A method to determine volatile contaminants in polyethylene terephthalate (PET) packages by HDC-GC-FID and its application to post-consumer materials Ciênc. Tecnol. Aliment.
Fabris,Samanta; Freire,Maria Teresa de Alvarenga; Wagner,Roger; Reyes,Felix Guillermo.
A simple and low cost method to determine volatile contaminants in post-consumer recycled PET flakes was developed and validated by Headspace Dynamic Concentration and Gas Chromatography-Flame Ionization Detection (HDC-GC-FID). The analytical parameters evaluated by using surrogates include: correlation coefficient, detection limit, quantification limit, accuracy, intra-assay precision, and inter-assay precision. In order to compare the efficiency of the proposed method to recognized automated techniques, post-consumer PET packaging samples collected in Brazil were used. GC-MS was used to confirm the identity of the substances identified in the PET packaging. Some of the identified contaminants were estimated in the post-consumer material at concentrations...

A real-time PCR genotyping assay to detect FAD2A SNPs in peanuts (Arachis hypogaea L.) Electron. J. Biotechnol.
Barkley,Noelle A; Wang,Ming Li; Pittman,Roy N.
The high oleic (C18:1) phenotype in peanuts has been previously demonstrated to result from a homozygous recessive genotype (ol1ol1ol2ol2) in two homeologous fatty acid desaturase genes (FAD2A and FAD2B) with two key SNPs. These mutant SNPs, specifically G448A in FAD2A and 442insA in FAD2B, significantly limit the normal function of the desaturase enzyme activity which converts oleic acid into linoleic acid by the addition of a second double bond in the hydrocarbon chain. Previously, a genotyping assay was developed to detect wild type and mutant alleles in FAD2B. A real-time PCR assay has now been developed to detect wild type and mutant alleles (G448A) in FAD2A using either seed or leaf tissue. This assay was demonstrated to be applicable for the...

A system to evaluate the performance of hydraulic nozzles used in stored grain preotection trials Scientia Agricola
Vásquez-Castro,Javier A.; Baptista,Gilberto C. de; Gadanha Jr.,Casimiro D.; Trevizan,Luiz R.P..
We developed a system to evaluate the performance of hydraulic nozzles used in stored corn and wheat grain protection experiments. An insecticidal mix was used as test fluid to determine the transversal volumetric distribution and droplets spectrum of a model TJ-60 8002EVS hydraulic nozzle. A mobile application system was built to apply a rate equivalent to 5 L t-1 and obtain theoretical concentrations of 10 and 0.5 mg kg-1 of fenitrothion and esfenvalerate, respectively. The corn and wheat grains were spread out as a fine layer. Three glass slides (0.1 × 0.05 m) were placed on the top surface of the grains to ensure that the intended application rate was achieved. After treatment, the deposits on both matrices were analyzed by gas chromatography. The...

Accumulation and depuration of hydrocarbons in the mangrove oyster Crassostrea rhizophorae ArchiMer
Delcastillo, C; Corredor, J; Morell, J.
Adult specimens of Crassostrea rhizophorae were exposed to diesel oil dispersed in seawater. The experiment consisted of a 10 days contamination period and 15 days depuration period. Oyster samples were extracted, and analysed by gas chromatography and UV fluorescence spectroscopy. The concentrations of n-alkanes, branched alkanes, unresolved complex mixture (UCM) and aromatic hydrocarbons were determined. The oysters accumulated the hydrocarbons rapidly, reaching a maximum concentration of 387.5 ug/g (wet weight). After exposure to uncontaminated seawater, the oysters were able to reduce their hydrocarbon load to 5.6 ug/g. The data indicates that C. rhizophorae is capable of preferential accumulation and depuration of hydrocarbons. Apparently, the oysters...

Comparison of the quantification of caffeine in human plasma by gas chromatography and ELISA BJMBR
Carregaro,A.B.; Woods,W.E.; Tobin,T.; Queiroz-Neto,A..
In the present study we evaluated the precision of the ELISA method to quantify caffeine in human plasma and compared the results with those obtained by gas chromatography. A total of 58 samples were analyzed by gas chromatography using a nitrogen-phosphorus detector and routine techniques. For the ELISA test, the samples were diluted to obtain a concentration corresponding to 50% of the absorbance of the standard curve. To determine whether the proximity between the I50 of the standard curve and that of the sample would bring about a more precise result, the samples were divided into three blocks according to the criterion of difference, in modulus, of the I50 of the standard curve and of the I50 of the sample. The samples were classified into three...

Convenient preparation of picolinyl derivatives from fatty acid esters ArchiMer
Dubois, Nolwenn; Barthomeuf, Chantal; Berge, Jean-pascal.
It is essential to have simple rapid methods for the determination of fatty acid structures. Traditionally, fatty acids are analysed by gas chromatography using their methyl ester derivatives (FAME). However, their corresponding mass spectra exhibit molecular ions but are usually devoid of ions indicative of structural features and, notably, the position of double bounds on the aliphatic chains [1]. In the most useful approach to structure determination, the carboxyl group is derivatised with a reagent containing a nitrogen atom. Recently, a convenient method for preparing picolinyl esters from intact lipids has been published [2]. However, some problems occurred in our laboratory when this method was used, leading to some modifications and optimisation....

Docosahexaenoic acid ethyl esther (DHAEE) microcapsule production by spray-drying: optimization by experimental design Ciênc. Tecnol. Aliment.
Rodrigues,Rodney Alexandre Ferreira; Rodrigues,Marili Villa Nova; Oliveira,Thais Isabel Vaccaro; Bueno,Cecilia Zorzi; Oliveira SOUZA,Ilza Maria de; Sartoratto,Adilson; Foglio,Mary Ann.
Docosahexaenoic acid is an essential polyunsaturated fatty acid with important metabolic activities. Its conjugated double bonds make it susceptible to decomposition. Its stability may be improved through fatty acid entrapment with a spray-drying technique; however, the many parameters involved in this technique must be considered to avoid affecting the final product quality. Therefore, this study aimed to evaluate the entrapment conditions and yields of fish oil enriched with docosahexaenoic acid ethyl ester. Microcapsules were obtained from Acacia gum using a spray-drying technique. The experimental samples were analyzed by chromatography and delineated by Statistica software, which found the following optimum entrapment conditions: an inlet temperature...

Evaluation of volatile profiles obtained for minimally-processed pineapple fruit samples during storage by headspace-solid phase microextraction gas chromatography-mass spectrometry Ciênc. Tecnol. Aliment.
TURAZZI,Francielle Crocetta; MORÉS,Lucas; MERIB,Josias; CARASEK,Eduardo; NARAIN,Narendra; LIMA,Lucélia Kátia de; NUNES,Maria Lúcia.
Abstract This paper describes the application of the solid-phase microextraction (SPME) technique for the determination and monitoring of the volatile profile of minimally-processed pineapple fruit stored at various temperatures (-12 °C, 4 °C and 25 °C) for different periods (1, 4 and 10 days). The SPME fiber coating composed of Car/PDMS presented the best performance. The optimal extraction conditions obtained through a Doehlert design were 60 min at 35 °C. The profiles for the volatile compounds content of the fruit at each stage of storage were determined by gas chromatography-mass spectrometry (GC-MS). The variation in the volatile profile over time was greater when the fruit samples were stored at 25 °C and at -12 °C compared to 4 °C. Thus, according...

Fenitrothion and esfenvalerate stability during corn and wheat sample processing Scientia Agricola
Vásquez-Castro,Javier Alberto; Baptista,Gilberto Casadei de; Trevizan,Luiz Roberto Pimentel; Gadanha Junior,Casimiro Dias.
The presence of insecticide residues in cereals represents a risk for the consumer, because these substances are used in large scale to protect stored grains from the attack of pests. The effects of three processing methods for corn and wheat samples on the stability of stored-grain protective insecticides were here evaluated. Fenitrothion and esfenvalerate were applied so as to produce theoretical concentrations of 10 and 0.5 mg kg-1, respectively. Two hours after treatment, the grains were processed and deposition was analyzed by gas chromatography. Grain species did not influence insecticide stability. This stability was only dependent upon the processing method and insecticide type. Grains processed together with dry ice provided the greatest...

Molecular and morphological characterization of hydrochar produced by microwave-assisted hydrothermal carbonization of cellulose PAB
Guiotoku,Marcela; Hansel,Fabricio Augusto; Novotny,Etelvino Henrique; Maia,Claudia Maria Branco de Freitas.
The objective of this work was to characterize the morphology and molecular composition of the hydrochar produced by microwave-assisted hydrothermal carbonization of cellulose. The produced hydrochar consists mainly of aggregate microspheres with about 2.0 µm in diameter, with aliphatic and aromatic structures and the presence of carbonyl functional groups. The aromatic groups are formed mainly by benzofuran-like structures, being chemically different from common cellulose char. Microwave-assisted hydrothermal carbonization yields a functionalized carbon-rich material similar to that produced by the conventional hydrothermal process.
