Chrysolaena flexuosa is a South American species of potential ornamental value (basic chromosome number of x=10). Diploid (n=10) and tetraploid (n=20) cytotypes have been reported for its distribution area, although one hexaploid (n=30-32ca.) cytotype has been reported for its most southern distribution. To investigate if ploidy and latitude are positively related in Ch. flexuosa natural populations and if sexual polyploidization could have had a role in the origin of the polyploid cytotypes, we determined chromosome numbers, DNA content, and pollen viability and size, and analyzed microsporogenesis in samples of seven Argentinian accessions. Abstract Sisyrinchium is the largest genus of Iridaceae in the Americas and has the greatest amount of cytological data available. This study aimed at investigating how genomes evolved in this genus. Chromosome number, genome size and altitude from species of sect. Viperella were analyzed in a phylogenetic context. Meiotic and pollen analyses were performed to assess reproductive success of natural populations, especially from those polyploid taxa. Character optimizations revealed that the common ancestor of sect. Viperella was probably diploid (2n = 2x =18) with two subsequent polyplodization events. Description of a ZZ/ZW sex chromosome system in Thoracocharax cf. stellatus (Teleostei, Characiformes, Gasteropelecidae) 74

The family Gasteropelecidae is composed of three genera and eight species. This study shows that Thoracocharax cf. stellatus has 2n = 52 chromosomes for both sexes. The five males studied showed 8 metacentric, 16 submetacentric, 4 subtelocentric, and 24 acrocentric chromosomes; the seven females showed only one submetacentric chromosome, belonging to pair 11, and one extra acrocentric chromosome, smaller than all the other chromosomes, characterizing the presence of a ZZ/ZW sex chromosome system in this species. DNA content estimation of Fig and Black Mulberry using flow cytometry 94

Abstract In this study, fig and black mulberry DNA contents were estimated using DAPI fluorescence stain in flow cytometry. The 2C DNA contents of the fig and black mulberry were found as 0.82 pg and 8.34 pg, respectively. The calculated 1C value of genome size of fig is 401.8 Mbp and that of black mulberry is 4086.6 Mbp. The ratio of 2C DNA content and 1C genome of the black mulberry was 10.17 times that of the fig although fig is diploid and black mulberry is decosaploid.

Flow cytometric determination of genome size in European seabass (Dicentrarchus labrax), gilthead seabream (Sparus aurata), thinlip mullet (Liza ramada), and European eel (Anguilla anguilla) 5

The nuclear DNA content in Dicentrarchus labrax, Sparus aurata, Liza ramada, and Anguilla anguilla, was measured by flow cytometric analysis. Male human leukocytes were used as internal reference cells. Nucleated fish erythrocytes were stained simultaneously to human leukocytes with a staining buffer containing propidium iodide (PI). Nuclear DNA contents of target species were estimated in relation to an assigned value of 7.0 pg DNA for male human leukocytes. The DNA content/nucleus (+/- CI) was 1.55 +/- 0.02 pg for D. labrax, 1.90 +/- 0.03 pg for S. aurata, 1.57 +/- 0.02 pg for L. ramada, and 2.43 +/- 0.04 pg for A. anguilla. Genetic diversity and DNA content of three South American and three Eurasiatic Trifolium species 74

Six species of Trifolium (T. polymorphum Poir., T. riograndense Burkart, T. argentinense Speg., T. medium L., T. pratense L. and T. repens L.) were analyzed using inter-simple sequence repeats (ISSR) markers. Six selected primers generated 186 polymerase chain reaction (PCR) products exploring 112 loci in 34 genotypes analyzed with molecular sizes ranging from 200 to 1300 bp. These primers were able to discriminate among and within species, with the PCR products being on average 41.6% species-specific and 59.9% polymorphic at the within species level. The objective of this work was to evaluate the genomic behavior of hybrid combinations between elephant grass (Pennisetum purpureum) and pearl millet (P. glaucum). Tetraploid (AAA'B) and pentaploid (AA'A'BB) chromosome races resulting from the backcross of the hexaploid hybrid to its parents elephant grass (A'A'BB) and pearl millet (AA) were analyzed as to chromosome number and DNA content. Genotypes of elephant grass, millet, and triploid and hexaploid induced hybrids were compared. ABSTRACTNapier grass and pearl millet are tropical forages from the genus Pennisetum. The variability in those species is explored in breeding programs of forages, as well as in the production of interspecific hybrids. Hybridization is a phenomenon that leads to intergenomic conflicts following the elimination of genomic sequences. In this sense, the present work aimed to study the genomic alterations occurring after interspecific hybridization of pearl millet and Napier grass with the use of cytogenetics and flow cytometry tools. Karyotype analysis, DNA content and molecular screening in Lippia alba (Verbenaceae) 42

Cytogenetic analyses, of pollen viability, nuclear DNA content and RAPD markers were employed to study three chemotypes of Lippia alba (Mill.) (Verbenaceae) in order to understand the genetic variation among them. Different ploidy levels and mixoploid individuals were observed. This work comprises the first report of different chromosome numbers (cytotypes) in L. alba. The chromosome numbers of La2-carvone and La3-linalool chemotypes suggested that they are polyploids. Flow cytometric analysis showed an increase of nuclear DNA content that was not directly proportional to ploidy level variation. Nuclear DNA content determination in Characiformes fish (Teleostei, Ostariophysi) from the Neotropical region 74

In the present study, nuclear DNA content was analyzed in 53 species of Characiformes fish from the Neotropical region. Diploid number ranged from 2n = 48 in Astyanax fasciatus, Gymnocorymbus ternetzi and Hyphessobrycon griemi to 2n = 102 in Potamorhina squamoralevis, with a modal number of 54 chromosomes. Nuclear DNA content ranged from 1.70 ± 0.04 pg of DNA per diploid nucleus in Acestrorhynchus pantaneiro to 3.94 ± 0.09 pg in Tetragonopterus chalceus. A general analysis showed a mean value of 2.9 pg of DNA per diploid nucleus. Nuclear DNA content in 20 species of Siluriformes (Teleostei: Ostariophysi) from the Neotropical region 74

In the present study, 20 species of Siluriformes fish were analyzed in order to determine their nuclear DNA content and compare these data with their diploid number. In addition, the extension and importance of the changes that occurred during the process of diversification in the group of Neotropical freshwater catfish were investigated. The only species studied of the family Doradidae, Rhinodoras d'orbignyi (2n = 58), presented 3.46 ± 0.13 pg of DNA. The objective of the present work was to induce chromosome doubling in genotypes of Brachiaria ruziziensis with potential for production of good-quality biomass and tolerance/resistance to stress caused by aluminum and spittlebugs. Germinated seeds of B. ruziziensis were immersed in 0.1% colchicine solution for 2 or 3 hours. Ploidy level was determined by flow cytometry and confirmed by chromosome count. Fertility of the tetraploid plants was assessed through evaluation of pollen viability by staining. Seeds were subjected to imbibition for 0, 24, 36, 48, 60 and 81 h, then dried to their initial moisture content (13%), rehydrated and evaluated for survival (resumption of growth and development of normal seedlings) and membrane system integrity (electrolyte leakage).
