Characterization of a new whitefly-transmitted (Bemisia tabaci Meam 1) cytorhabdovirus infecting common bean. Repositório Alice

Common bean (Phaseolus vulgaris) is an important crop in Latin America and Africa. The bean yield may be affected by the incidence of different viruses. As part of a virus survey in beans, we have used high-throughput sequencing approach to identify viruses with RNA genome in bean plants collected in Goiás (GO) state. Bioinformatic analysis of the de novoassembled contigs identified a putative cytorhabdovirus (family Rhabdoviridae) with low similarities with Northern cereal mosaic virus (NCMV). This new cytorhabdovirus was denominated Bean associated cytorhabdovirus (BaC). Cytorhabdoviruses have enveloped particles, negative ssRNA of 11-14 kb genome and are usually transmitted by aphids or leafhoppers.

Using double-strand RNA (dsRNA) high-throughput sequencing, we identified five RNA viruses in a bean golden mosaic virus (BGMV)-resistant common bean transgenic line with symptoms of viral infection. Four of the identified viruses had already been described as infecting common bean (cowpea mild mottle virus, bean rugose mosaic virus, Phaseolus vulgaris alphaendornavirus 1, and Phaseolus vulgaris alphaendornavirus 2) and one is a putative new plant rhabdovirus (genus Cytorhabdovirus), tentatively named bean-associated cytorhabdovirus (BaCV). Occurrence of a closely-related isolate to Maize yellow striate virus in wheat plants Agriscientia (Córdoba)

In Argentina, wheat fields have exhibited virus-like symptoms, such as chlorotic streaking, dwarfing, yellowing and empty ears since 2007. Symptomatic plants and leaves samples were collected in Marcos Juárez (2007) and Río Cuarto (2008 and 2013), both of them in Córdoba province. The virus was experimentally transmitted from symptomatic wheat plants to wheat cv. Baguette 10 and cv. BioINTA 3005 using the vector Delphacodes kuscheli Fennah (Delphacidae); symptoms of chlorotic streaking, dwarfing and yellowing appeared in the inoculated cereals at 10-15 days post-inoculation. Virus presence was confrmed by electron microscopy and RT-PCR using degenerated primers, which amplified a conserved region of the plant rhabdovirus polymerase (L) gene. Sequence...
