Action of pesticides to Metarhizium anisopliae in soil Neotropical Entomology
Mochi,Dinalva A.; Monteiro,Antonio C.; Barbosa,José C..
The present study aimed to analyze the action of some acaricides, fungicides, insecticides and herbicides containing different active ingredients on Metarhizium anisopliae Metsch. (Sorokin) inoculated into autoclaved soil. The action of the pesticides was evaluated based on the fungal respiratory activity. The first assessment was done at 48h after inoculation. The pesticides were then added and respiratory activity was determined nine times every 48h and an additional five times every 4 days. Except for the fungicides, no significant effect (P > 0.05) of the pesticides on M. anisopliae was observed. A reduction in CO2 production was observed for the mancozeb treatment from day 4 to day 6 of incubation, and for tebuconazol between days 4 and 6, 8 and...

Biocontrol of pigeon tick Argas reflexus (Acari: Argasidae) by entomopathogenic fungus Metarhizium Anisopliae (Ascomycota: Hypocreales) BJM
Tavassoli,Mosa; Pourseyed,Seyed Hassan; Ownagh,Abdulghaffar; Bernousi,Iraj; Mardani,Karim.
The pigeon tick Argas reflexus is a pathogen-transmitting soft tick that typically feeds on pigeons, but can also attack humans causing local and systemic reactions. Chemical control is made difficult due to environmental contamination and resistance development. As a result, there is much interest in increasing the role of other strategies like biological control. In this study, the efficacy of three strains (V245, 685 and 715C) of entomopathogenic fungus Metarhizium anisopliae for biological control of three life stages of pigeon tick A. reflexus including eggs, larvae, engorged and unfed adults was investigated under laboratory conditions. Five concentrations of different strains of M. anisopliae ranging from 10³ to 10(7) conidia/ml were used. All...

Compatibility of Beauveria bassiana with acaricides Neotropical Entomology
Oliveira,Renato C. de; Neves,Pedro M.O.J..
The in vitro compatibility of the entomopathogenic fungus Beauveria bassiana (Bals.) Vuil. with 12 acaricide formulations was evaluated. The active ingredients Clorfenapyr, Fenpyroximate, Amitraz, Acrinathrin, Hexythiazox, Abamectin, Pyridine, Dimethoate, Pyridaphethion, Fenbutatin Oxide, Azocyclotin and Cyhexatin were tested in three different concentrations (mean concentration - MC, half MC and twice the MC). The formulations tested affected conidial germination, vegetative growth and sporulation of the fungus in different levels. The formulations more compatible with B. bassiana were Avermectin and the pyrethroids. The acaricides belonging to the organophosphate and organostanic chemical groups, however, drastically affected conidial germination as well...

Xavier-Santos,Solange; Magalhães,Bonifácio; Lima,Elza A. Luna-Alves.
The differentiation of a Brazilian isolate of Metarhizium flavoviride (CG 423), a promising candidate for the biocontrol of grasshoppers, was investigated. Conidia were spread onto solid medium (1% yeast extract, 2.8% agar, 96.2% distilled water), incubated at 28°C and observed during 26 h. Germination initiated as conidia size increased from 5.3 (±0.6) x 3.1 (±0.3) µm (0 h incubation) to 8.1 (±0.2) x 6.1 (±0.2) µm (8 h incubation). Germ tubes started to appear after 10 h incubation showing a high degree of multipolarity. Twenty six hours after inoculation, hyphal differentiation and anastomosis among hyphae from adjacent conidia were recorded. Appressoria were formed only from conidia incubated in liquid medium containing minimum concentration of yeast...

Genetic diversity of Hirsutella thompsonii isolates from Thailand based on AFLP analysis and partial beta-tubulin gene sequences Genet. Mol. Biol.
Tigano,Myrian S.; Adams,Byron; Maimala,Saowanit; Boucias,Drion.
Amplified fragment length polymorphism (AFLP) was used to investigate polymorphism among 43 Hirsutella thompsonii isolates (33 from Thailand) obtained from various mite species. The outgroups were an unidentified Hirsutella isolate along with Hirsutella nodulosa and Hirsutella kirchneri. Phylogenetic analyses of the AFLP data showed significant variation among isolates and the existence of three H. thompsonii clades. We also investigated the isolates using PCR with specific primers for the Hirsutella exotoxin gene Hirsutelin A (HtA) and 18 of these isolates were used for sequencing of the partial beta-tubulin gene. Phylogenetic analyses of beta-tubulin sequences showed two distinct H. thompsonii clades, one of which included AFLP clades I and II. For both...

Horizontal transmission and effect of the temperature in pathogenicity of Beauveria bassiana against Diatraea saccharalis (Lepidoptera: Crambidae) BABT
Svedese,Virgínia Michelle; Lima,Elza Áurea de Luna Alves; Porto,Ana Lúcia Figueiredo.
The aim of this work was to evaluate the effect of temperature on the pathogenicity of Beauveria bassiana against Diatraea saccharalis and evaluate the horizontal transmission capacity among the sugarcane borer larvae. The strains were most pathogenic at 26ºC, followed by 32ºC than at 20ºC, with respective mortalities of 100, 50 and 30.3%. The lethal time was also affected by the temperature, with the shortest LT 50 observed at 26ºC (2.2 days) and the longest at 20ºC (16.2 days). The URM2915 strain appeared most effective in all the experiments as having potential for deployment in biological control programs for the sugarcane borer. Results in horizontal transmission of B. bassiana showed that this procedure could also be adopted as control strategy for...

In vitro efficacy of Metarhizium anisopliae sensu lato against unfed Amblyomma parvum (Acari: Ixodidae). Repositório Alice
GARCIA, M. V.; RODRIQUES, V. da S.; MONTEIRO, A. C.; SIMI, L. D.; HIGA, L. de O. S.; MARTINS, M. M.; PRETTE, N.; MOCHI, D. A.; ANDREOTTI, R.; JUAN SZABÓ, M. P..
Amblyomma parvum Aragão (Acari: Ixodidae) is a tick species found with wide distribution in the Neotropical region. Even though it is a wildlife-related tick, it is also a frequent parasite of domestic animals, is aggressive to human beings and may harbor pathogenic microorganisms. Therefore, it is a target species for control on domestic animals, particularly those at the rural?wildlife interface. Herein, the efficacy of two isolates (E9 and IBCB 425) of an entomopathogenic fungus, Metarhizium anisopliae sensu lato, already evaluated for ticks that parasitize domestic animals, was tested against unfed A. parvum adults. Both isolates displayed high acaricidal efficacy after immersion in fungal conidial suspensions for 5 min. Isolate E9 killed all ticks by...

Mechanism of infection and colonization of Rhipicephalus sanguineus eggs by Mertarhizium anisopliae as revealed by scanning eletron microscopy and histopathology BJM
Garcia,Marcos Valerio; Monteiro,Antonio Carlos; Szabo,Matias Juan Pablo; Prette,Nancy; Bechara,Gervásio Henrique.
The present work aimed to verify the penetration pattern of the Metarhizium anisopliae fungus into Rhipicephalus sanguineus tick eggs, as well as the lesions occurred inside the egg. The fungus adherence and penetration were studied by scanning electron microscopy and the action of fungus in the internal tissues was evaluated through conventional histological sessions. For the observation of these events, experimental infections were performed in 11 groups of R. sanguineus eggs containing 25 mg each. The eggs were bathed during 3 minutes under manual shaking in a 10(8) conidia/mL suspension. The bath was performed only in the suspension vehicle for the control groups. The eggs were processed for histopathological analysis and scanning electron microscopy...

Molecular characterization of Paecilomyces fumosoroseus (Deuteromycotina: Hyphomycetes) isolates Scientia Agricola
Azevedo,Andréia Cristiane Souza; Furlaneto,Márcia Cristina; Sosa-Gómez,Daniel Ricardo; Fungaro,Maria Helena Pelegrinelli.
ITS and RAPD analyses were used to investigate molecular variations within samples of Paecilomyces isolates and to resolve five morphologically atypical isolates resembling P. fumosorosus, obtained from whitefly in Northern Paraná State. The ITS4-ITS5 amplicon was 700 base pairs (bp) long in all isolates. The five isolates of Paecilomyces not assigned to species produced restriction profiles identical to all the reference strains of P. fumosoroseus. The extent of fingerprint variability observed by RAPD was sufficient to discriminate all the isolates. The genetic similarity among unidentified isolates and strains of P. fumosoroseus was even higher than that observed among reference strains of this species, allowing us to conclude that isolates CNPso-P77,...

Polymorphism of the grasshopper Schistocerca pallens (Thunberg) (Orthoptera: Acrididae) and its natural pathogen Metarhizium flavoviride Gams & Rozsypal (Hyphomycetes), revealed by RAPD analysis Anais da SEB
Silveira,Eni B.; Al-Janabi,Salah M.; Magalhães,Bonifácio P.; Carvalho,Luiz J.C.B.; Tigano,Myrian S..
The genetic variability of 27 individuals from two distinct populations (Northeast and Central Brazil) of the grasshopper Schistocerca pallens (Thunberg) and eight isolates of its natural pathogen, the fungus Metarhizium flavoviride Gams & Rozsypal, was investigated using RAPD analysis. For the grasshoppers, ten different 10-mer oligonucleotide primers of arbitrary sequence were selected for analysis, resulting in 79 scorable binary characters. The program RAPDDIP applied to S. pallens revealed nucleotide diversity of 2.3 and 2.2% for the populations from Rio Grande do Norte (Northeast Brazil) and Federal District (Central Brazil), respectively. These values indicate the presence of high genetic variability within these populations. Conversely, the...
