Assessment of genetic diversity in Venezuelan rice cultivars using simple sequence repeats markers Electron. J. Biotechnol.
Ghneim Herrera,Thaura; Posso Duque,Duina; Pérez Almeida,Iris; Torrealba Núñez,Gelis; Pieters,Alejandro J; Martinez,César P; Tohme,Joe M.
In Venezuela, pedigree analyses indicate that the rice varieties currently under cultivation are closely related. Effective breeding programs, based on knowledge of the genetic diversity of cultivars, are needed to broaden the genetic bases of rice germplasm in the country. In this study, we used a set of 48 simple-sequence-repeat (SSR) markers to assess the genetic diversity of 11 Venezuelan rice cultivars, released by the National Rice Breeding Program between 1978 and 2007. A total of 203 alleles were detected, the number of alleles (NA) per marker ranged from 2 to 9, with an average of 4.23. The average genic diversity (H) over all SSR loci for the 18 genotypes was 0.524, ranging from 0.105 to 0.815. Positive correlations were found between H at each...

Evaluation of Xylella fastidiosa genetic diversity by fAFPL markers Rev. Bras. Frutic.
Kishi,Luciano Takeshi; Wickert,Ester; Lemos,Eliana Gertrudes de Macedo.
The first phytopathogenic bacterium with its DNA entirely sequenced is being detected and isolated from different host plants in several geographic regions. Although it causes diseases in cultures of economic importance, such as citrus, coffee, and grapevine little is known about the genetic relationships among different strains. Actually, all strains are grouped as a single species, Xylella fastidiosa, despite colonizing different hosts, developing symptoms, and different physiological and microbiological observed conditions. The existence of genetic diversity among X. fastidiosa strains was detected by different methodological techniques, since cultural to molecular methods. However, little is know about the phylogenetic relationships developed by...

Fingerprinting of cell lines by directed amplification of minisatellite-region DNA (DAMD) BJMBR
Silva,L.M.; Montes de Oca,H.; Diniz,C.R.; Fortes-Dias,C.L..
The development of in vitro propagation of cells has been an extraordinary technical advance for several biological studies. The correct identification of the cell line used, however, is crucial, as a mistaken identity or the presence of another contaminating cell may lead to invalid and/or erroneous conclusions. We report here the application of a DNA fingerprinting procedure (directed amplification of minisatellite-region DNA), developed by Heath et al. [Nucleic Acids Research (1993) 21: 5782-5785], to the characterization of cell lines. Genomic DNA of cells in culture was extracted and amplified by PCR in the presence of VNTR core sequences, and the amplicons were separated by agarose gel electrophoresis. After image capture with a digital camera, the...

Genetic evaluation of the mating system in the blue-and-yellow macaw (Ara ararauna, Aves, Psittacidae) by DNA fingerprinting Genet. Mol. Biol.
Caparroz,Renato; Miyaki,Cristina Y.; Baker,Allan J..
More than 90% of birds are socially monogamous, although genetic studies indicate that many are often not sexually monogamous. In the present study, DNA fingerprinting was used to estimate the genetic relationships between nestlings belonging to the same broods to evaluate the mating system in the socially monogamous macaw, Ara ararauna. We found that in 10 of 11 broods investigated, the nestlings showed genetic similarity levels congruent with values expected among full-sibs, suggesting that they shared the same parents. However, in one brood, the low genetic similarity observed between nestlings could be a result of intraspecific brood parasitism, intraspecific nest competition or extra-pair paternity. These results, along with available behavioral and...

Genetic, morphological, and chemical patterns of plant hybridization RChHN
López-Caamal,Alfredo; Tovar-Sánchez,Efraín.
Natural hybridization is a frequent phenomenon among vascular plants. Hybridization is considered an important evolutionary force since it may lead to (1) an increase of the intraspecific genetic diversity of the participating populations, (2) the creation of new species, (3) species extinction through genetic assimilation, and (4) the generation of highly invasive genotypes. Because of the importance of plant hybridization in evolution, it is of great importance to accurately identify hybrid individuals. In this review, we give a general historical background of the study of plant hybridization. Also, we review some of the tools employed for hybrid recognition and their pattern of expression in hybrid individuals (morphological, chemical, chromosome...

Genetic variation among species, races, forms and inbred lines of lac insects belonging to the genus Kerria (Homoptera, Tachardiidae) Genet. Mol. Biol.
Ranjan,Sanjeev Kumar; Mallick,Chandana Basu; Saha,Dipnarayan; Vidyarthi,Ambarish S; Ramani,Ranganathan.
The lac insects (Homoptera: Tachardiidae), belonging to the genus Kerria, are commercially exploited for the production of lac. Kerria lacca is the most commonly used species in India. RAPD markers were used for assessing genetic variation in forty-eight lines of Kerria, especially among geographic races, infrasubspecific forms, cultivated lines, inbred lines, etc., of K. lacca. In the 48 lines studied, the 26 RAPD primers generated 173 loci, showing 97.7% polymorphism. By using neighbor-joining, the dendrogram generated from the similarity matrix resolved the lines into basically two clusters and outgroups. The major cluster, comprising 32 lines, included mainly cultivated lines of the rangeeni form, geographic races and inbred lines of K. lacca. The...

LaNe RAGE: a new tool for genomic DNA flanking sequence determination Electron. J. Biotechnol.
Park,Daniel J.
The determination of genomic DNA sequence flanking a known region is often problematic. Existing technologies depend on multiple, efficient enzyme-catalysed preparative processing steps and/or rely on relatively inefficient ‘one-sided' PCR mechanisms. I demonstrate the application of a simple ‘two-sided' PCR-based approach, lariat-dependent nested PCR for rapid amplification of genomic DNA ends (LaNe RAGE), applied to the mouse GAPDH and PGK1 gene flanking sequences. This demonstration offers great promise in applications such as genome walking, transposon mutagenesis mapping and DNA fingerprinting

Molecular markers useful to discriminate Coffea arabica cultivars with high genetic similarity. Repositório Alice
SOUSA, T. V.; CAIXETA, E. T.; ALKIMIM, E. R.; OLIVEIRA, A. C. B. de; PEREIRA, A. A.; ZAMBOLIM, L.; SAKIYAMA, N. S..
New cultivars are released every year to meet market demands. However, in species with a narrow genetic base, such as Coffea arabica, the cultivars are closely related and phenotypically similar. This hinders the accurate discrimination of genotypes sing morphological descriptors in distinctness, uniformity, and stability (DUS) testing, which is required for the registration and protection of new cultivars. In this sense, molecular markers are an auxiliary tool for accurate and precise discrimination of cultivars. This study aimed to verify the informative capacity and effectiveness of a molecular marker set to discriminate among C. arabica varieties, create a database of DNA profiles and allele frequencies, analyze the genetic diversity in this...

Polymorphism in the internal transcribed spacer (ITS) of the ribosomal DNA of 26 isolates of ectomycorrhizal fungi Genet. Mol. Biol.
Gomes,Eliane A.; Kasuya,Maria Catarina M.; Barros,Everaldo G. de; Borges,Arnaldo C.; Araújo,Elza F..
Inter- and intraspecific variation among 26 isolates of ectomycorrhizal fungi belonging to 8 genera and 19 species were evaluated by analysis of the internal transcribed sequence (ITS) of the rDNA region using restriction fragment length polymorphism (RFLP). The ITS region was first amplified by polymerase chain reaction (PCR) with specific primers and then cleaved with different restriction enzymes. Amplification products, which ranged between 560 and 750 base pairs (bp), were obtained for all the isolates analyzed. The degree of polymorphism observed did not allow proper identification of most of the isolates. Cleavage of amplified fragments with the restriction enzymes Alu I, Hae III, Hinf I, and Hpa II revealed extensive polymorphism. All eight genera...

Use of molecular markers SSR and SCAR for identification of olive accessions. Repositório Alice
VAL, A. D. B. do; BREVES, S. de S.; CANÇADO, G. M. de A.; FERREIRA, J. L.; PASQUAL, M..
ABSTRACT: Morphological and agronomical describers are traditionally used in plant characterization. However, the usage of these describers have some limitations such as susceptibility to abiotic and biotic stress and environmental factors. Furthermore, the describers are not stable over time and many can only be evaluated during the adult phase of the plants, which requires time and physical space. Molecular markers offer numerous advantages compared to the conventional alternatives based on phenotype: they are stable and detectable in all vegetable tissues, and are independent of the environment and development phase. One of the main advantages of the use of molecular markers is the time reduction in the identification of genetic diversity among the...

SSR molecular markers SSR are tandem arranged repeats of short DNA motifs (1-6 bp in length) distributed throughout the genome that frequently exhibit variation in the number of repeats at the locus of each rice variety. The SSR markers, therefore, are suitable for detecting polymorphisms between closely related genotypes of Thai local rice varieties from the National Rice Germplasm Bank. In this research, DNA fingerprinting was conducted using 8 Thai local rice varieties. i.e., Niaw Sanpahtawng (NSPT_GB), Khao Gaw Diaw (KGD_GB), Niaw Dam (ND_GB), Khao tah Haeng 17 (KTH17_GB), Jao’ Haw (JH_GB), Pin Gaew 56 (PG56_GB), Jek Chuey Gahb Khiaw (JCGK_GB), Sang Yod Pattalung (SYPL_GB), and 5 standard varieties, i.e., Khao Dawk Mali 105 (KDML105_GB, KDML105_C),...
