18S-rDNA SEQUENCING, ENZYME PATTERNS AND MORPHOLOGICAL CHARACTERIZATION OF TRICHOPHYTON ISOLATES BJM
Nascimento,Adriana Mendes do; Martinez-Rossi,Nilce M..
Dermatophytes, capable to use keratin of the host for nutrition, belong to one of the major groups of pathogenic fungi. Since dermatophytes are a closely related group they share various common features, and the morphology of isolates of a given species can be atypical, making species identification and differentiation even more difficult. Many methods have been explored in attempts to distinguish dermatophytes, but the combined use of different approaches for the investigation of the intraspecific and interspecific variability of Trichophyton continues to be scarce. Some studies have shown that amplified fragments of the small ribosomal DNA subunit 18S contains variable regions which can be used to discriminate between medically relevant yeast species,...

Antagonism of Trichoderma species on Cladosporium herbarum and their enzimatic characterization BJM
Barbosa,Maria Angélica G.; Rehn,Kurt Georg; Menezes,Maria; Mariano,Rosa de Lima R..
The verrucose caused by Cladosporium herbarum reduces production and quality of Passion fruit (Passiflora edulis Sims.), a largely consumed tropical fruit. This work aimed to investigate the antagonism of Trichoderma species (T. polysporum, T. koningii, T. viride and T. harzianum) against Cladosporium herbarum, and to study the production of extracellular hydrolytic enzymes by the pathogen and the antagonists. The results showed considerable antagonistic potential for the biocontrol of C. herbarum isolates by all Trichoderma species, except T. koningii. The most distinguished effect was observed for T. polysporum. In relation to the pattern of esterase obtained by electrophoresis in poliacrylamyde gel, the major activity presented by the isolates was...

Characterization of streptomycetes with potential to promote plant growth and biocontrol Scientia Agricola
Sousa,Carla da Silva; Soares,Ana Cristina Fermino; Garrido,Marlon da Silva.
Studies with streptomycetes in biocontrol programs and plant growth promotion are presented as technological alternatives for environmental sustainable production. This work has the objective of characterizing six isolates of streptomycetes aiming the production of extracellular enzymes, indole acetic acid, capacity for phosphate solubilization, root colonization and growth under different pH and salinity levels. For detection of enzyme activity the isolates were grown in culture media with the enzyme substrates as sole carbon source. The root colonization assay was performed on tomato seedlings grown on 0.6% water-agar medium. Growth under different pH and salinity levels was evaluated in AGS medium with 1%, 1.5%, 2%, 2.5%, and 3% NaCl, and pH levels...

Enzymatic activity measured by microcalorimetry in soil amended with organic residues Rev. Bras. Ciênc. Solo
Cenciani,Karina; Santos Freitas,Sueli dos; Critter,Silvana Auxiliadora Missola; Airoldi,Claudio.
Enzymatic activity is an important property for soil quality evaluation. Two sequences of experiments were carried out in order to evaluate the enzymatic activity in a soil (Rhodic Eutrudox) amended with cattle manure, earthworm casts, or sewage sludges from the municipalities of Barueri and Franca. The activity of commercial enzymes was measured by microcalorimetry in the same soil samples after sterilization. In the first experiment, the enzyme activities of cellulase, protease, and urease were determined in the soil samples during a three month period. In the second sequence of experiments, the thermal effect of the commercial enzymes cellulase, protease, and urease on sterilized soil samples under the same tretaments was monitored for a period of 46...

Extracellular enzymes of Fusarium graminearum isolates BABT
Kikot,Gisele Eleonora; Hours,Roque Alberto; Alconada,Teresa María.
Fusarium graminearum isolates from three different agroecological regions in Argentina were examined according to the production of different extracellular enzyme activities of potential biotechnological interest: pectinases (PGase: polygalacturonase and PMGase: polymethylgalacturonase), cellulase (CMCase: carboxymethylcellulase) and hemicellulase (xylanase). The isolates were grown in minimum salt medium supplemented with 0.25% glucose, 0.125% citric pectin and 0.125% oat bran as carbon sources and/or enzyme inducers. PGase activity was detected early (after two days of incubation) in all the cultures; it was found to be the highest for all the isolates. PMGase was high only for those isolates of the II region. CMCase and endoxylanase activities were...

Model of experimental infection in healthy and immunosuppressed swiss albino mice (Mus musculus) using Candida albicans strains with different patterns of enzymatic activity BJM
Chaves,Guilherme M.; Cavalcanti,Maria Auxiliadora de Q.; Carneiro-Leão,Ana Maria dos A.; Lopes,Silvia L..
With the purpose to evaluate Candida albicans virulence in vivo, two strains selected were based on their phospholipase and proteinase activity, and used in a model of experimental infection. One strain, isolated from vaginal secretion, was stocked at the URM Culture Collection for 43 years and presented high phospholipase (Pz=0.217) and proteinase (1.386 U.mL-1) activity. The other strain was a fresh strain isolated from oropharyngeal secretion of an AIDS patient, and presented low phospholipase (Pz=0.482) and proteinase (0.780 U.mL-1) activity. The strains were inoculated via intraperitoneum in immunosuppressed and non-immunosuppressed mice (Mus musculus) and the infection was evaluated over a period of 21 days. Liver, spleen, lungs and kidneys were...

Optimization of the production of extracellular α-amylase by Kluyveromyces marxianus IF0 0288 by response surface methodology BABT
Stergiou,Panagiota-Yiolanda; Foukis,Athanasios; Theodorou,Leonidas; Papagianni,Maria; Papamichael,Emmanuel.
The aim of this work was to study the production of extracellular α-amylase by Kluyveromyces marxianus IF0 0288 using optimized nutritional and cultural conditions in a complex yeast medium under aerobic batch fermentation. By applying the conventional "one-variable-at-a-time" approach and the response surface methodology, the effect of four fermentation parameters (type of carbon source, initial culture pH, temperature, and incubation time) on the growth and α-amylase production was evaluated. The production of α-amylase during 60 h of fermentation increased 13-fold under optimized conditions (1% starch, pH 6.0, 30ºC) in comparison to the conventional optimization method. The initial pH value of 6.13 and temperature of 30.3ºC were optimal conditions by...

Polyphasic characterization of endophytic bacteria of sorghum grown on cerrado soil of the Goiás State - Brazil. Repositório Alice
SILVA, L. B. da; SILVA, L. L. da; FERREIRA, E. P. de B.; SANTOS, K. F. E. N.; MARTIN-DIDONET, C. C. G..
Sorghum is a cereal of great commercial importance adapted to the tropical and subtropical regions. Isolation of plant-associated bacteria of agronomic interest has been the subject of several research works, however, there are few studies on bacteria isolating from sorghum. The objective of this work was to characterize as plant growth promoters endophytic bacteria isolated from sorghum roots. A total of 24 isolates were selected using the NFb-lactate, LGI-P, and YM semi-selective medium. The characterization was based on the resistance to antibiotics, use of carbon sources, enzyme activity and 16S-23S intergenic analysis. The isolates were more sensitive to the antibiotic streptomycin and showed a higher capacity to produce lipases and cellulases. Carbon...

Population dynamics and extracellular enzymes activity of mesophilic and thermophilic bacteria isolated from semi-arid soil of Northeastern Brazil BJM
Gorlach-Lira,Krystyna; Coutinho,Henrique D.M..
The dynamics of mesophilic and thermophilic bacterial population was studied in the bulk soil and rhizosphere of the grass Aristida adscensionis L. in a caatinga of the semi-arid Brazilian Northeast. Mesophilic heterotrophic bacteria, sporeforming bacteria and actinomycetes were significantly more abundant than thermophiles, and their counts were 10(6) - 10(7) CFU g-1 dry soil, showing weak fluctuations over the one-year study period. Thermophiles were below 10(6) CFU g-1 dry soil and, in general, demonstrated stronger fluctuations over the studied period. Among them, thermophilic actinomycetes showed the greatest decline during the dry season. No significant differences in microbial number were observed between bulk and rhizosphere soil. Mesophilic...

Presence of intra and extracellular enzymes in strains of Acinetobacter spp, produced in different inducers. Repositório Alice
SCRAMIN, S.; ROQUE, M. R. A.; DENTZIEN, A. F. M.; ROSSO, C. R. S. de; MELO, I. S. de.
Two Acinetobacter spp. strains, denominated D-12 and D-16 (extracted from Diuron TM treated sugarcane rhizosphere) were grown in different substrates in order to determine whether catechol 1,2-dioxygenase (pyrocatechase) and catechol 2,3-dioxygenase (metapyrocatechase) are constitutive or induced enzymes. Extracellular enzymes were also measured. Although biomass was measured, the conditions required for best production of microorganisms were not established. Five different inducers were added to culture media: glucose, sodium benzoate, Diuron TM, dichloroaniline and Diuron TM plus glucose. Both strains were harvested by centrifugation and disrupted by sonication for intracellular enzyme extraction. Extracellular enzyme assays were carried out with...
