ABSTRACT The efficacy of an antisepsis protocol comprising chlorhexidine gluconate and ethyl alcohol in combination with prophylactic antimicrobial therapy in controlling surgical site infection in horses was studied. To that purpose, seven mixed breed horses received potassium penicillin and gentamicin at least 30 minutes prior to surgery. The surgical site was scrubbed with chlorhexidine gluconate and rinsed with ethyl alcohol. Samples were collected at four time points: (A) - before and (B) - immediately following shaving of the hair coat, (C) - at the end of antisepsis procedures, and (D) - at the end of the surgical procedure. Assessment of the metabolism of different strains of Bacillus megaterium BABT

Bacillus megaterium is a promising host for expression of heterologous proteins. This paper reports the nutrient consumption patterns and production of metabolites for three different strains of B. megaterium, ATCC 14945, QMB 1551 and PV 361, which is QMB 1551 with seven constitutive plasmids deleted. 14 h cultivations in agitated flasks were run, for two different media: A (LB plus 10g/L glucose) and B (medium A, with the yeast extract replaced by tryptone). Strains PV361 and QMB 1551 showed higher maximum specific growth rates in medium B, reaching 0.42 h-1 and 0.48 h-1 respectively. Asymbiotic germination in three Chloraea species (Orchidaceae) from Chile Gayana Botánica

ABSTRACT Orchids require symbiotic fungi and/or specific conditions to germinate. Asymbiotic techniques have been shown successful for orchid germination. In Chile, Chloraea include many endemic, and potentially ornamental, terrestrial orchid species. In this study, individuals of Chloraea crispa, C. gavilu and C. virescens were manually autopollinated. The resulting capsules were sterilized and seeds were aseptically obtained. We evaluated asymbiotic germination in: Agar Water (AW), Knudson C (KC), Banana Culture Media (CMB), Tomato Culture Media (CMT), Malmgren Modified (MM), Murashige and Skoog (MS), and MS modified (MS1/2). The objective of this study was to establish cell suspension culture and plant regeneration via somatic embryogenesis of a Brazilian plantain, cultivar Terra Maranhão, AAB. Immature male flowers were used as explant source for generating highly embryogenic cultures 45 days after inoculation, which were used for establishment of cell suspension culture and multiplication of secondary somatic embryos. Five semisolid culture media were tested for differentiation, maturation, somatic embryos germination and for plant regeneration. Morchella esculenta es un hongo comestible de alto valor comercial y la obtención de esclerocios es considerada como la clave para su cultivo. En este trabajo se evaluó la cepa IE-750 en ocho medios de cultivo sólido y seis en forma líquida, utilizando como parámetros el crecimiento micelial, la producción de biomasa, la habilidad para producir esclerocios y su biomasa. El mejor crecimiento micelial se obtuvo en el tratamiento con composta (T7: 53.87 cm2), y en cuanto a la biomasa, el medio de cultivo con levadura fue mejor (T3: 80.3 mg). Effects of environmental conditions on the sessile existence of an estuarine sediment bacterium. ArchiMer

A submerged glass coverslip technique was developed to determine dissolved organic nutrient uptake by adherent cells of a sediment isolate of Enterobacter cloacae . Cells which colonized the coverslips in swirling culture flasks during anaerobic growth remained firmly attached during manipulations employed to determine uptake of radiolabeled glucose by the adherent population. The attached cells were capable of a more rapid rate of glucose uptake than free cells. Lineweaver-Burk plots demonstrated different glucose uptake kinetics for the 2 cell populations. The data suggest that physiological changes occur in cells soon after they become attached to surfaces. These changes appear to enhance the metabolic activity of the adherent population.

ABSTRACT The present study evaluated the effects of the culturing media and the levels of nitrogen and phosphorus on the growth, biomass productivity and lipid production of four species of Microcystis (M. novacekii, M. aeruginosa, M panniformis and M. protocystis). The lipid extract was obtained by refluxing with dichloromethane (Soxhlet). The biomass and biomass productivity yields were maximized with ASM-1 medium treatment enriched with nitrogen and/or phosphorus (0.25-0.65 g/L and 25-50.7 mg/L d-1, respectively). The lipid extract yields from M. panniformis and M. novacekii were inversely correlated with the concentration of nitrogen and directly correlated with the concentration of phosphorus (35.8 % and 31.7 %). EMJH medium with 5-fluorouracil and nalidixic acid associated with serial dilution technique used to recover Leptospira spp from experimentally contaminated bovine semen BJM

Bovine semen experimentally contaminated with Leptospira santarosai serovar Guaricura was submitted to the modified EMJH medium with 5-fluorouracil (300mg/L) and nalidixic acid (20mg/L), named as "selective medium" and using the serial dilution technique, in order to evaluate the percentage of recovery of the added microorganism. The selective EMJH medium was found with higher percentage of recovery of leptospiras and minor losses of samples due to contamination with opportunistic microorganisms than the non-selective EMJH medium: 151/376 (40.0%) of positive growth; and 38/376 (10.0%) contamination and 58/376 (15%) and 129/376 (34.0%), respectively. Anthers of two soybean cultivars were cultured in B5 long basal culture media gelled with agarose or PhytagelTM. Cytological examinations of the anthers were carried out during the first 45 days of culture to assay the viability and developmental stage of microspores. Frequency of callus formation was recorded at 45 days of culture. The analysis of variance of the microspore viability assay showed significant Cultivar X Gelling Agent X Day of Culture interactions. Growth capacity of thermotolerant campylobacters in culture media supplemented with pig and cow blood BABT

In this work 60 thermotolerant Campylobacter strains (37 C. jejuni and 23 C. coli) isolated from the cows, pigs, chickens and ducks (15 strains of each type of animal) were used to establish their growth capacity on media containing cow or swine blood as potential substitutes of sheep or horse blood. The growth capacity was assessed by viable counts on cow and swine blood media, using the modified Miles and Misra method. Campylobacter strains showed better growth in the media supplemented with pig or sheep blood than with cow blood. Thus, the use of pig blood could be a supplement for Campylobacter culture medium, when there was no availability of sheep or horse blood.
