Agronomical, genetical and developmental characterization of fs6.4: a Quantitative Trait Locus controlling melon fruit shape National Institute of Agronomic Research

The genetic control of melon fruit shape is complex. Recently, several Quantitative Trait Loci (QTLs) affecting melon fruit shape have been identified using an introgression line population derived from the cross between a Cucumis melo ‘Piel de Sapo’ cultivar and the Korean accession PI 161375 ‘Songwan Charmi’ (SC). One of these QTLs, fs6.4, was selected for further genetical and developmental characterization. The SC allele of fs6.4 induces round fruit by limiting the longitudinal development of the fruit. Its mode of gene action is largely additive, showing low interactions with environment, genetic background or when combined with other fruit shape QTLs. The efficiency of simulated annealing algorithms and rapid chain delineation in establishing the best linkage order, when constructing genetic maps, was evaluated. Linkage refers to the phenomenon by which two or more genes, or even more molecular markers, can be present in the same chromosome or linkage group. In order to evaluate the capacity of algorithms, four F2 co-dominant populations, 50, 100, 200 and 1000 in size, were simulated. For each population, a genome with four linkage groups (100 cM) was generated. Construction of a fosmid library of cucumber (Cucumis sativus) and comparative analyses of the eIF4E and eIF(iso)4E regions from cucumber and melon (Cucumis melo) National Institute of Agronomic Research

We undertook comparative sequence analyses to assess synteny between the cucumber and melon genomes. A fosmid library of cucumber was synthesized and end sequencing of random fosmids produced over 680 kilobases, of which 25 % was similar to ribosomal DNAs, 23 % to satellite sequences, 22 % to coding regions in other plants, 4 % to transposable elements, 12 % to mitochondrial and chloroplast sequences, and 13 % showed no hits to the databases. The relatively high frequencies of ribosomal and satellite DNAs are consistent with previous analyses of cucumber DNA. The objective of this work was to verify the existence of a lethal locus in a eucalyptus hybrid population, and to quantify the segregation distortion in the linkage group 3 of the Eucalyptus genome. A E. grandis x E. urophylla hybrid population, which segregates for rust resistance, was genotyped with 19 microsatellite markers belonging to linkage group 3 of the Eucalyptus genome. Genetic linkage maps consist of ordering molecular markers across the genome and require a high number of markers for a good coverage of the genome. Such maps represent a framework which enables the identification and localisation of Quantitative Trait Loci (QTLs) for traits of interest, such as growth or disease resistance, with the final aim of achieving genetic improvement through marker-assisted selection (MAS).
