The introduction of CRISPR technologies has revolutionized strain engineering in filamentous fungi. However, its use in commercial applications has been hampered by concerns over intellectual property (IP) ownership, and there is a need for implementing Cas nucleases that are not limited by complex IP constraints. One promising candidate in this context is the Mad7 enzyme, and we here present a versatile Mad7-CRISPR vector-set that can be efficiently used for the genetic engineering of four different Aspergillus species: Aspergillus nidulans, A. niger, A. oryzae and A. campestris, the latter being a species that has never previously been genetically engineered. Mutations related to gene methG1 of Aspergillus nidulans were analyzed, in order to study a mathematical model for the determination of the mutation rate per nucleus per generation, in filamentous fungi. A replica plating technique was used to inoculate, in a single operation, 26 colonies of the strain, into Petri dishes containing culture medium, and the nine central colonies were analyzed for size and number of conidia in each colony. Using this technique, several central colonies were analyzed with regard to the appearance of mutation, and the number and type of reversions were determined for each colony. Anti dermatophytic therapy: prospects for the discovery of new drugs from natural products BJM

Millions of people and animals suffer from superficial infections caused by a group of highly specialized filamentous fungi, the dermatophytes, which only infect keratinized structures. With the appearance of AIDS, the incidence of dermatophytosis has increased. Current drug therapy used for these infections is often toxic, long-term, and expensive and has limited effectiveness; therefore, the discovery of new anti dermatophytic compounds is a necessity. Natural products have been the most productive source for new drug development. This paper provides a brief review of the current literature regarding the presence of dermatophytes in immunocompromised patients, drug resistance to conventional treatments and new anti dermatophytic treatments.

Air conditioned environments were sampled using a passive sedimentation technique, the air I ratio and the presence of aflatoxigenic strains evaluated for ANVISA. Abstract Filamentous fungi are widely diverse and ubiquitous organisms. Such biodiversity is barely known, making room for a great potential still to be discovered, especially in tropical environments - which are favorable to growth and species variety. Filamentous fungi are extensively applied to the production of industrial enzymes, such as the amylases. This class of enzymes acts in the hydrolysis of starch to glucose or maltooligosaccharides. Various microorganisms including bacteria, yeast and fungi can degrade caffeine. There are few publications about caffeine degradation pathway in filamentous fungi, mainly by solid-state fermentation (SSF). Studies were carried out on degradation of caffeine and their metabolites by filamentous fungi in SSF using coffee husk as substrate. The purpose of this work was to investigate the caffeine degradation pathway by Rhizopus delemar in packed bed column fermenter and to compare this degradation metabolism with glass flasks fermentation. Concentration determination of extracellular and intracellular red pigments produced by Monascus sp BABT

In this work red pigments production was evaluated (by spectrophotometry) in semi-synthetic medium by rotary shaker cultures, utilizing seven strains of Monascus sp. The strains isolated at LEB/DEQ/EPUSP had a absorbance average of extracellular red pigments higher than the others strains (13.0 and 9.6 U, respectively) and the specific production of extracellular red pigments was from 1.7 until 3.5 times higher than the specific production of intracellular ones.

This research study was conducted to evaluate, by means of microbiological analyses, the preservation of wet brewery waste stored under aerobic and anaerobic conditions, regarding the development of filamentous fungi, yeasts and lactic-acid bacteria. The following treatments were used: untreated brewery waste, pre-dried brewery waste silage, brewery waste silage with effluent drainage and BW silage without effluent drainage. Silos made of PVC and equipped with Bunsen valves to allow gases to escape were used. The present work reports factors affecting the production and regeneration of protoplasts from Colletotrichum lindemuthianum. The usefulness of protoplast isolation is relevant for many different applications and has been principally used in procedures involving genetic manipulation. Osmotic stabilizers, lytic enzymes, incubation time and mycelial age were evaluated in terms of their effects on protoplast yield. Filamentous fungi isolated from Brazilian semiarid tolerant to metallurgical industry wastes: an ex situ evaluation BABT

The purpose of this study was to assess the impact of metallurgical industry wastes on the semiarid soil microbiota using physico-chemical and microbiological parameters, highlighting the filamentous fungi assembly. Soil samples were collected in an area of industrial waste deposit contaminated with lead and mixed with natural soil (control soil) in seven different concentrations (0, 7.5, 15, 30, 45, 60 and 100%). The treatment of acute lymphoblastic leukemia is challenging due to side effects, efficacy of the available drugs, and costs. Utilization of L-asparaginase as a therapeutic agent is essential to increase survival of patients. However, costs are elevated and the bacterial forms of the enzyme cause reactions that result in its inhibition by the immune system. Therapeutics alternatives may be searched among eukaryote producers, like fungi. Twelve strains of filamentous fungi were evaluated regarding expression of L-asparaginase activity. Incidence and distribution of filamentous fungi during fermentation, drying and storage of coffee (Coffea arabica L.) beans BJM

The objective of this work was to isolate and characterize filamentous fungi present in different stages of harvest, fermentation, drying and storage of coffee beans processed by natural method. The cherries were hand-picked and then placed on a cement drying platform where they remained until reached 11% of humidity. Microbial counts were found in all samples during fermentation and drying of the coffee beans. Counts of fungi in the coffee cherries collected from the tree (time 0) were around 1.5 x 10³ CFU/g. This number increased slowly during the fermentation and drying reaching values of 2 x 10(5) CFU/g within 22 days of processing. Intra and extracellular nuclease production by strains of Aspergillus niger and Aspergillus nidulans was estimated using a modified DNAse test agar and cell-free extract assays. Differences in the production of nucleases by A. niger and A. nidulans were observed. These observations suggest that the DNAse test agar can be helpful for a quick screening for some types of nucleases in filamentous fungi. The assays using cell-free extracts can also be useful for initial characterization of other types of nucleases.
