This study characterized seven Pennisetum purpureum varieties, namely v. Anão, Bajra, Cameroon, Guaçu, Roxo, Taiwan A-144 and Uruckwami, through biochemical analyses, including protein, glucose and fructose contents, and polyacrylamide gel electrophoresis using the esterase system, by sampling 30, 60, 90, 120 and 150 day-old leaves. The number of nodes per stem and the percentage of bud emergence were also recorded. Variety Taiwan A-144 presented the highest number of nodes per stem and percentage of emerging buds. Protein concentration decreased gradually after 60 days for all varieties, except for Anão. Variety Guaçu presented the highest level of glucose in 90 day-old plants, whereas Cameroon presented the highest levels at 120 and 150 days. The... |

The interaction of proteins with polysaccharides represents a major and challenging topic in glycobiology, since such complexes mediate fundamental biological mechanisms. An affinity capillary electrophoresis method has been developed to evidence the complex formation and to determine the binding properties between an anticoagulant polysaccharide of marine origin, fucoidan, and a potential target protein, antithrombin. This method is a variant of zonal electrophoresis in the mobility shift format. A fixed amount of protein was injected into a capillary filled with a background electrolyte containing the polysaccharide in varying concentrations. The effective mobility data of the protein were processed according to classical linearization treatments to... |

Abstract Obtaining collagen and hydrolysates from mechanically separated chicken meat residue is an excellent way to add value to this waste. The aims of this study were to obtain collagen hydrolysates from chicken MSM residue using Alcalase® and Flavourzyme® enzymes, as well characterizing the functional, structural and thermal stability properties. The highest degree of hydrolysis was obtained using Alcalase®, 36.11%, while using Flavourzyme® resulted in 12.02%. The DSC analysis of the collagen indicated a denaturation temperature of 46.47 °C. The FTIR spectra of the crude collagen showed absorption peaks that were characteristic of amide bands A, B, I, II and III. In the spectra of the hydrolysed the area of the amide bands was reduced, and some peaks... |

Background Scorpion venoms are rich bioactive peptide libraries that offer promising molecules that may lead to the discovery and development of new drugs.Leiurus abdullahbayrami produces one of the most potent venoms among Turkish scorpions that provokes severe symptoms in envenomated victims. Methods In the present study, the peptide profile of the venom was investigated by electrophoretic methods, size-exclusion and reversed-phase chromatography and mass spectroscopy. Cytotoxic and antimicrobial effects were evaluated on a breast cancer cell line (MCF-7) and various bacterial and fungal species. Results Proteins make up approximately half of the dry weight of L. abdullahbayrami crude venom. Microfluidic capillary electrophoresis indicated the presence of... |

The objective of this work was to characterize 27 potato genotypes, using molecular markers. Polyacrylamide gel electrophoresis, RAPD techniques and isozymes of esterase, phosphoglucomutase and soluble proteins were analyzed in tubers, and isocitrate dehydrogenase, aspartate transaminase, phosphoglucomutase and peroxidase, in leaves. Eighteen primers were tested and four were chosen, kits OPX (01, 04 and 09) and OPY (07), to analyze RAPD markers in leaf extracts. Similarity and cluster analysis were conducted using Jaccard coefficient and the unweighted pair-group method using arithmetic average. Despite the differences detected in the analysis of proteins and isozymes in the tubers, as well as of isozymes in the leaves, the characterization of all... |

Electrophoretic analysis of eight polymorphic loci in 23 mixed or monospecific populations of Cepaea nemoralis and Cepaea hortensis indicates important genetic dissimilarities between Breton and Pyrenean areas. The concordance between groups of samples and various latitudinal or altitudinal zones suggests that the influence of climatic factors could be involved. Founder effects also seem to increase the differentiation in some populations; the lower genetic variability in three pyrenean samples probably submitted to frequent extinctions could illustrate this process. Nevertheless, divergences corresponding to plurispecific samples suppose the absence of interactions between the two species. |

Of eleven proteins analyzed in four Amazonian populations, the esterases showed the greatest variation, with five activity zones. EST1, EST2 and EST5 showed variation in each of the populations studied. EST1 and EST2 are each controlled by two, and EST5 by four, codomi-nant alleles. LAP presented six activity zones, with codominant variation in LAP5and LAP6.oc—GPDH was monomorphic with one activity band on starch gel and two on polyacrylamide gel. 1DH presented two activity zones, with variation in the IDHl region. PGM had a single activity zone, with variation in all populations. The Ariquemes populations showed five alleles and the other populations three, all of then codominant. Three activity zones with two codominant alleles were observed for ODH.... |

Beetles of the species Sitophilus oryzae and S. zeamais are pests of great economic importance since they attack not only rice and maize but also several other cereals. In fact, these beetles are one of the most visible threats to sustainable food production. Current study estimated the genetic variability of S. oryzae in two samples, one from the State of Paraná (PR), Brazil, and another from the state of Rio Grande do Sul (RS), Brazil, and a sample of S. zeamais from the State of Santa Catarina (SC), Brazil. Isozyme electrophoresis in starch gel technique was employed to analyze eight enzyme systems (AAT, ACP, GDH, GPI, IDH, MDH, PGM and ME). Average heterozygosity rates were 0.0091, 0.0100 and 0.0000 and expected heterozygosity rates were 0.0419, 0.0452... |

Eighty-nine of 144 isolates of Clostridium perfringens obtained from 187 samples of 71 bovine in several Brazilian states were submitted to esterase electrophoresis for typing. Mobilites electrophoresis, as parameter, were settled down by isolates from ATCC pattern of types A, B, C, and D. Of the 89 isolates, 43 (48.3%) were characterized as electrophoretic type A, 20 (22.5%) as D, 18 (20.2%) as C, and 3 (3.4%) as B. Five (5.6%) isolates did not identity with any type. Similarly, esterase electrophoresis enabled the typing of 94.4 % of the isolates, demonstrating to be an appropriate method for animal sample analyses. |
