In Venezuela, pedigree analyses indicate that the rice varieties currently under cultivation are closely related. Effective breeding programs, based on knowledge of the genetic diversity of cultivars, are needed to broaden the genetic bases of rice germplasm in the country. In this study, we used a set of 48 simple-sequence-repeat (SSR) markers to assess the genetic diversity of 11 Venezuelan rice cultivars, released by the National Rice Breeding Program between 1978 and 2007. A total of 203 alleles were detected, the number of alleles (NA) per marker ranged from 2 to 9, with an average of 4.23. Se utilizó la técnica RAPD (amplificación al azar de ADN polimórfico) para el estudio de la diversidad genética de Oreochromis sp. (tilapia roja) en cinco piscícolas del Valle del Cauca (Colombia) y en la determinación del nivel de introgresión de las especies parentales Oreochromis mosambicus, O. niloticus y O. aureus. Se evaluaron 25 cebadores, ocho fueron polimórficos y se obtuvieron 109 bandas. Los valores de heterocigosidad esperada (0.196 a 0.256) y la estructura genética (Gst = 0.22) para Oreochromis sp. indicaron un elevado grado de polimorfismo y alta estructuración genética. More than 90% of birds are socially monogamous, although genetic studies indicate that many are often not sexually monogamous. In the present study, DNA fingerprinting was used to estimate the genetic relationships between nestlings belonging to the same broods to evaluate the mating system in the socially monogamous macaw, Ara ararauna. We found that in 10 of 11 broods investigated, the nestlings showed genetic similarity levels congruent with values expected among full-sibs, suggesting that they shared the same parents. Natural hybridization is a frequent phenomenon among vascular plants. The lac insects (Homoptera: Tachardiidae), belonging to the genus Kerria, are commercially exploited for the production of lac. Kerria lacca is the most commonly used species in India. RAPD markers were used for assessing genetic variation in forty-eight lines of Kerria, especially among geographic races, infrasubspecific forms, cultivated lines, inbred lines, etc., of K. lacca. In the 48 lines studied, the 26 RAPD primers generated 173 loci, showing 97.7% polymorphism. LaNe RAGE: a new tool for genomic DNA flanking sequence determination Electron. J. Biotechnol.

The determination of genomic DNA sequence flanking a known region is often problematic. Existing technologies depend on multiple, efficient enzyme-catalysed preparative processing steps and/or rely on relatively inefficient ‘one-sided' PCR mechanisms. I demonstrate the application of a simple ‘two-sided' PCR-based approach, lariat-dependent nested PCR for rapid amplification of genomic DNA ends (LaNe RAGE), applied to the mouse GAPDH and PGK1 gene flanking sequences. 以逢機增殖多型性DNA分子標誌鑑別番椒屬野生馴化種原 Taiwan Agricultural Research Institute

[[abstract]]本試驗以源自日本、美國、巴西和匈牙利等地區之番椒屬14個品系,探討利用逢機增殖多型性DNA(RAPD)建立種間之分子標誌,以鑑別番椒屬重要野生馴化種之種原。試驗結果顯示,以UBC327,UBC346,UBC441及UBC457等四個隨意引子,經聚合酵素連鎖反應(PCR),其RAPD之產物具有明顯的多型性表現,且在增殖之多型性DNA片段中,可獲得11個有效之多型性DNA片段,提供作為不同「種」辨識之標誌。其中UBC 327,UBC 346及UBC 457三個引子產生的多型性DNA片段,更具有指紋比對之功效,可以清楚地將14個不同品系加以區別。利用上述四個引子產生之所有多型性DNA片段,一併進行相似性群集分析(UPGMA cluster analysis)之結果顯示,供試的14個品系可分成四個群,第一群包含2個品系,屬於C. chinense;第二群包含4個品系,屬於C. annuum;第三群包含5個品系,也屬於C. annuum;第四群則包含3個品系,屬於C. baccatum。試驗結果證實,RAPD標誌對番椒屬具有指紋比對之效,可作為一種有效且值得信賴之種原鑑別工具。
