ABSTRACT The agro-industrial waste deposited in the environment causes problems in nature that can be solved with the use and generation of bioproducts. Thus, the objective was to study the lignocellulosic fraction of cashew (Anacardium occidentale L.) peduncle bagasse and fermentation on large scale (8-16 times) using the strain Candida guilliermondii CCT-3544 as production agent. According to the obtained results, it can be noted that the dry cashew peduncle bagasse has 21.45% of cellulose, 10.96% of hemicellulose and 35.39% of lignin. During fermentation, C. guilliermondii 3544-CAT was able to grow on medium containing hydrolysate, with maximum cell growth concentration of 3.5 g L-1. The behavior of the sugars in the fermentation process was similar in... |

UDP-glucose dehydrogenase (UGDH) catalyzes the oxidation of UDP-glucose (UDP-Glc) to UDP-glucuronate (UDP-GlcA), a key sugar nucleotide involved in the biosynthesis of plant cell wall polysaccharides. A full-length cDNA fragment coding for UGDH was cloned from the cambial region of 6-month-old E. grandis saplings by RT-PCR. The 1443-bp-ORF encodes a protein of 480 amino acids with a predicted molecular weight of 53 kDa. The recombinant protein expressed in Escherichia coli catalyzed the conversion of UDP-Glc to UDP-GlcA, confirming that the cloned cDNA encodes UGDH. The deduced amino acid sequence of the cDNA showed a high degree of identity with UGDH from several plant species. The Southern blot assay indicated that more than one copy of UGDH is present... |

Abstract The filter cake from sugar cane processing is rich in organic matter and nutrients, which favors the proliferation of microorganisms with potential to deconstruct plant biomass. From the metagenomic data of this material, we assembled a draft genome that was phylogenetically related to Thermomonospora curvata DSM 43183, which shows the functional and ecological importance of this bacterium in the filter cake. Thermomonospora is a gram-positive bacterium that produces cellulases in compost, and it can survive temperatures of 60 ºC. We identified a complete set of biomass depolymerizing enzymes in the draft genome of Thermomonospora sp. CIT 1, such as α-amylase, catalase-peroxidases, β-mannanase, and arabinanase, demonstrating the potential of this... |

The foldable, low thermal expansion, and optically transparent are main properties of paper produced with cellulose nanofibers (nanopaper), being able to be used ifl the development of flexible circuit technologies. In this context, this work evaluates the importance of hemicellulose in the properties of nanofiber paper (NFP). The hemicellulose extraction from Pinus sp bleached cellulose pulp was performed with NaOH 17.5% for 1.5 h. Thereafter, the pulp was washed with 8.3% NaOH, then was add 10% acetic acid until neutralization, and washed with water. The nanofibrils were obtained by mechanical defibrillation in a Super Masscoloider Masuko Sangyo mill. The NFP with the grammage of 40 and 50 g/m2 were produced by filtration and drying under pressure. The... |

This research aimed to characterize the tolerance to flooding and alterations in pectic and hemicellulose fractions from mesocotyl of maize tolerant to flooding when submitted to hypoxia. In order to characterize tolerance seeds from maize cultivars Saracura BRS-4154 and BR 107 tolerant and sensitive to low oxygen levels, respectively, were set to germinate. Plantlet survival was evaluated during five days after having been submitted to hypoxia. After fractionation with ammonium oxalate 0.5% (w/v) and KOH 2M and 4M, Saracura BRS-4154 cell wall was obtained from mesocotyl segments with different damage intensities caused by oxygen deficiency exposure. The cell wall fractions were analyzed by gel filtration and gas chromatography, and also by Infrared... |

The endo-?-mannanase acts on the hemicellulose fraction of the endosperm cell walls, mainly mannans and galactomannans. This process weakens cell walls and allows radicle protrusion during seed germination, but may also occur during the deterioration process. Thus, the aim of this research was to determine the activity of endo-?-mannanase enzyme in dry coffee seeds and in soaked seeds, evaluating its relationship between physiological qualities. Coffee seeds obtained by different processing methods (natural, fermented and demucilated) and drying (sun, shade and dryer) were used. Seed quality was evaluated by germination and tetrazolium tests, and the endo-?-mannanase enzyme activity was determined in dry seeds and after 10 days of soaking. From the... |

Corn fiber (CF) is an agroindustrial residue obtained from starch or corn syrup. Currently, there has been increasing research interest in lignocellulosic materials from these residual materials. Consequently, this paper is a novel approach that complements the structural and chemical characterizations already published in the literature for CF. In this study, isolation, chemical composition and characterization was conducted of hemicellulose by means of 1H NMR and FTIR-ATR and Klason lignin by means of acid soluble lignin (ASL) and acid insoluble lignin AIL) determination using UV spectrophotometry and Py?GC?MS. Hemicellulose was obtained after aqueous extraction, lipid removal, delignification and alkaline extraction. The isolated hemicellulose from CF... |
