Background The production of biofuels from renewable energy sources is one of the most important issues in industrial biotechnology today. The process is known to generate various by-products, for example crude glycerol, which is obtained in the making of biodiesel from rapeseed oil. Crude glycerol may be utilized in many ways, including microbial conversion to 1,3-propanediol (1,3-PD), a raw material for the synthesis of polyesters and polyurethanes. The immobilization of Saccharomyces cerevisiae DSM 70424, Saccharomyces ludwigii DSM 3447 and Saccharomyces rouxii DSM 2531 on brewer's spent grain and then ethanol production and sugar consumption of these immobilized yeasts were investigated. The aim of this study was to investigate the abilities of these three immobilized yeasts for producing alcohol for brewing at two temperatures (7 and 12 °C) using two different sugar levels (one at original level supplied in the brewery and one with 2.5% (w/v), added glucose to the wort). Increasing both parameters resulted in higher alcohol production by all the yeasts studied. A cost effective fermentative production of glutathione by Saccharomyces cerevisiae with cane molasses and glycerol BABT

This work aimed to evaluate the effect of sugar cane molasses and glycerol on glutathione (GSH) fermentation by Saccharomyces cerevisiae ATCC 7754 in flask culture using response surface methodology. Under optimized conditions (80 g/L of molasses and 60 g/L of glycerol), the highest GSH and biomass concentration achieved were 119.6 mg/L and 25.3 g/L, respectively. Further studies done in 5 L bioreactor resulted 241.3 mg/L GSH after 96 h in batch fermentation without amino acids addition and the concentration of biomass was 12.1 g/L. Abstract White wheat flour, concentrated full fat yoghurt, tomato paste, onion, red and green paprika, and mint and salt are used in the preparation of Tarhana. During the 7-day Tarhana fermentation period, the acidity increased from 1.10% to 3.25%, the pH decreased from 5.22 to 4.13, and the moisture decreased from 70.12% to 26.15%. The chemical composition of the Tarhana at the end of fermentation was determined as: moisture 9.55%, protein 12.05%, total ash 5.65%, salt 5.65%, and fat 4.88%. A new alkalophilic isolate of Bacillus as a producer of cyclodextrin glycosyltransferase using cassava flour BJM

Abstract Cyclodextrin glycosyltransferase (CGTase) catalyzes the conversion of starch into non-reducing cyclic sugars, cyclodextrins, which have several industrial applications. This study aimed to establish optimal culture conditions for β-CGTase production by Bacillus sp. SM-02, isolated from soil of cassava industries waste water lake. The optimization was performed by Central Composite Design (CCD) 2, using cassava flour and corn steep liquor as substrates. The maximum production of 1087.9 U mL−1 was obtained with 25.0 g L−1 of cassava flour and 3.5 g L−1 of corn steep after 72 h by submerged fermentation. Alcoholic fermentation by the wild yeasts under thermal, osmotic and ethanol stress BABT

This study aimed to explore the variability in the metabolism of nine wild yeasts isolated from the sugarcane juice from a distillery in the Brazilian State of Mato Grosso. Cell viability under the stress conditions was evaluated. The yeasts were inoculated in the test tubes containing sugarcane juice adjusted from 12 to 21º Brix, ethanol from 6 to 12% in volume and temperature at 30, 35 and 40ºC. The viability was established by the growth in Petri dishes and visually by the CO2 production in the test tubes. None of the evaluated yeasts showed simultaneous resistance to the three stress conditions. The potential of yeast BB.09 could be emphasized due to its ability to ferment up to12% ethanol at 30°C.

Alkali pretreated of wheat straw and its enzymatic hydrolysis BJM

The efficiency of enzymatic hydrolysis of cellulose can be improved by various pretreatments of the substrate. In order to increase the efficiency of enzymatic saccharification of the wheat straw, we determined the effect of different pretreatments on the physical structure, chemical components and enzymatic saccharification of wheat straw. Our results showed that combination of grinding and sodium hydroxide (NaOH) treatment had high effect on the enzymatic hydrolysis of wheat straws. The optimal pretreatment condition was to grind the wheat straws into the sizes of 120 meshes followed by treatment with 1.0% NaOH for 1.5 h (121°C/15psi). Agroindustrial co-products are a viable alternative for use in animal nutrition. Tests were conducted using eight different types of co-products and feed to evaluate the chemical composition, in vitro digestibility of dry matter, crude protein and neutral detergent fiber, and gas production by them. Treze genotipos de sorgo foram agrupados pela suculencia de seus colmos e apresentaram niveis de materia seca (MS) variando entre 24,54 e 34,27%, para o material original, e de 23,11 a 36,12% para suas silagens. Diferencas significativas foram observadas entre genotipos quando comparados os niveis de carboidratos soluveis (CS) nas amostras originais, sendo que estes oscilaram de 2,20 a 17,22%. Ja os valores medios obtidos para pH das silagens variaram de 3,52 a 4,34. A avaliacao de qualidade final das silagens permitiu classifica-las como sendo de boa qualidade.

Resumo: Avaliou-se o potencial de uso de sacos de tecido com diferentes características e porosidades para acondicionamento de amostras em ensaios de produção de gases in vitro. Para isso, amostras de feno de capim coastcross (Cynodon sp.), secas em estufa de ventilação forçada e trituradas (1 mm) foram incubadas durante 96 horas em frascos de vidro com 160 mL de capacidade, contendo 72 mL de meio de cultura e 8 mL de inóculo ruminal. Foram avaliadas as silagens de quatro genótipos de girassol (Helianthus annus) (Rumbosol 91, Victoria 627, Victoria 807 e Mycogen 93338), pela técnica semiautomática de produção de gases. O delineamento experimental utilizado foi o de blocos ao acaso, em esquema de parcelas subdivididas, sendo as médias comparadas pelo teste SNK (p<0,05). O genótipo Rumbosol 91 apresentou a maior produção acumulativa de gases para o tempo de 96 horas de incubação com valor de 128,47mL/g de MS, e o menor valor foi observado para o genótipo Victoria 807 com 92,88. Não foram observadas diferenças entre os valores de degradabilidade da matéria seca (DMS) para as silagens avaliadas nos diferentes tempos.
