMI-9 liquid media to develop a drug screening assay for T.?equiperdum. An intracellular ATP-based luciferase assay using CellTiter-Glo reagent...
