Chromosome numbers, meiotic abnormalities, and 2n pollen formation in accessions of the wild species Chrysolaena flexuosa (Vernonieae, Compositae) from its distribution range in Argentina Boletín de la Sociedad
Echeverría,María Lis; Camadro,Elsa Lucila.
Chrysolaena flexuosa is a South American species of potential ornamental value (basic chromosome number of x=10). Diploid (n=10) and tetraploid (n=20) cytotypes have been reported for its distribution area, although one hexaploid (n=30-32ca.) cytotype has been reported for its most southern distribution. To investigate if ploidy and latitude are positively related in Ch. flexuosa natural populations and if sexual polyploidization could have had a role in the origin of the polyploid cytotypes, we determined chromosome numbers, DNA content, and pollen viability and size, and analyzed microsporogenesis in samples of seven Argentinian accessions. Two of the northeastern accessions were diploid and one was tetraploid, whereas the four southeastern accessions...

Coconut genome size determined by flow cytometry: tall versus dwarf types. Repositório Alice
FREITAS NETO, M.; PEREIRA, T. N. S.; GERONIMO, I. G. C.; AZEVEDO, A. O. N.; RAMOS, S. R. R.; PEREIRA, M. G..
Coconuts (Cocos nucifera L.) are tropical palm trees that are classified into Tall and Dwarf types based on height, and both types are diploid (2n = 2x = 32 chromosomes). The reproduction mode is autogamous for Dwarf types and allogamous for Tall types. One hypothesis for the origin of the Dwarf coconut suggests that it is a Tall variant that resulted from either mutation or inbreeding, and differences in genome size between the two types would support this hypothesis. In this study, we estimated the genome sizes of 14 coconut accessions (eight Tall and six Dwarf types) using flow cytometry. Nuclei were extracted from leaf discs and stained with propidium iodide, and Pisum sativum (2C = 9.07 pg DNA) was used as an internal standard. Histograms with good...

Cytological and genome size data analyzed in a phylogenetic frame: Evolutionary implications concerning Sisyrinchium taxa (Iridaceae: Iridoideae) Genet. Mol. Biol.
Burchardt,Paula; Souza-Chies,Tatiana T.; Chauveau,Olivier; Callegari-Jacques,Sidia M.; Brisolara-Corrêa,Lauís; Inácio,Camila Dellanhese; Eggers,Lilian; Siljak-Yakovlev,Sonja; de Campos,José Marcello Salabert; Kaltchuk-Santos,Eliane.
Abstract Sisyrinchium is the largest genus of Iridaceae in the Americas and has the greatest amount of cytological data available. This study aimed at investigating how genomes evolved in this genus. Chromosome number, genome size and altitude from species of sect. Viperella were analyzed in a phylogenetic context. Meiotic and pollen analyses were performed to assess reproductive success of natural populations, especially from those polyploid taxa. Character optimizations revealed that the common ancestor of sect. Viperella was probably diploid (2n = 2x =18) with two subsequent polyplodization events. Total DNA content (2C) varied considerably across the phylogeny with larger genomes detected mainly in polyploid species. Altitude also varied across the...

Description of a ZZ/ZW sex chromosome system in Thoracocharax cf. stellatus (Teleostei, Characiformes, Gasteropelecidae) Genet. Mol. Biol.
Carvalho,Margarida Lima; Oliveira,Claudio; Foresti,Fausto.
The family Gasteropelecidae is composed of three genera and eight species. This study shows that Thoracocharax cf. stellatus has 2n = 52 chromosomes for both sexes. The five males studied showed 8 metacentric, 16 submetacentric, 4 subtelocentric, and 24 acrocentric chromosomes; the seven females showed only one submetacentric chromosome, belonging to pair 11, and one extra acrocentric chromosome, smaller than all the other chromosomes, characterizing the presence of a ZZ/ZW sex chromosome system in this species. Nucleolus organizing regions (NORs) were detected on the short arms of the subtelocentric chromosome pair 13. Constitutive heterochromatin was identified at pericentromeric and terminal positions in almost all chromosomes. The W chromosome was...

Development and application of high-throughput amplified fragment length polymorphism technique in Calluna vulgaris (Ericaceae) Electron. J. Biotechnol.
Borchert,Thomas; Gawenda,Inka.
Calluna vulgaris is an important ornamental crop of the horticultural industry in Europe. In order to improve breeding of this species, especially of the most important trait of ‘bud-flowering', the implementation of molecular techniques that allow rapid, reproducible and efficient screening of whole segregating populations e.g. for molecular marker and mapping approaches is a requirement. We therefore aimed to introduce the powerful tool of amplified fragment length polymorphisms (AFLP®), a widely and successfully applied method, into our methodological assortment. As an essential prerequisite, the isolated DNA should be of adequate quality which is a common obstacle when dealing with woody species and their interfering secondary...

Rizza,Marco Dalla; Real,Daniel; Reyno,Rafael; Porro,Valentina; Burgueño,Juan; Errico,Eugenia; Quesenberry,Kenneth H..
Six species of Trifolium (T. polymorphum Poir., T. riograndense Burkart, T. argentinense Speg., T. medium L., T. pratense L. and T. repens L.) were analyzed using inter-simple sequence repeats (ISSR) markers. Six selected primers generated 186 polymerase chain reaction (PCR) products exploring 112 loci in 34 genotypes analyzed with molecular sizes ranging from 200 to 1300 bp. These primers were able to discriminate among and within species, with the PCR products being on average 41.6% species-specific and 59.9% polymorphic at the within species level. Nuclear DNA content was determined by flow cytometry and revealed variation among species. The 1Cx genome size values were calculated and were found to range from 0.46 pg (T. pratense) to 0.96 pg (T....

Interspecific hybrids between Paspalum plicatulum and P. oteroi: a key tool for forage breeding Scientia Agricola
Novo,Patricia Elda; Valls,José Francisco Montenegro; Galdeano,Florencia; Honfi,Ana Isabel; Espinoza,Francisco; Quarin,Camilo Luis.
ABSTRACT Grama-tio-pedro (Paspalum oteroi Swallen) is a rare stoloniferous grass of the Plicatula group of Paspalum, well adapted to continuous grazing in areas subject to seasonal flooding in the Pantanal region, in central western Brazil. The species is a facultative apomictic (asexual reproduction by seed) tetraploid, sporadically cultivated on Pantanal farms, propagated either by cuttings or seed. Due to its potential for extensive cultivation and forage quality, Grama-tio-pedro appears as a candidate for genetic improvement within the Plicatula group through plant breeding. We used a colchicine-induced sexual autotetraploid genotype of P. plicatulum Michx. to obtain interspecific hybrids using the apomictic species, P. oteroi, as pollen donor. The...

KARYOTYPE ALTERATIONS AFTER HYBRIDIZATION BETWEEN Pennisetum purpureum AND Pennisetum glaucum Ciência e Agrotecnologia
Reis,Gabriela Barreto dos; Andrade-Vieira,Larissa Fonseca; Campos,José Marcello Salabert de; Pereira,Antonio Vander; Davide,Lisete Chamma.
ABSTRACTNapier grass and pearl millet are tropical forages from the genus Pennisetum. The variability in those species is explored in breeding programs of forages, as well as in the production of interspecific hybrids. Hybridization is a phenomenon that leads to intergenomic conflicts following the elimination of genomic sequences. In this sense, the present work aimed to study the genomic alterations occurring after interspecific hybridization of pearl millet and Napier grass with the use of cytogenetics and flow cytometry tools. These methods allowed the evaluation of chromosome morphometry, DNA content and genomic ratio in pearl millet, Napier grass and hybrids. It was observed that pearl millet and Napier grass have chromosomes with superposed size....

Karyotype analysis, DNA content and molecular screening in Lippia alba (Verbenaceae) Anais da ABC (AABC)
Pierre,Patrícia M.O.; Sousa,Saulo M.; Davide,Lisete C.; Machado,Marco A.; Viccini,Lyderson F..
Cytogenetic analyses, of pollen viability, nuclear DNA content and RAPD markers were employed to study three chemotypes of Lippia alba (Mill.) (Verbenaceae) in order to understand the genetic variation among them. Different ploidy levels and mixoploid individuals were observed. This work comprises the first report of different chromosome numbers (cytotypes) in L. alba. The chromosome numbers of La2-carvone and La3-linalool chemotypes suggested that they are polyploids. Flow cytometric analysis showed an increase of nuclear DNA content that was not directly proportional to ploidy level variation. A cluster analysis based on RAPD markers revealed that La3-linalool shares genetic markers with La1-citral and La2-carvone. The analysis showed that the majority...

Karyotype asymmetry in Cynodon Rich. (Poaceae) accessions. Repositório Alice
CHIAVEGATTO, R. B.; PAULA, C. M. P.; SOUZA SOBRINHO, F. de; BENITES, F. R. G.; TECHIO, V. H..
Abstract Cynodon is a genus of plants with forage potential that has attracted the interest of breeders. These species have high morphological variability in a large number of varieties and cytotypes, hampering identification. This study aimed to determine the karyotype asymmetry index among accessions of Cynodon to discriminate between them. Karyotype symmetry was based on three estimates, which were compared. The basic number for the genus is x = 9. The results of the chromosome count and DNA quantification, respectively, were as follows: two diploid accessions (2n = 2x = 18 and 1.08 ± 0.094 to 1.17 ± 0.036 pg DNA and ± standard deviation), one triploid accession (2n = 3x = 27 and 1.63 ± 0.017 pg DNA), four tetraploid accessions (2n = 4x = 36 and 1.88 ±...
