A DNA pooling based system to detect Escherichia coli virulence factors in fecal and wastewater samples BJM
Luz María Chacón,J; Lizeth Taylor,C; Carmen Valiente,A; Irene Alvarado,P; Ximena Cortés,B.
The availability of a useful tool for simple and timely detection of the most important virulent varieties of Escherichia coli is indispensable. To this end, bacterial DNA pools which had previously been categorized were obtained from isolated colonies as well as selected in terms of utilized phenotype; the pools were assessed by two PCR Multiplex for the detection of virulent E. coli eaeA, bfpA, stx1, stx2, ipaH, ST, LT, and aatA genes, with the 16S gene used as DNA control. The system was validated with 66 fecal samples and 44 wastewater samples. At least one positive isolate was detected by a virulent gene among the 20 that were screened. The analysis of fecal samples from children younger than 6 years of age detected frequencies of 25% LT positive...

A high-copy T7 Escherichia coli expression vector for the production of recombinant proteins with a minimal N-terminal His-tagged fusion peptide BJMBR
Ramos,C.R.R.; Abreu,P.A.E.; Nascimento,A.L.T.O.; Ho,P.L..
We report here the construction of a vector derived from pET3-His and pRSET plasmids for the expression and purification of recombinant proteins in Escherichia coli based on T7 phage RNA polymerase. The resulting pAE plasmid combined the advantages of both vectors: small size (pRSET), expression of a short 6XHis tag at N-terminus (pET3-His) and a high copy number of plasmid (pRSET). The small size of the vector (2.8 kb) and the high copy number/cell (200-250 copies) facilitate the subcloning and sequencing procedures when compared to the pET system (pET3-His, 4.6 kb and 40-50 copies) and also result in high level expression of recombinant proteins (20 mg purified protein/liter of culture). In addition, the vector pAE enables the expression of a fusion...

A microbiological survey on tomatoes (Lycopersicon esculentum) marketed in Rio de Janeiro, Brazil. Repositório Alice
PENTEADO, A. L.; CASTRO, M. F. D. P. M. de; PAZIANOTTO, R. A. A..
Abstract: Fresh produce can be a possible source of microbiological contamination. In the past 20 years, several salmonellosis outbreaks due to the consumption of tomato have been reported, mainly in the USA. Organic raw vegetables pose a risk for the transmission of foodborne pathogens since they are often cultivated using manure as a fertilizer. The aim of this study was to conduct a survey of the presence of Salmonella spp, total coliforms and Escherichia coli on the surface of tomatoes from two different production systems. A total of 262 samples of organic and conventional tomatoes were collected from supermarkets and open street markets in the city of Rio de Janeiro, Brazil and analyzed for these microorganisms. To increase the probability of...

A new sterile technique effective on capturing tramp ants for microbiological investigations Neotropical Entomology
Schuller,Lucia; Matté,Glavur R; Matté,Maria H.
Tramp ants are an outstanding group of organisms that have a definitive and important role in carrying pathogens. The purpose of this study was to develop a microbiological sterile technique of collecting ants from contaminated areas. The traps were composed of Petri dishes containing culture media, and were tested to verify the applicability of the system. A positive correlation between the microorganism growth and the presence of ants inside or around traps was observed. The technique described demonstrated to be useful to collect ants from different environments, helping the surveillance of pathogenic microorganisms that are of public health concern.

Action of peracetic acid on Escherichia coli and Staphylococcus aureus in suspension or settled on stainless steel surfaces BJM
Kunigk,Leo; Almeida,Maria C.B..
The efficiency of a commercial peracetic acid sanitizer on destruction of Staphylococcus aureus and Escherichia coli was evaluated using two distinct methods. The first method is the AOAC suspension test and the second is a method proposed by one of the authors in which the microbial cells are settled on a stainless steel surface and then treated with the sanitizer. The results showed that when in suspension S. aureus was more resistant to the sanitizer than E. coli. When S. aureus was settled on the stainless steel surface, the contact time between the sanitizer and the microorganisms to attain a 6.5 log reduction in the number of viable cells was three times greater than when the cells were in suspension.

Adaptative responses of E-coli to marine environmental stresses. a modelling approach based on viability and dormancy concepts ArchiMer
Martin, Y; Troussellier, Marc; Bonnefont, Jl.
In an attempt to synthesize conceptual and experimental information on the behaviour of enteric bacteria in seawater, a mathematical model has been developed. This model is based on changes in intracellular metabolisable components depending an physiological responses of the cell when subjected to nutrient starvation, salinity stress and solar radiation. Following a strategy which takes into account short- and mid-term physiological adaptation and reversal processes, the cells can develop in different states: culturable (B-1), viable and definitively nonculturable (B-2), and reversibly dormant (B-3) cells. Model parameters were deduced either from original microcosm experiments, from literature data or calibration procedures. Validation of the model was...

Alkaline gel electrophoresis assay to detect DNA strand breaks and repair mechanisms in Escherichia coli BABT
Mattos,José Carlos Pelielo de; Motta,Ellen Serri da; Oliveira,Márcia Betania Nunes de; Dantas,Flávio José da Silva; Araujo,Adriano Caldeira de.
Reactive oxygen species (ROS) can induce lesions in different cellular targets, including DNA. Stannous chloride (SnCl2) is a ROS generator, leading to lethality in Escherichia coli (E. coli), with the base excision repair (BER) mechanism playing a role in this process. Many techniques have been developed to detect genotoxicity, as comet assay, in eukaryotic cells, and plasmid DNA agarose gel electrophoresis. In this study, an adaptation of the alkaline gel electrophoresis method was carried out to ascertain the induction of strand breaks by SnCl2 in bacterial DNA, from E. coli BER mutants, and its repair pathway. Results obtained show that SnCl2 was able to induce DNA strand breaks in all strains tested. Moreover, endonuclease IV and exonuclease III play...

Antagonic effect of the inhibition of inducible nitric oxide on the mortality of mice acutely infected with Escherichia coli and Bacteroides fragilis BJMBR
Astolfi,R.S.; Khouri,D.G.; Brandizzi,L.I.V.; Ávila-Campos,M.J.; Andrade Jr.,H.F. de.
Sepsis, the leading cause of death in intensive care units, is associated with overproduction of nitric oxide (NO) due to inducible NO synthase (iNOS), responsible for some of the pathologic changes. Aminoguanidine (AG) is a selective iNOS inhibitor with reported inconsistent actions in sepsis. To investigate the influence of iNOS, we studied models of acute bacterial sepsis using acute challenges with aerobic (Escherichia coli) and anaerobic (Bacteroides fragilis) bacteria in the presence of AG. Six-week-old, 23 g, male and female BALB/c and C57Bl/6j mice, in equal proportions, were inoculated (ip) with bacteria in groups of 4 animals for each dose and each experiment in the absence or presence of AG (50 mg/kg, ip, starting 24 h before challenge and daily...

ANTAGONISM AGAINST VIBRIO CHOLERAE BY BACTERIAL DIFFUSIBLE COMPOUND IN THE FECAL MICROBIOTA OF RODENTS Rev. Microbiol.
Silva,Simone Helena da; Vieira,Enio Cardillo; Nicoli,Jacques Robert.
In an ex vivo agar plate assay, we monitored the appearance of an inhibitory halo against Vibrio cholerae from the feces of Wistar and Fischer rats aged 10 to 42 days. The frequency of Wistar rats showing halo increased from 0% (10 days) to a maximum of 80.0% (29 days) and then decreased to 53.3% (42 days). A similar pattern was obtained with Fischer rats but with a lower intensity (maximum frequency of 50.0% by day 36). In a separate experiment, when Wistar rats were fed a low-protein diet for 7 days, the inhibitory halo decreased drastically. Three apparently different colony morphologies were isolated from the dominant fecal microbiota: a facultative anaerobe (FAN) and two strict anaerobes (SAN). The ex vivo inhibitory test showed a halo around the...

Antibiotic resistance patterns of pediatric community-acquired urinary infections BJID
Guidoni,Eliana Biondi Medeiros; Berezin,Eitan N.; Nigro,Stanley; Santiago,Nataly A; Benini,Vanda; Toporovski,Julio.
Knowledge about antimicrobial resistance patterns of the etiological agents of urinary tract infections (UTIs) is essential for appropriate therapy. Urinary isolates from symptomatic UTI cases attended at Santa Casa University Hospital of São Paulo from August 1986 to December 1989 and August 2004 to December 2005 were identified by conventional methods. Antimicrobial resistance testing was performed by Kirby Bauer's disc diffusion method. Among the 257 children, E. coli was found in 77%. A high prevalence of resistance was observed against ampicillin and TMP/SMX (55% and 51%). The antibiotic resistance rates for E. coli were: nitrofurantoin (6%), nalidixic acid (14%), 1st generation cephalosporin (13%), 3rd generation cephalosporins (5%), aminoglycosides...

Camargo,L.R.P.; Suffredini,I.B..
