A practical molecular identification of nonfermenting Gram-negative bacteria from cystic fibrosis BJM

Abstract Identification of nonfermenting Gram-negative bacteria (NFGNB) of cystic fibrosis patients is hard and misidentification could affect clinical outcome. This study aimed to propose a scheme using polymerase chain reaction to identify NFGNB. This scheme leads to reliable identification within 3 days in an economically viable manner when compared to other methods.

Bovine schistosomiasis, caused by Schistosoma bovis, is a serious veterinary problem in many parts of the worid. The current methods used for the diagnosis of the disease include clinical signs, pathological lesions, parasitological and serological techniques. As clinical signs and parasitological lesions caused by S. bovis are indistinguishable from those induced by other trematode parasites, confirmation of diagnosis by these methods is unreliable. Accuracy of a self-collection kit for the microbiological study of the vaginal content BJID

Diagnosis of vaginal discharge is frequently performed in an empirical way, leading to inadequate treatment. This study tested the accuracy of a self-collection kit for microbiological study of the vaginal content. One hundred and forty-two women of Family Health Program units in Niterói and Piraí cities were enrolled in order to have their vaginal content studied. A brief explanation and a self-collection kit were provided in order to sample the vaginal content. The self-collection kit was composed of one empty plastic tube, two glass slides, a long handle cytobrush, an identification card and guideline notes. ABSTRACT The purpose of the present work was to evaluate the accuracy of quantitative polymerase chain reaction (qPCR) performed on samples of fresh frozen tissue (FT) and formalin-fixed, paraffin-embedded (FFPE) healthy skin. This is a validation study conducted with samples from 46 dogs from an endemic area in Brazil. After sample collection, DNA extractions were conducted using commercial kits and qPCR was oriented to kinetoplast DNA (kDNA) targets of the Leishmania infantum species. The results obtained for the FFPE samples showed 63.6% sensitivity and 77.1% specificity, whereas those obtained for the FT samples showed 100% and 48.6%, respectively. Alteration in the erythrocyte sedimentation rate in dengue patients: analysis of 1,398 cases BJID

A study of the erythrocyte sedimentation rate during the first hour (ESR) in dengue patients would help determine how this parameter is affected by this disease, as well as whether it can be used for diagnosis. One thousand, three hundred and ninety-eight cases of dengue attended at the Dengue Treatment Center, Campos dos Goytacazes Rio de Janeiro, Brazil, were included. The ESR values were classified as normal or elevated and compared by gender and clinical form of the disease. Among the 1,398 cases ESR was normal in 81.25% (n = 1,136), while in 18.75% (n=262) it was elevated. ABSTRACT Glässer's disease is an emergent bacterial disease that affects swine husbandries worldwide causing important economic losses. The aetiological agent, Haemophilus parasuis, is currently divided in fifteen serovars but an increasing number of non-typeable serovars have been reported. Indirect hemagglutination (IHA) is indicated as a serotyping method for H. parasuis. Visceral leishmaniasis is a zoonotic disease caused by parasites of the Leishmania genus. Dog is the major source of infection to man, especially in urban areas. The authors report a case of visceral leishmaniasis in a pit bull female dog from Bocaina, São Paulo, Brazil. The animal presented clinical signs compatible with leishmaniasis, including skin lesions in the body and partial damage of the external ears. The indirect fluorescent antibody test (IFAT) demonstrated a titer of 1280, and promastigote forms of Leishmania sp were isolated by the culture of bone marrow puncture. This paper presents an object-oriented approach to support, on the web, an integrated framework for diagnosis, investigation and treatment of disorders (diseases, computer faults, etc). In this framework, called Fuzzy Covering Theory (FCT), the knowledge is basically modeled through causal associations between objects and inference is abductive. In this new approach, it was necessary to specify the classes model using object - oriented methodology and the knowledge base model that was a flat file in Prolog. These models are discussed in this paper as well as the advantages and limitations of this new approach.

Application of direct agglutination test and fast agglutination screening test for serodiagnosis of visceral leishmaniasis in endemic area of Minas Gerais, Brazil OAK

We have tested an indirect fluorescence technique (IFAT), fast agglutination screening test (FAST) and direct agglutination test (DAT) for the detection of anti-Leishmania antibodies in serum and blood samples from patients with visceral and cutaneous leishmaniasis in Brazil. For visceral leishmaniasis the results obtained with DAT showed a sensitivity and specificity of 100% with a cut-off value of 1:800. The blood and serum samples could also be clearly read in FAST using a 1:100 dilution with the same high sensitivity. Both DAT and FAST were not able to detect significant amounts of antibodies in samples from cutaneous leishmaniasis patients.

A seven month old female goat showed neck swelling, apathy, appetite and weight loss, restlessness, increased salivation, cough, and regurgitation episodes which occurred post feeding. The animal was evaluated through both clinical and radiographic examinations. Plain radiography was performed and contrasted X-ray was done using barium sulfate. For radiological examination, the lateral projection was used for analysis. The results showed a radiopaque content in the esophageal lumen dorsally positioned to heart with density and appearance similar to the rumen. A dilation of the esophagus was reported and a diagnosis of megaesophagus was made. In conclusion, contrast esophagography must be included in the diagnosis of megaesophagus in goats.

Abstract Scientific literature on broiler chicken welfare in Brazilian industrial systems is scarce. This study aimed at assessing broiler chicken welfare on industrial farms in the State of Rio Grande do Sul, Southern Brazil, using the Welfare Quality(r) assessment protocol for poultry, to provide directly applicable scientific information. Results are presented as criteria scores ranging from 0 to 100, with higher scores indicating better welfare; and percentages of prevalence. The scores classified as excellent (above 80) were absence of prolonged thirst, absence of prolonged hunger, litter quality, breast blister and touch test. To assess the potency of the PPD-mallein produced in Brazil, five animals were from a property identified as a focus of glanders. These animals had suggestive clinical signs of the disease and the other five, from a property free from glanders, showed no clinical signs and were serology negative (control group). PPD-mallein from Burkholderia mallei was obtained by precipitation with trichlo-roa-cetic acid and ammonium sulfate. A tricomonose é uma doença venérea de bovinos causada por um protozoário flagelado denominado Tritrichomonas foetus (T.foetus), cujo habitat é o trato genital de bovinos sendo transmitido do macho para a fêmea através da monta ou pelo uso de sêmen contaminado. O T.foetus é um protozoário de morfologia piriforme com três flagelos em posição cranial e um caudal, cujo tamanho oscila entre entre 8-18mm de comprimento e 4-9 de largura. Se caracteriza por possuir 4 flagelos, 3 deles são anteriores e livres e o 4o acompanha a membrana ondulante, que é perfeitamente visível quando em microscopia, localizando-se na parte posterior.

Este trabalho teve como objetivo comparar os resultados do diagnóstico do Lentivírus Caprino, por Imunodifusão em Gel de ágar - IDGA, utilizando o kit comercial americano e o kit nacional produzido com cepa CAEV Cork. Foram utilizados dois rebanhos, sendo um da Embrapa Caprinos submetido a doze anos de programa de controle e um outro rebanho infectado pelo CAEV, que não teve nenhuma ação prévia de controle. Analisando os resultados dos antígenos (nacional e americano) no rebanho não controlado, verificou-se que o antígeno comercial americano, quando foi utilizado pela primeira vez para o diagnóstico apresentou resultados mais significativos do que o nacional. Resumo: O objetivo deste trabalho foi avaliar a utilização periódica de testes de diagnóstico mais sensíveis aliados às práticas de manejo, visando ao controle eficaz da artrite encefalite caprina (CAE). Foram realizadas oito coletas de sangue em matrizes e reprodutores. Da primeira à sétima análise, as coletas foram quadrimestrais, utilizando-se os testes de imunodifusão em gel de agarose (IDGA), ensaio imunoenzimático indireto (ELISA-i) e Western Blot (WB). A oitava coleta aconteceu seis meses após a sétima, utilizando-se o WB e a reação em cadeia de polimerase (PCR).
