Abstract The objective of this work was to evaluate the genetic variability for the synthesis of bioactive compounds and minerals in pumpkin (Cucurbita moschata) landraces. Total phenolic compounds, carotenoids, antioxidant activity and minerals were evaluated in 10 accessions of Cucurbita moschata landraces from the Genebank of Embrapa Temperate Agriculture (Pelotas - RS, Brazil). Twenty plants of each accession were cultivated in the field during the spring/summer of 2013/2014. After harvesting of mature fruits, the seeds were discarded and opposite longitudinal portions of the pulp were manually prepared for analysis of the bioactive compounds. Crop genetic resources are the basis of agricultural production, and significant economic benefits have resulted from their conservation and use. However, crop genetic resources are largely public goods, so private incentives for genetic resource conservation may fall short of achieving public objectives. Within the U.S. germplasm system, certain crop collections lack sufficient diversity to reduce vulnerability to pests and diseases. Many such genetic resources lie outside the United States. As a consequence of the difficulty in conventional coffee seed storage, biotechnological alternatives such as cryopreservation have been investigated. The objective of this study was to develop a protocol for the cryopreservation of Coffea arabica L. (cv. ?Catuaí Vermelho? - IAC 144) zygotic embryos by vitrification. For the cryopreservation study, the embryos were immersed in Plant Vitrification Solution 2 at different times (0, 10, 25, 50, 100, and 250 min) and two temperatures (0 and 25 °C). ABSTRACT Conservation of plant genetic resources is important to prevent genetic erosion. Seed banks are the most common method of ex situ conservation; however, coffee seeds can not be stored by conventional methods. Cryopreservation is a viable alternative for long-term conservation of species that produce intermediate or recalcitrant seeds, as coffee. Cryopreservation of zygotic embryos of the Brazilian Green Dwarf coconut PAB

Abstract: The objective of this work was to evaluate rewarming procedures and recovery media for Brazilian Green Dwarf coconut (Cocos nucifera) embryos cryopreserved by vitrification. The rewarming procedures evaluated were: T1, water bath at 38±2°C; and T2, unloading solution consisting of Y3 medium + 1.2 mol L-1 sucrose. The recovery media assessed were: M1, Y3 medium + 45 g L-1 sucrose + 1 mg L-1 gibberellic acid + 1 g L-1 activated charcoal + 2.2 g L-1 Gelrite; and M2, Y3 medium + 60 g L-1 sucrose + 2.2 g L-1 Gelrite. Rewarming procedures showed no significant effect, and the M1 media induced 72.5% regeneration.

The present study aimed at estimating genetic parameters and variability among and within provenances for silvicultural traits of Handroanthus vellosoi (Toledo) Mattos, in provenances and progenies test stands, located at the Luiz Antônio Experimental Station (São Paulo State, Brazil). The test was established in the compact family block design, with two provenances (allocated in the plots), six replicates and sub-linear plots of five plants, totalizing 35 progenies. Foram avaliados os parâmetros genéticos e fenotípicos da bifurcação (BIF), forma do fuste (RET) e diâmetro à altura do peito (DAP) para as procedências e progênies de Dipteryx alata. O experimento foi instalado em delineamento em blocos ao acaso, com 43 tratamentos (26 e 17 progênies dos municípios de Três Lagoas e Brasília no Estado de Mato Grosso do Sul e Distrito Federal, respectivamente) e cinco plantas por parcela. O espaçamento entre plantas foi de 3 x 3 metros. A análise dos dados foi realizada pelo método REML / BLUP (máxima verossimilhança restrita melhor predição linear não viesada). In vitro conservation of blackberry genotypes under minimal growth conditions and subsequent large-scale micropropagation PAB

Abstract: The objective of this work was to evaluate the micropropagation of blackberry (Rubus spp.) cultivars, after in vitro conservation under minimal growth conditions. Nodal segments of the 'Guarani', 'Caingangue', 'Ébano', and 'Xavante' genotypes were conserved under minimal growth conditions at 20ºC, for 15 months. Microshoots were regenerated and multiplied by up to five successive subcultures, when they were rooted and acclimatized. After 30 days of acclimatization in a greenhouse, rooted plantlets showed no significant losses. Blackberry cultivars can be conserved in vitro for 15 months, without subcultures and, after this time, they can be micropropagated on a large-scale, maintaining the regenerative potential and multiplication.

Abstract: The objective of this work was to evaluate the effects of temperature and osmotic agents on the in vitro conservation of Cape gooseberry (Physalis peruviana). Temperatures at 18 and 25°C, as well as the osmotic agents sucrose, mannitol, and sorbitol were tested. A short-term in vitro conservation of Cape gooseberry can be achieved at 18°C, using 30 g L-1 sucrose.

In vitro conservation of Piper aduncum and Piper hispidinervum under slow-growth conditions PAB

The objective of this work was to evaluate in vitro storage of Piper aduncum and P. hispidinervum under slow-growth conditions. Shoots were stored at low temperatures (10, 20 and 25°C), and the culture medium was supplemented with osmotic agents (sucrose and mannitol - at 1, 2 and 3%) and abiscisic acid - ABA (0, 0.5, 1.0, 2.0 and 3.0 mg L-1). After six-months of storage, shoots were evaluated for survival and regrowth. Low temperature at 20ºC was effective for the in vitro conservation of P. aduncum and P. hispidinervum shoots. In vitro cultures maintained at 20ºC on MS medium showed 100% survival with slow-growth shoots. Acceleration of environment degradation and climate change poses serious threats to both natural and agricultural ecosystems. In fact, the number of endangered plant species at the global level has increased yearly in the last decades, due to anthropic interventions that sum up to the climate change. Samples of natural plant populations, particularly of wild crop relatives, are ex situ conserved in germplasm banks, which have become active in the provision of genetic resources to breeders. The aim of this article is to generate discussion on the adequacy of (a) current protocols for sampling and ex situ conservation of the natural genetic diversity and (b) the actual species paradigm in plants, due to their consequences for this and future generations.
