Addition of benzyladenine to coconut explant cultured In vitro improves the formation of somatic embryos and their germination Agrociencia
Montero-Cortés,Mayra I.; Chan-Rodríguez,José L.; Cordova-Lara,Ivan; Oropeza-Salin,Carlos; Sáenz-Carbonell,Luis.
Coconut (Cocos nucífera L.) production has been declining in México and other countries due to deadly diseases; therefore, massive production of palms resistant to diseases is needed for replanting. Production of somatic embryos and micropropagation could be a very useful technique to produce free disease coconut population. Explants (embryogenic-structures) of Pacific Tall 2 ecotype were cultured in an induction medium defined as I (semisolid Y3 medium containing 0.65 mM 2,4-dichlorophenoxyacetic acid) and treated with the cytokinin benzyladenine (BA) at 0, 25, 100 or 200 ,µM to evaluate its effect on embryogenic callus formation. The BA treatments did not affect the percentage of embryogenic callus formation. In order to evaluate the effect of the BA...

Different alternatives for producing sprouts of avocado (Persea americana) rootstocks Ciencia e Investigación Agraria
Castro,Mónica; Fassio,Claudia; Darrouy,Nicole; Aedo,Marcela; Jorquera,Lorena.
In Chile, most avocado orchards have been planted using 'Hass' as the scion variety on seedling rootstocks. This has led to the presence of a large variety of individuals, some of which show, among other characteristics, high productivity and a low degree of biennial bearing cycle in the same growing conditions. The most common method used in other countries to propagate such rootstocks from trees involves severe topping; however, this technique is notably destructive, making it difficult to employ in commercial orchards. With the goal of developing alternate methods for forcing shoot production from the rootstock, we tested two different procedures in July 2002 and March 2003, respectively: wounding in conjunction with the application of 1.1 M or 2.2 M...

Effect of preharvest spraying with thidiazuron on fruit quality and maturity of apples Rev. Bras. Frutic.
Amarante,Cassandro Vidal Talamini do; Megguer,Clarice Aparecida; Blum,Luiz Eduardo Bassay.
Apple trees, cultivars Gala and Fuji, were sprayed at full bloom with thidiazuron (TDZ) at the doses of 0, 5, 10, or 20 g (a.i.) ha-1 and fruit were assessed for quality and maturity. In both cultivars, the increase of TDZ dose had detrimental effects on fruit quality, causing a reduction of fruit red surface and an increase of percentage of fruit that was asymmetrical and with calyx-end rot. TDZ caused an increment of calyx-end aperture that might have increased calyx-end rot. TDZ increased density and the length : diameter (L : D) ratio of the fruit. The number of viable seeds was not affected by TDZ. TDZ resulted in a substantial delay of fruit maturity in 'Gala' but not in 'Fuji'. TDZ also reduced soluble solids content (SSC) in 'Gala' and reduced...

FAGUNDES,EVERLAN; PETRI,JOSÉ LUIZ; ARGENTA,LUIZ CARLOS; HAWERROTH,FERNANDO JOSÉ; COUTO,MARCELO.
ABSTRACT ‘Gala’ apple trees have low fruit set in restrictive pollination situations, being an obstacle to the achievement of high production rates in orchards in southern Brazil, which can be minimized by the use of growth regulators. The aim of this study was to evaluate the effect of thidiazuron concentrations in ‘Royal Gala’ apple fruiting and production, grown in mild winter conditions. The experiment was conducted during the 2011/2012, 2012/2013 and 2013/2014 crop years in Fraiburgo, SC, on ‘Royal Gala’ apple trees in Marubakaido rootstock M9 with filter. In all evaluation cycles, the experimental design was a randomized block with factorial arrangement (6x2) with six TDZ concentrations and two application forms, with six replicates, and the...

Endogenous auxin and cytokinin contents associated with shoot formation in leaves of pineapple cultured in vitro Braz. J. Plant Physiol.
Mercier,Helenice; Souza,Beatriz Maia; Kraus,Jane Elizabeth; Hamasaki,Regina Mayumi; Sotta,Bruno.
The in vitro culture of pineapple leaves on a shoot induction medium (SIM) results in the formation of protuberances and further development in shoots, and plantlets. The contents of endogenous indoleacetic acid (IAA) and five cytokinins (Cks), N6(2-isopentenyl)adenine (iP), N6(2-isopentenyl)adenosine (iPR), zeatin (Z), zeatin riboside (ZR) and N6-benzyladenine (BA), present in the basal portion of those leaves, were correlated to the organogenic response that occurs over 15 days of culture. The endogenous auxin/cytokinins ratio was lowest on the 3rd day, mainly due to a strong increase in the iP level. It seems that endogenous iP concentration triggered the induction signal for an organogenic response in pineapple leaf bases. The rise in iP content...

Gas exchange in Annona emarginata (Schltdl.) H. Rainer subjected to salt stress and application of plant growth regulator Sci. Agrar.Paran. / SAP
Dalanhol, Samanta Jaqueline; Mantoan, Luís Paulo Benetti; Amaro, Amanda Cristina Esteves; Ferreira, Gisela.
The aim of this study was to evaluate the effects of the application of plant growth regulator on gas exchange in A. emarginata seedlings subjected to different NaCl concentrations. We evaluated the gas exchange in A. emarginata seedlings subject to the presence and absence of plant growth regulator consisting of kinetin, gibberellic acid, and indolbutyric acid and three concentrations of NaCl (0 mM, 50 mM, and 100 mM). When assessing these factors individually, it was observed that in the presence of plant growth regulators, there was a reduction in stomatal conductance and transpiration, and NaCl concentration (50 mM and 100 mM) caused a reduction in CO2 assimilation, stomatal conductance, and transpiration. There was no significant interaction between...

Braga, Karina de Queiroz; Coimbra, Mairon César; Castro, Ana Hortência Fonsêca.
A protocol for the in vitro germination and callus induction in Pyrostegia venusta, a medicinal plant species from the Brazilian savannah (Cerrado) is provided. The establishment of cultures of P. venusta was originally obtained from seeds germinated in vitro and induced callus directly from leaf explants of seedlings. Seeds were germinated on MS and WPM media containing 100 and 50% of salt concentration, supplemented with 30 g L-1 sucrose. Callus induction consisted of the inoculation of leaf segments on MS medium plus 2,4-D or BAP in the presence or absence of light. The germination percentage averaged 85% and the aerial parts and roots of seedlings obtained in WPM with 50 and 100% of salt concentration showed elevated contents of total phenolic...

In vitro multiplication of 'Flordaguard' rootstock: cytokinin source and concentration effects, explants orientation and period of permanence in the culture medium BABT
Radmann,Elizete Beatriz; Bianchi,Valmor João; Fachinello,José Carlos; Ferreira,Letícia Vanni; Oliveira,Roberto Pedroso de.
This work aimed at studying the effects of different sources and cytokinin concentrations, as well as explants orientation and time of their permanence in shoots induction medium to obtain high multiplication rate. Four experiments were carried out to evaluate the sprout percentage mean, shoots number per explant, elongated shoots percentage and shoots length. The treatments with 2iP and zeatin did not promote in vitro plant multiplication. When different BAP concentrations were tested, an increase in sprout percentage was obtained using BAP concentration up to 3.66 mg L-1 (80%), and for shoots number per explant a linear behavior was observed, showing a mean of four shoots per explant with 4.0 mg L-1 BAP. For the 'Flordaguard' rootstocks, the explants...

In vitro organogenesis and genetic transformation of mandarin cultivars Rev. Bras. Frutic.
Soriano,Leonardo; Tavano,Eveline Carla da Rocha; Correa,Marcelo Favaretto; Harakava,Ricardo; Mendes,Beatriz Madalena Januzzi; Mourão Filho,Francisco de Assis Alves.
Abstract The in vitro organogenesis of Fremont (Citrus clementina x ), Citrus reticulataThomas (Citrus reticulata), and Nules (Citrus clementina) mandarins was evaluated aiming to optimize a regeneration protocol that could be applied in genetic transformation. The use of epicotyl-derived explants resulted in higher explant responsiveness and number of shoots developed per explant when compared with the use of internodal-derived explants. The highest efficiency in shoot regeneration was observed in the presence of 1 mg L-1 of BAP, regardless of the explant type and cultivar. The in vitro organogenesis protocol produced transgenic plants from three mandarin cultivars expressing attA gene under the control of phloem-specific promoters.

Influence of 2,4-D and BAP on callus growth and the subsequent regeneration of somatic embryos in long-term cultures of Pelargonium x domesticum cv. Madame Layal Electron. J. Biotechnol.
Haensch,Klaus-Thomas.
This long-term study demonstrates for the first time that it is possible to propagate embryogenic cultures in pelargoniums and to subsequently initiate the differentiation of embryos using the cultivar Madame Layal (Pelargonium x domesticum). Propagation of callus was only possible with combinations of 2,4-dichlorophenoxyacetic acid (2,4-D) and 6-benzylaminopurine (BAP), which gave rise to embryos from the primary culture stage on. However, the propagation of cells, as well as the differentiation of embryos, was inhibited by a continuous application of these growth regulators. For this reason, a long period on medium lacking growth regulators was necessary before the differentiation of embryos occurred again. The consequences for improving the propagation...

Influence of Benzyladenine and Thidiazuron on Shoot Regeneration from Leaf and Shoot tip Explants of Sedum sarmentosum Bunge BABT
Kim,Doo Hwan; Sivanessan,Iyyakkannu.
Effects of N6-benzyladenine (BA) or thidiazuron (TDZ) on adventitious shoot regeneration and axillary shoot multiplication of Sedum sarmentosum was investigated. Leaf and shoot tip explants obtained from in vitro-grown shoots of S. sarmentosum were cultured on Murashige and Skoog (MS) medium supplemented with 0, 2.0, 4.0 or 8.0 µM BA or TDZ. Of the two cytokinins studied, BA was found to be more responsive as compared to TDZ with respect to number of shoots produced per explant. High frequency of shoot regeneration (92.2%) with a mean of 12.3 shoots was produced when the leaf explants were cultured on MS medium supplemented with 8.0 µM BA. The highest number of shoots (25.4) was obtained when shoot tip explants were cultured on MS medium devoid of...

Micropropagation of Homalomena pineodora Sulaiman & Boyce (Araceae): a new species from Malaysia Horticultura Brasileira
Stanly,Christine; Bhatt,Arvind; Sulaiman,Baharuddin; Keng,Chan Lai.
Homalomena pineodora (family Araceae) is a species found to have impressive foliage characteristics which remain evergreen throughout the year. Therefore, H. pineodora can be grown as an ornamental plant. Generally H. pineodora needs 3-5 years to propagate and multiply. However, the demand for new ornamental plants is increasing worldwide and the quality of planting material is a basic need for boosting productivity. Therefore an efficient micropropagation protocol for large-scale production of H. pineodora was developed. In vitro shoot cultures were initiated from the rhizomatous buds on MS basal medium. The best conditions for propagating H. pineodora was found to be MS medium supplemented with 3% sucrose and 0.5 mg L-1 BA (6-benzyladenine) under 24 h of...

Oxidative metabolism altered by plant growth regulators in atemoia seeds (Annona x atemoya Mabb.) ‘Thompson’ Rev. Bras. Frutic.
Hata,Ana Paula Yuki; Ferreira,Gisela; Sousa,Marília Caixeta; Bezerra,Breno Kennedy Lima; Kissmann,Camila.
Abstract The objective of this work was to validate the enzymatic activity in Annona x atemoyaseeds submitted to treatments with plant growth regulators belonging to the group of gibberellins and cytokinins for overcoming dormancy during the germination process. Initially, the water acquisition curve was determined, where two points of phase II (72 and 144 hours) were determined to evaluate the activity of superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) enzymes in atemoya seeds after the application of two plant growth regulators. Treatments with GA resulted in higher activity of SOD enzyme, which may be favored the higher germination percentage and speed in comparison to control treatment. In cases in which SOD enzyme presented high...

Reproductive physiology of mango Braz. J. Plant Physiol.
Davenport,Thomas L..
Mango flowering involves hormonal regulation of shoot initiation and induction events resulting in reproductive shoot formation. A balance or ratio of endogenously regulated phytohormones, thought to be auxin from leaves and cytokinins from roots, appears to govern the initiation cycle independently from inductive influences. Induction of reproductive or vegetative shoots is thought to be governed by the ratio of a temperature-regulated florigenic promoter and an age regulated vegetative promoter at the time of shoot initiation. Management of off-season flowering in mango trees is being accomplished in the tropics by successfully synchronizing shoot initiation through tip pruning and use of nitrate sprays coupled with management of the stem age to induce...

Thidiazuron affects the quality of 'Gala' apples stored under controlled atmosphere Ciência Rural
Giehl,Ricardo Fabiano Hettwer; Sestari,Ivan; Eisermann,Ana Cristina; Brackmann,Auri.
This work was carried out aiming to evaluate the effect of thidiazuron (TDZ) on the quality of 'Gala' apples stored under controlled atmosphere (CA) conditions. Plants were sprayed at full bloom with TDZ at 0, 10, 20, or 40g ha-1 (a.i.). Harvested fruits were then stored in CA with 1.0+2.5, 1.5+2.5 and 1.5+5.0 (kPa O2+kPa CO2), both at 0.5°C. In addition, the partial pressure of 1.5kPa of O2+2.5kPa of CO2 was also evaluated at -0.5°C. Higher flesh firmness and titratable acidity was obtained by applying 20 or 40g ha-1 TDZ. Furthermore, these TDZ doses decreased the respiration rate as well as the occurrence of flesh breakdown, mealiness and decay. The application of TDZ at doses ranging from 20 to 40g ha-1 maintains higher fruit quality for up to 8 months...

Thidiazuron effects on shoot growth, return bloom, fruit set and nutrition of apples PAB
Amarante,Cassandro Vidal Talamini do; Ernani,Paulo Roberto; Blum,Luiz Eduardo Bassay; Megguer,Clarice Aparecida.
Apple (Malus domestica Borkh.) trees, cultivars Gala and Fuji, were sprayed at full bloom with thidiazuron (TDZ) at doses of 0, 5, 10, or 20 g ha-1 of a.i. in order to evaluate its effects on plant growth and development, return bloom, fruit set, nutrition, and fruit yield. Fruit set increased with TDZ dose in 'Gala' but not in 'Fuji'. TDZ did not affect fruit yield in any cultivar. In 'Gala', the return bloom was reduced in about 50% at TDZ doses of 10 or 20 g ha-1. TDZ increased shoot growth in both cultivars. In leaves, TDZ decreased the concentration of Ca and Mg in 'Gala' and of Mg in 'Fuji', but did not affect the chlorophyll content, leaf area, length, width, and dry mass per cm² regardless of cultivar. In fruits, the effect of TDZ varied according...
