An improved system for competent cell preparation and high efficiency plasmid transformation using different Escherichia coli strains Electron. J. Biotechnol.

This paper describes an efficient bacterial transformation system that was established for the preparation of competent cells, plasmid preparation, and for the storage in bacterial stocks in our laboratory. Using this method, a number of different plasmids have been amplified for further experiments. Competent cells for bacterial transformation were prepared by the calcium chloride method with an optimum concentration of 75 mM. Three different strains of Escherichia coli that were tested are DH5α, TG1 and XL1 blue, and the most efficient strain being XL1 blue. Coliformes fecais são definidos como coliformes capazes de fermentar a lactose com produção de gás em 48 h a 45ºC. Escherichia coli, juntamente com algumas cepas de Enterobacter e Klebsiella, podem apresentar essas características. Entretanto, apenas a presença de Escherichia coli em alimentos indica contaminação fecal por ser encontrada em grande quantidade no trato gastrointestinal do homem e animais de sangue quente, não sendo isolada normalmente em outros nichos. Capture immunoassay for LT detection produced by enterotoxigenic Escherichia coli in bacterial isolates BJM

A capture enzyme-linked immunosorbent assay (ELISA), which detects LT-I toxin produced by enterotoxigenic Escherichia coli strains, has beendeveloped. This capture assay was performed using the IgG enriched fraction of anti-LT-I antiserum and IgG2b anti-LT-I monoclonal antibody and allowed a clear distinction between E. coli LT-I - producing and non-producing strains. The estimated accuracy of the assay is 78% for sensitivity, 94% for specificity and 92% for efficiency. Thus, the capture immunoassayis a sensitive tool for detection of E. coli, which produces heat-labile enterotoxin, and is suitable for use in clinical laboratories and epidemiological surveys in developing world.

Different types of water bodies, including lakes, streams, and coastal marine waters, are often susceptible to fecal contamination from a range of point and nonpoint sources, and have been evaluated using fecal indicator microorganisms. The most commonly used fecal indicator is Escherichia coli, but traditional cultivation methods do not allow discrimination of the source of pollution. ABSTRACT Rumen fungi inhabit the gastro-intestinal tract of ruminants and the most non-ruminant herbivores. Rumen fungi produce highly active plant cell wall degrading enzymes, therefore they have gained scientific interest. In this study, genes encoding xylanase (xynA-7) and cellulase (celA-5) were amplified from Neocallimastix sp. GMLF7 and Orpinomyces sp. GMLF5, respectively, and expressed in Escherichia coli. XynA-7 was found to be active only on xylan, however CelA-5 had activity both on carboxymethyl cellulose and lichenan. Fecal bacterial indicator analyses have been widely used for monitoring the water quality. This study was designed to determine the ratio between the density of Escherichia coli and other Thermotolerant Coliforms (TtC) bacteria from freshwater samples collected for a two-year period of monitoring. TtC were enumerated by membrane filtration on mFC agar. E. coli enumeration was done by two methods: TtC colonies identified in mFC were inoculated in EC-MUG or water samples were filtered and inoculated in modified mTEC agar media, and both methods were compared for quantitative recovery of E. coli. One-day-old Cobb 500 broilers were randomly divided into 2 groups and treated with intraperitoneal E. coli or broth injection. Clinical signs of the birds were assessed every day. Spleen and bursa of Fabricius of the birds, post-infection (pi), were collected to evaluate immune organ index. Jejunal tissues were collected to ascertain the expression of TGF-βs, TβRs, and Smads. Après un rappel des objectifs, du fonctionnement et de la méthode d’interprétation des résultats du réseau de contrôle microbiologique REMI, ce rapport présente le bilan national et décrit le programme annuel pour le département de la Charente-Maritime. Il présente l’ensemble des résultats obtenus, en particulier l’estimation de la qualité des zones de production de coquillages classées et l’évolution de leur qualité sur la période 2014-2016. Le programme de surveillance 2016 a été réalisé à 99%. Les données manquantes résultent d’un manque de ressource ou de problèmes de disponibilité. Au total, le dispositif d’alerte a été déclenché à 25 reprises en 2016 et a donné lieu à la réalisation de 37 prélèvements et analyses supplémentaires. Toutes les zones de...
