Cloning and chloroplast-targeted expression studies of insect-resistant gene with ricin fusion-gene under chloroplast transit peptide in cotton Electron. J. Biotechnol.
Kiani,Sarfraz; Ali,Arfan; Bajwa,Kamran Shehzad; Muzaffar,Adnan; Ashraf,Muhammad Aleem; Samiullah,Tahir Rehman; Shahid,Ahmad Ali; Husnain,Tayyab.
Background: Transgenic plants inhabiting single Bt gene are prone to develop insect resistance and this resistance has been reported in case of some important yield-devastating insect larvae of commercial crops, such as cotton and rice. Therefore, it has become essential to adapt new strategies to overcome the problem of insect resistance and these new strategies should be sophisticated enough to target such resistant larvae in broad spectrum. Among these, plants may be transformed with Bt gene tagged with some fusion-protein gene that possesses lectin-binding capability to boost the binding sites for crystal protein gene within insect mid-gut in order to overcome any chances of insect tolerance against Bt toxin. Enhanced chloroplast-targeted Bt gene...

Collagen arrangement in hepatic granuloma in mice infected with Schistosoma mansoni: dependence on fiber radiation centers BJMBR
Lenzi,H.L.; Kimmel,E.; Schechtman,H.; Pelajo-Machado,M.; Vale,B.S.; Panasco,M.S.; Lenzi,J.A..
The collagen structure of isolated and in situ liver granuloma from Swiss Webster mice infected with Schistosoma mansoni was sequentially and three-dimensionally analyzed during different times of infection (early acute, acute, transitional acute-chronic, and chronic phases) by laser scanning confocal microscopy and electron scanning variable vacuum microscopy. The initial granuloma structure is characterized by vascular collagen residues and by anchorage points (or fiber radiation centers), from where collagenous fibers are angularly shed and self-assembled. During the exudative-productive stage, the self-assembly of these fibers minimizes energy and mass through continuous tension and focal compression. The curvature or angles between collagen fibers...

Early colonization pattern of maize (Zea mays L. Poales, Poaceae) roots by Herbaspirillum seropedicae (Burkholderiales, Oxalobacteraceae) Genet. Mol. Biol.
Monteiro,Rose A.; Schmidt,Maria A.; Baura,Valter A. de; Balsanelli,Eduardo; Wassem,Roseli; Yates,Marshall G.; Randi,Marco A.F.; Pedrosa,Fábio O.; Souza,Emanuel M. de.
The bacterium Herbaspirillum seropedicae is an endophytic diazotroph found in several plants, including economically important poaceous species. However, the mechanisms involved in the interaction between H. seropedicae and these plants are not completely characterized. We investigated the attachment of Herbaspirillum to maize roots and the invasion of the roots by this bacterium using H. seropedicae strain SMR1 transformed with the suicide plasmid pUTKandsRed, which carries a mini-Tn5 transposon containing the gene for the Discosoma red fluorescent protein (Dsred) constitutively expressed together with the kanamycin resistance gene. Integration of the mini-Tn5 into the bacterial chromosome yielded the mutant H. seropedicae strain RAM4 which was capable of...

Embryo sac development in yellow passion fruit Passiflora edulis f. flavicarpa (Passifloraceae) Genet. Mol. Biol.
Souza,Margarete Magalhães de; Pereira,Telma Nair Santana; Hoffmann,Magali; Melo,Edésio J.T. de; Louro,Ricardo Pereira.
The yellow passion fruit, Passiflora edulis f. flavicarpa, is one of the most important Brazilian fruit crops. It is an allogamous, diploid, and self-incompatible species. It has hermaphrodite, solitary flowers, located in the leaf axils and protected by leaf bracts. The flower has an androgynophore, which is a straight stalk supporting its reproductive parts. There are usually five anthers, located at the tip of each of the five filaments. The ovary is borne just above the filaments, at the top of the androgynophore; there are three styles that are united at their base, and at the top there are three stigmas. The objective of this research was to observe embryo sac development in yellow passion flowers. Ovaries at different stages of development were...

Evidence for the endophytic colonization of Phaseolus vulgaris(common bean) roots by the diazotroph Herbaspirillum seropedicae BJMBR
Schmidt,M.A.; Souza,E.M.; Baura,V.; Wassem,R.; Yates,M.G.; Pedrosa,F.O.; Monteiro,R.A..
Herbaspirillum seropedicae is an endophytic diazotrophic bacterium, which associates with important agricultural plants. In the present study, we have investigated the attachment to and internal colonization of Phaseolus vulgaris roots by the H. seropedicae wild-type strain SMR1 and by a strain of H. seropedicae expressing a red fluorescent protein (DsRed) to track the bacterium in the plant tissues. Two-day-old P. vulgaris roots were incubated at 30°C for 15 min with 6 x 10(8) CFU/mL H. seropedicae SMR1 or RAM4. Three days after inoculation, 4 x 10(4) cells of endophytic H. seropedicae SMR1 were recovered per gram of fresh root, and 9 days after inoculation the number of endophytes increased to 4 x 10(6) CFU/g. The identity of the recovered bacteria was...

Evidence of accelerated and altered pollen development after Imazapyr treatment in resistant sunflower Bragantia
Ochogavía,Ana Claudia; Bianchi,Marta Beatríz; Picardi,Liliana; Nestares,Graciela María.
ABSTRACT Imazapyr (IM) treatment applied at early reproductive developmental stages have been associated with induced male sterility in sunflower (Helianthus annuus L.). The aim of this work was to determine the effects induced by two different doses of IM-treatment (1X and 2X; 80 and 160 g a.i.·ha-1) on pollen grain yield, pollen viability and on the exine surface, in an intermediate resistant (I) and a completely resistant (R) genotype. An anatomical analysis of some stages during the microsporogenesis and microgametogenesis was also achieved by confocal microscopy. Plants of both genotypes survived the herbicide treatments and showed no external injury symptoms. Differential response to different doses of IM was obtained in R and I genotypes. The pollen...

Fluorescent ligands for studying neuropeptide receptors by confocal microscopy BJMBR
Beaudet,A.; Nouel,D.; Stroh,T.; Vandenbulcke,F.; Dal-Farra,C.; Vincent,J.-P..
This paper reviews the use of confocal microscopy as it pertains to the identification of G-protein coupled receptors and the study of their dynamic properties in cell cultures and in mammalian brain following their tagging with specific fluorescent ligands. Principles that should guide the choice of suitable ligands and fluorophores are discussed. Examples are provided from the work carried out in the authors' laboratory using custom synthetized fluoresceinylated or BODIPY-tagged bioactive peptides. The results show that confocal microscopic detection of specifically bound fluorescent ligands permits high resolution appraisal of neuropeptide receptor distribution both in cell culture and in brain sections. Within the framework of time course experiments,...

Immunohistochemistry of GluR1 subunits of AMPA receptors of rat cerebellar nerve cells Biocell
Castejón,Orlando J.; Dailey,Michael E..
The localization of GluR1 subunits of ionotropic glutamate receptors in the glial cells and inhibitory neurons of cerebellar cortex and their association with the climbing and parallel fibers, and basket cell axons were studied. Samples of P14 and P21 rat cerebellar cortex were exposed to a specific antibody against GluR1 subunit(s) of AMPA receptors and were examined with confocal laser scanning microscopy. GluR1 strong immunoreactivity was confined to Purkinje cell and the molecular layer. Weak GluR1 immunoreactivity was observed surrounding some Golgi cells in the granule cell layer. Intense GluR1 immunoreactivity was localized around Purkinje, basket, and stellate cells. Purkinje cells expressed strong GluR1 immunoreactivity surrounding the cell body,...

NEW PERSPECTIVES ON THE DETERMINATION OF PHOSPHATASE ACTIVITY IN ECTOMYCORRHIZAE OF NOTHOFAGUS OBLIQUA FORESTS IN SOUTHERN CHILE Gayana Botánica
Alvarez,Maricel; Härtel,Steffen; Godoy,Roberto; Heyser,Wolfgang.
We present a method to localise and quantify surface-bound phosphatase activity (SBPA) in mycorrhizal fungi and mycorrhizae of Nothofagus obliqua (Mirb.) Oerst., using image processed confocal fluorescent microscopy. ELF-97 is a hydrophilic substrate which turns into a strongly fluorescent precipitate upon activation by phosphomonoesterases. In fungal mycelium, this technique has recently been approved by comparison with a standard method (p-nitrophenyl-phosphate). The microscopic technique based on ELF-97 revealed that Paxillus involutus (Batsch: Fr.) Sing. and Austropaxillus boletinoides (Sing.) Brsky. & Jarosch provide different adaptive strategies to changing phosphate concentrations and different pH (3-7). We also analysed SBPA in four...
