Effect of a submaxillary gland extract on Ehrlich tumor growth in mice BJMBR
Weill,P.; Frussa-Filho,R.; Bonamin,L.V..
Ablation of host submaxillary glands modifies Ehrlich tumor growth and tumor-infiltrating leukocytes, possibly by modifications in the serum level of growth factors produced by this gland. To extend this research, 7-month-old male EPM-1 mice (N = 30) were divided into two groups: 1) inoculated with tumor cells previously incubated with submaxillary salivary gland extract (SGE) in PBS for 30 min at 37%; 2) inoculated with tumor cells previously incubated with PBS, under the same conditions. Animals were inoculated into the footpad with 40 µl of a suspension containing 4.5 x 107 tumor cells/ml, and footpad thickness was measured daily for 10 days. Sections and smears of tumor cells were prepared from the tumor mass to determine mitosis frequency, percent of...

Glial fibrillary acidic protein (GFAP): modulation by growth factors and its implication in astrocyte differentiation BJMBR
Gomes,F.C.A.; Paulin,D.; Moura Neto,V..
Intermediate filament (IF) proteins constitute an extremely large multigene family of developmentally and tissue-regulated cytoskeleton proteins abundant in most vertebrate cell types. Astrocyte precursors of the CNS usually express vimentin as the major IF. Astrocyte maturation is followed by a switch between vimentin and glial fibrillary acidic protein (GFAP) expression, with the latter being recognized as an astrocyte maturation marker. Levels of GFAP are regulated under developmental and pathological conditions. Upregulation of GFAP expression is one of the main characteristics of the astrocytic reaction commonly observed after CNS lesion. In this way, studies on GFAP regulation have been shown to be useful to understand not only brain physiology but...

Heparan sulfate and cell division BJMBR
Porcionatto,M.A.; Nader,H.B.; Dietrich,C.P..
Heparan sulfate is a component of vertebrate and invertebrate tissues which appears during the cytodifferentiation stage of embryonic development. Its structure varies according to the tissue and species of origin and is modified during neoplastic transformation. Several lines of experimental evidence suggest that heparan sulfate plays a role in cellular recognition, cellular adhesion and growth control. Heparan sulfate can participate in the process of cell division in two distinct ways, either as a positive or negative modulator of cellular proliferation, or as a response to a mitogenic stimulus.

Heparan sulfate proteoglycans: structure, protein interactions and cell signaling Anais da ABC (AABC)
Dreyfuss,Juliana L.; Regatieri,Caio V.; Jarrouge,Thais R.; Cavalheiro,Renan P.; Sampaio,Lucia O.; Nader,Helena B..
Heparan sulfate proteoglycans are ubiquitously found at the cell surface and extracellular matrix in all the animal species. This review will focus on the structural characteristics of the heparan sulfate proteoglycans related to protein interactions leading to cell signaling. The heparan sulfate chains due to their vast structural diversity are able to bind and interact with a wide variety of proteins, such as growth factors, chemokines, morphogens, extracellular matrix components, enzymes, among others. There is a specificity directing the interactions of heparan sulfates and target proteins, regarding both the fine structure of the polysaccharide chain as well precise protein motifs. Heparan sulfates play a role in cellular signaling either as receptor...

Immunolocalization of HB-EGF in Human Skin by Streptavidin-Peroxidase (HRP) Conjugate Method International Journal of Morphology
Castrogiovanni,P; Mazzone,V; Imbesi,R.
EGF family growth factors consists of growth factors, such as transforming growth factor (TGF)-a, heparin-binding EGF-like growth factor (HB-EGF), amphiregulin (AR) and epiregulin, autocrine growth factors in normal human keratinocytes. HB-EGF is mitogen for epithelial cells and like other members of the EGF family, HB-EGF exerts its biological effects via interaction with the EGF receptor (EGFR). HB-EGF is an autocrine growth factor for human keratinocytes, and has a possible role as a paracrine growth factor for fibroblast. Our report concerning immunohistochemical localization of HB-EGF in normal skin by using the streptavidin-peroxidase (HRP) conjugate method, confirms previous data, revealing specific patterns of HB-EGF localization. Identification of...

Impact of cigarette smoke and aerobic physical training on histological and molecular markers of prostate health in rats BJMBR
Veras,A.S.C.; Baptista,D.B.; dos Santos,N.J.; Thorpe,H.H.A.; Seraphim,P.M.; Florido Neto,A.R.; Teixeira,G.R..
Recent evidence suggests that aerobic physical training may attenuate the deleterious effects of cancer risk factors, including smoking. We investigated the effects of cigarette smoke inhalation and aerobic physical training on the expression of steroid receptors and inflammatory and apoptotic proteins in the prostate. Forty male Wistar rats were distributed in four groups: control (CO), exercise (EXE), cigarette smoke exposure (CS), and cigarette smoke exposure with exercise (CS+EXE). For eight weeks, animals were repeatedly exposed to cigarette smoke for 30 min or performed aerobic physical training either with or without the cigarette smoke inhalation protocol. Following these experiments, we analyzed prostate epithelial morphology and prostatic...

Neoangiogenesis Induced by Progenitor Endothelial Cells: Effect of Fucoidan from Marine Algae ArchiMer
Boisson-vidal, C; Zemani, F; Calliguiri, G; Galy-fauroux, I; Colliec-jouault, Sylvia; Helley, D; Fischer, A-m.
Fucoidans -- sulphated polysaccharides extracted from brown algae - could be beneficial in patients with ischemic diseases. Their antithrombotic and proangiogenic properties promote in animals, neovascularization and angiogenesis which prevent necrosis of ischemic tissue. In 1997, endothelial progenitor cells were first identified in human peripheral blood. They are recruited from bone marrow and contribute to neovascularization after ischemic injury. Mobilization of these cells in ischemic sites is an important step in new vessel formation. It is thought that the progenitors interact with endothelial cells, then extravasate and reach ischemic sites, where they proliferate and differentiate into new blood vessels. Although chemokines, cytokines and...

Osteogenic molecules for clinical applications: improving the BMP-collagen system Biol. Res.
Arrabal,Pilar M; Visser,Rick; Santos-Ruiz,Leonor; Becerra,José; Cifuentes,Manuel.
Among the osteogenic growth factors used for bone tissue engineering, bone morphogenetic proteins (BMPs) are the most extensively studied for use in orthopaedic surgery. BMP-2 and BMP-7 have been widely investigated for developing therapeutic strategies and are the only two approved for use in several clinical applications. Due to the chemical and biological characteristics of these molecules, their authorised uses are always in combination with a carrier based on collagen type I. Although the use of these growth factors is considered safe in the short term, the very high doses needed to obtain significant osteoinduction make these treatments expensive and their long-term safety uncertain, since they are highly pleiotropic and have the capacity to induce...

Sombreamento para controle da Antracnose na produção de mudas de açaí-solteiro. Infoteca-e
NOGUEIRA, S. R.; ANDRADE NETO, R. de C.; LUNZ, A. M. P..
O açaí-solteiro (Euterpe precatoria Mart.)

Specific structural features of syndecans and heparan sulfate chains are needed for cell signaling BJMBR
Lopes,C.C.; Dietrich,C.P.; Nader,H.B..
The syndecans, heparan sulfate proteoglycans, are abundant molecules associated with the cell surface and extracellular matrix and consist of a protein core to which heparan sulfate chains are covalently attached. Each of the syndecan core proteins has a short cytoplasmic domain that binds cytosolic regulatory factors. The syndecans also contain highly conserved transmembrane domains and extracellular domains for which important activities are becoming known. These protein domains locate the syndecan on cell surface sites during development and tumor formation where they interact with other receptors to regulate signaling and cytoskeletal organization. The functions of cell surface heparan sulfate proteoglycan have been centered on the role of heparan...

Temporal expression pattern of the insulin-like growth factor II and fibroblast growth factor transcripts in avian embryogenesis BABT
Gabriel,Jane Eyre; Alvares,Lúcia Elvira; Coutinho,Luiz Lehmann.
In this study, the abundance of IGF-II and bFGF transcripts was estimated in the chicken embryos using the competitive RT-PCR analysis. Significant enhancements in the abundance of IGF-II mRNA were observed at stages HH1 and 5, and a new accumulation in these levels was observed at stage HH18 in comparison to the basal levels. The abundance of bFGF mRNA increased significantly at stages HH18 and 20, followed by an upregulation in the expression of these transcripts at stage HH26. These findings provided important information about the temporal expression pattern of IGF-II and bFGF transcripts in the whole chicken embryos during in ovo development.

The specificity of interactions between proteins and sulfated polysaccharides Anais da ABC (AABC)
Mulloy,Barbara.
Sulfated polysaccharides are capable of binding with proteins at several levels of specificity. As highly acidic macromolecules, they can bind non-specifically to any basic patch on a protein surface at low ionic strength, and such interactions are not likely to be physiologically significant. On the other hand, several systems have been identified in which very specific substructures of sulfated polysaccharides confer high affinity for particular proteins; the best-known example of this is the pentasaccharide in heparin with high affinity for antithrombin, but other examples may be taken from the study of marine invertebrates: the importance of the fine structure of dermatan sulfate (DS) to its interaction with heparin cofactor II (HCII), and the...
