Biochemical and isoenzyme analyses of elephant grass, Pennisetum purpureum (Schum) varieties Scientia Agricola
Bach,E.E.; Alcântara,V.B.G.; Alcântara,P.B.; Veasey,E.A..
This study characterized seven Pennisetum purpureum varieties, namely v. Anão, Bajra, Cameroon, Guaçu, Roxo, Taiwan A-144 and Uruckwami, through biochemical analyses, including protein, glucose and fructose contents, and polyacrylamide gel electrophoresis using the esterase system, by sampling 30, 60, 90, 120 and 150 day-old leaves. The number of nodes per stem and the percentage of bud emergence were also recorded. Variety Taiwan A-144 presented the highest number of nodes per stem and percentage of emerging buds. Protein concentration decreased gradually after 60 days for all varieties, except for Anão. Variety Guaçu presented the highest level of glucose in 90 day-old plants, whereas Cameroon presented the highest levels at 120 and 150 days. The...

Capillary electrophoresis determination of the binding affinity of bioactive sulfated polysaccharides to proteins: study of the binding properties of fucoidan to antithrombin ArchiMer
Varenne, A; Gareil, P; Colliec-jouault, Sylvia; Daniel, R.
The interaction of proteins with polysaccharides represents a major and challenging topic in glycobiology, since such complexes mediate fundamental biological mechanisms. An affinity capillary electrophoresis method has been developed to evidence the complex formation and to determine the binding properties between an anticoagulant polysaccharide of marine origin, fucoidan, and a potential target protein, antithrombin. This method is a variant of zonal electrophoresis in the mobility shift format. A fixed amount of protein was injected into a capillary filled with a background electrolyte containing the polysaccharide in varying concentrations. The effective mobility data of the protein were processed according to classical linearization treatments to...

Abstract Obtaining collagen and hydrolysates from mechanically separated chicken meat residue is an excellent way to add value to this waste. The aims of this study were to obtain collagen hydrolysates from chicken MSM residue using Alcalase® and Flavourzyme® enzymes, as well characterizing the functional, structural and thermal stability properties. The highest degree of hydrolysis was obtained using Alcalase®, 36.11%, while using Flavourzyme® resulted in 12.02%. The DSC analysis of the collagen indicated a denaturation temperature of 46.47 °C. The FTIR spectra of the crude collagen showed absorption peaks that were characteristic of amide bands A, B, I, II and III. In the spectra of the hydrolysed the area of the amide bands was reduced, and some peaks...

Characterization of potato genotypes using molecular markers PAB
Collares,Eliana Antonia Silveira; Choer,Eva; Pereira,Arione da Silva.
The objective of this work was to characterize 27 potato genotypes, using molecular markers. Polyacrylamide gel electrophoresis, RAPD techniques and isozymes of esterase, phosphoglucomutase and soluble proteins were analyzed in tubers, and isocitrate dehydrogenase, aspartate transaminase, phosphoglucomutase and peroxidase, in leaves. Eighteen primers were tested and four were chosen, kits OPX (01, 04 and 09) and OPY (07), to analyze RAPD markers in leaf extracts. Similarity and cluster analysis were conducted using Jaccard coefficient and the unweighted pair-group method using arithmetic average. Despite the differences detected in the analysis of proteins and isozymes in the tubers, as well as of isozymes in the leaves, the characterization of all...

Divergence genetique de quelques populations naturelles de Cepaea nemoralis et Cepaea hortensis en France ArchiMer
Guiller, A; Madec, L.
Electrophoretic analysis of eight polymorphic loci in 23 mixed or monospecific populations of Cepaea nemoralis and Cepaea hortensis indicates important genetic dissimilarities between Breton and Pyrenean areas. The concordance between groups of samples and various latitudinal or altitudinal zones suggests that the influence of climatic factors could be involved. Founder effects also seem to increase the differentiation in some populations; the lower genetic variability in three pyrenean samples probably submitted to frequent extinctions could illustrate this process. Nevertheless, divergences corresponding to plurispecific samples suppose the absence of interactions between the two species.

ELECTROPHORETIC ANALYSIS OF 11 ENZYMES IN NATURAL POPULATIONS OFROOT, 1926 (DIPTERA: CULICIDAE) IN THE AMAZON REGION Acta Amazonica
SANTOS,Jaselita M.M. dos; TADEI,Wanderli Pedro; CONTEL,Eucléia P.B.
Of eleven proteins analyzed in four Amazonian populations, the esterases showed the greatest variation, with five activity zones. EST1, EST2 and EST5 showed variation in each of the populations studied. EST1 and EST2 are each controlled by two, and EST5 by four, codomi-nant alleles. LAP presented six activity zones, with codominant variation in LAP5and LAP6.oc—GPDH was monomorphic with one activity band on starch gel and two on polyacrylamide gel. 1DH presented two activity zones, with variation in the IDHl region. PGM had a single activity zone, with variation in all populations. The Ariquemes populations showed five alleles and the other populations three, all of then codominant. Three activity zones with two codominant alleles were observed for ODH....

Electrophoretic behavior of ewe milk proteins from local breeds Rembi and Ouled-Djellal of the Algerian central steppe Acta Agron. (Palmira)
Zobiri-Illoul,Assia; Yabrir,Benalia; Laoun,Abbas; Mati,Abderrahmane.
Abstract In order to characterize the production of sheep milk in Algeria (North of Africa) and to detect molecular markers related to the constitution of the protein phase of this milk, we proposed to analyze the electrophoretic behaviors of caseins and serum proteins under various migration conditions (native, urea and SDS-PAGE), from milk collected during the first three months of the year, from two breeds Ouled-Djellal and Rembi, living in the central area steppe. The profiles obtained show a great similarity and homogeneity between the different samples of the milk of the two breeds of ewes studied as to the number and intensity of the revealed migration bands. Some of the latter are nevertheless distinguished from cow’s milk by different levels of...

Electrophoretic spectra of seed storage protein in Cucumis melo and Citrullus vulgaris as a tool for cultivar differentiation National Institute of Agronomic Research
Vladova, R.; Zamfirova, M.; Andreev, T.; Krasteva, L..
Reduced seed storage proteins from 60 cultivars of melon (Cucumis melo) and 52 cultivars of watermelon (Citrullus lanatus) have been investigated electrophoretically using polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate. The cultivars are from the collection of the Institute of Plant Genetics Resources, Sadovo (Bulgaria). The data obtained suggest clearly expressed differences in electrophoretic profiles of components of 7S globulins both for melon and watermelon cultivars. Four phenotypic groups are proposed for melon cultivars (I, II, III, IV) and two (I, II) for watermelon cultivars. This information can be used for cultivar differentiation.
