A New Photobioreactor for Continuous Microalgal Production in Hatcheries Based on External-Loop Airlift and Swirling Flow ArchiMer
Loubiere, K; Olivo, Erell; Bougaran, Gael; Pruvost, J; Robert, Rene; Legrand, J.
This study deals with the scale of a new photobioreactor for continuous microalgal production in hatcheries. The combination of the state-of-art with the constraints inherent to hatcheries has turned the design into a closed, artificially illuminated and external-loop airlift configuration based on a succession of elementary modules, each one being composed of two transparent vertical interconnected columns. The liquid circulation is ensured pneumatically (air injections) with respect to a swirling motion (tangential inlets). A single module of the whole photobioreactor was built-up to scale its geometry (diameter and length) and to optimize its design (air sparger, tangential inlets). The volumetric productivities were predicted by modeling radiative...

Action of Antimicrobial Copper on Bacteria and Fungi Isolated from Commercial Poultry Hatcheries Rev. Bras. Ciênc. Avic.
Depner,RFR; Pontin,KP; Depner,RA; Flores Neto,A; Lucca,V; Lovato,M.
ABSTRACT Since 2008, when the US Environmental Protection Agency (EPA) registered copper and its alloys as an antimicrobial agent for contact surfaces, research has demonstrated their antimicrobial activity. The aim of this study was to evaluate the efficacy of antimicrobial copper against bacteria and fungi isolated from commercial poultry hatcheries in order to develop a microbiological control alternative in these environments. Samples were collected from the surfaces of hatcher baskets from two hatcheries. Mesophilic microorganisms and fungi/yeasts were isolated and standardized in concentration of 105 cells/mL. Four copper plates and four stainless steel plates were completely immersed for one minute in bacteria and fungi/yeasts solutions and left to...

in mollusc hatcheries. Consequently, we assessed themost commonly used species to establish their growth and size profiles, their cytomorphological, biochemical, biomolecular identification as well as their cytometer signs, and also their ability to grow under extreme conditions of temperature and salinity. This work aimed at matching the conformity of the strains used in Argenton (dispatched to most of the French commercial hatcheries) to those of a referenced culture collection. A previous screening preceded this study, because of the 45 species and/or clones held originally, Argenton kept no more than 15 species. As most of the latter are also represented in referenced collection, our strains are therefore correctly identified. On the other hand, the...

Analysis of hemocyte parameters in Pacific oysters, Crassostrea gigas, reared in the field — Comparison of hatchery diploids and diploids from natural beds ArchiMer
Gagnaire, Beatrice; Soletchnik, Patrick; Faury, Nicole; Kerdudou, Nolwenn; Le Moine, Olivier; Renault, Tristan.
Internal defence mechanisms of bivalves include the hemocytes which assume several functions. Relationships exist between environmental factors, physiological state of oysters, defence system and pathogens. Hatchery oysters are more and more produced and reared in the field. The characteristics and particularly the defence system of these animals are poorly documented. In this context, two field trials were carried out from May to September 2002 and from April to July 2003 in order to monitor hemocyte parameters of diploid oysters from natural beds and hatchery-produced diploid oysters using flow cytometry. Several hemocyte parameters (cell mortality, granulocyte percentage, phagocytosis, esterase and peroxidase activities) were monitored. For both oyster...

Bivalve hatchery technology: The current situation for the Pacific oyster Crassostrea gigas and the scallop Pecten maximus in France ArchiMer
Robert, Rene; Gerard, Andre.
The French oyster farming industry relies almost exclusively on juveniles collected from the natural environment; the supply of spat produced by hatcheries is low, about 10% of the industry's requirements. Development through selective breeding of oyster stocks which are better suited for aquaculture purposes, is likely to reverse this tendency since only hatcheries will be able to supply such animals. Scallop farming, which at present is poorly developed in France, relies exclusively on hatchery produced spat. Although hatchery technology is constantly being improved, significant production problems remain which must be solved before hatcheries become a major supplier of juveniles for the industry. This paper describes the present state of hatchery...

Comparison of growth, digestive system maturation and skeletal development in sea bass larvae reared in an intensive or a mesocosm system ArchiMer
Zouiten, Dora; Ben Khemis, Ines; Masmoudi, Ahmed Slaheddin; Huelvan, Christine; Cahu, Chantal.
The quality of development in intensive or mesocosm hatchery-reared Dicentrarchus labrax larvae was investigated using physiological indicators assessing ontogeny. Larvae were reared in intensive (120 L tanks) and in mesocosm systems (20 m(3) enclosures) with the same feeding sequence, excluding the wild zooplankton naturally available in mesocosms. Faster growth was recorded since early development [16 day after hatching (DAH)] in the mesocosm. Maturation of the digestive system also occurred earlier as indicated by the higher amylase secretion ratios, the intestinal maturation index (alkaline phosphatase/leucine-alanine peptidase and aminopeptidase-N/leucine-alanine peptidase ratios) and the more developed intestinal epithelium at 23 DAH. Nevertheless,...

Conception et étude d'un photobioréacteur pour la production en continu de microalgues en écloseries aquacoles ArchiMer
Olivo, Erell.
This thesis describes the conception and the study of a photobioreactor dedicated to aquaculture. The state of the art and the technico-economic constraints proper to this activity led to the choices to a simple reactor, volume variable, cheap to produce and reliable for continuous cultures. The elementary module is made of two vertical columns, connected by flanges. An airlift injection located at the base of the column (airlift reactor) generates a swirling flow circulation in order to minimize the biofilm development on the inner side of the column. A pilot of 6.1 liters has been conceived to study the radiative transfers, the hydrodynamics and the biological cultures in continuous mode. Coupling a radiative transfer model (two flux model) with the...

Conséquences génétiques de la production de larves d'huîtres en écloserie : étude des processus de dérive et de sélection ArchiMer
Taris, Nicolas; Sauvage, Christopher; Batista, Frederico; Baron, Sophie; Ernande, Bruno; Haffray, Pierrick; Boudry, Pierre.
Previous studies have shown heritable variation in larval developmental traits in the Pacific oyster Crassostrea gigas. In order to study the genetic consequences of production of oyster larvae in hatcheries, two factors, specific to hatcheries, were examined: the effect of discarding the smallest larvae (i.e. culling) and the effect of temperature (20°C versus 26°C). A mixed-family approach was used in order to infer the genetic composition of larval populations and family assignment, limiting possible environmental bias and allowing the study of a relatively large number of families using a limited number of larval tanks. Our results show that three multiplexed highly polymorphic microsatellite markers are a powerful tool for family assignment and,...

Conséquences génétiques de la production intensive de larves d'huîtres en écloserie : étude des processus de dérive et de sélection liés aux pratiques d'élevage. ArchiMer
Taris, Nicolas.
In order to study the genetic influence of intensive production of the larval stage in commercial hatcheries, two types of specific selective processes were examined: the effects of discarding the smallest larvae and temperature. A mixed-family approach was used in order to infer the genetic composition of the larvae. The results show that our mixed-family approach, combined with microsatellite-based family assignment, is a powerful tool for the study of bivalve larvae genetics. Selective sieving is an advantageous practice at a phenotypic scale, but also represents a substantial risk for diversity loss. The culled larvae are important for the minimization of the variability in reproductive success and the maximization of broodstock genetic diversity....

Cryopreservation of Pacific oyster ( Crassostrea gigas ) larvae: Revisiting the practical limitations and scaling up the procedure for application to hatchery ArchiMer
Labbe, Catherine; Haffray, Pierrick; Mingant, Christian; Quittet, Benjamin; Diss, Blandine; Tervit, H. Robin; Adams, Serean L.; Rimond, Flore; Suquet, Marc.
Pacific oyster Crassostrea gigas is one major species for aquaculture, and the development of breeding programs and the need for preservation of wild stock genetic resources prompted the need for larvae cryopreservation. The objective of the present study was to choose the most reliable protocol from several existing publications, to test its biological and practical limitations, and to adapt it to hatchery conditions. The selected protocol was characterized by a very slow freezing rate without seeding, and by the use of ethylene glycol and sucrose as cryoprotectant. The best survivals after thawing and rearing up to 48 h post fertilization (hpf) were obtained with larvae that were frozen at late trochophore (20 hpf) and early-D (24 hpf) stages. Increasing...

Effect of the probiotic strain Phaeobacter gallaeciensis after bacterial challenge on the complete larval development of Pecten maximus ArchiMer
Genard, Bertrand; Larouche, Olivier; Nicolas, Jean-louis; Miner, Philippe; Beaudin, Marie-lou; Tremblay, Rejean.
The aim of this project was to evaluate the impact of probiotic (Phaeobacter gallaeciensis, X34 strain) treatment on the complete development (from veliger to metamorphosis) of Pecten maximus larvae in the context of a bacterial challenge and in conditions more representative of hatchery practices. To that effect, the present study was divided into two main steps. In the first, we used in vitro analyses (antibiograms and microplate assays) to validate the inhibition abilities of X34 on the growth of four Vibrio pathogen species. During the second step, we added pathogens (Vibrio pectenicida) into rearing tanks after two weeks of pre-treatment with the probiotic and then followed the larval development of Pecten maximus through the monitoring of survival...

Epigenomic modifications induced by hatchery rearing persist in germ line cells of adult salmon after their oceanic migration ArchiMer
Leitwein, Maeva; Laporte, Martin; Le Luyer, Jeremy; Mohns, Kayla; Normandeau, Eric; Withler, Ruth; Bernatchez, Louis.
Human activities induce direct or indirect selection pressure on natural population and may ultimately affect population’s integrity. While numerous conservation programs aimed to minimize human‐induced genomic variation, human‐induced environmental variation may generate epigenomic variation potentially affecting fitness through phenotypic modifications. Major questions remain pertaining to how much epigenomic variation arises from environmental heterogeneity, whether this variation can persist throughout life, and whether it can be transmitted across generations. We performed whole genome bisulfite sequencing (WGBS) on the sperm of genetically indistinguishable hatchery and wild born migrating adults of Coho salmon (Oncorhynchus kisutch) from two...

This study has two main objectives: (1) to implement a recycling aquaculture system (RS) for the larvae of the oyster Crassostrea gigas, and (2) to characterise the bacterial communities established in different compartments of this system. An RS with 25% fresh seawater addition per hour and another with no addition (0%) were compared with a flow-through system (FT). Larval survival was equivalent in RS and FT, but growth rate was 17% slower in RS than in FT. The physical chemical parameters remained stable, except for pH that decreased to 7.75 and salinity that increased to 37.5‰ in the RS 0%. In both systems, the cultivable bacteria were present in similar numbers in seawater (around 105 ml− 1) and in larvae (103 larva− 1) on day 15. Bacterial...

Estimating the impact of environmental conditions on hatching results using multivariable analysis Rev. Bras. Ciênc. Avic.
Nääs,IA; Gigli,ACS; Baracho,MS; Almeida Paz,ICL; Salgado,DD.
Hatching results are directly related to environmental and biological surroundings. This research study aimed at evaluating the influence of incubation environmental conditions on hatchability and one-day-old chickling quality of five production flocks using multivariable analysis tool. The experiment was carried out in a commercial hatchery located in the state of São Paulo, Brazil. Environmental variables such as dry bulb temperature, relative humidity, carbon dioxide concentration, and number of colony forming units of fungi were recorded inside a broiler multi-stage setter, a hatcher after eggs transference, and a chick-processing room. The homogeneity of parameter distribution among quadrants inside the setter, the hatcher, and the chick room was...

Etudes sur les mortalites larvaires de la coquille st Jacques Pecten maximus en ecloserie ArchiMer
Robert, Rene; Miner, Philippe; Nicolas, Jean-louis; Mazuret, Michel; Connan, Jean-paul.
Larval mortalities occurring in molluscan hatcheries have often been associated with bacterial contamination. Although batches of oyster and clam larvae have been routinely reared in the hatchery of Argenton without antibiotics, high larval mortalities have been recorded with Pecten maxirnus under similar conditions. For this species, an addition of chloramphenicol was found necessary. However, this chemical has now been banned in Europe. Thus either substitition products or an improvement in the rearing procedures is essential. Studies carried out have shown that neither a decrease in larval density (1 larvae. ml"l) nor sea water change frequency (1 per day) had any positive effects. Furthermore, elective substances such as sugars were not suitable and...
