Agrowaste-based Polyhydroxyalkanoate (PHA) production using hydrolytic potential of Bacillus thuringiensis IAM 12077 BABT

The study identified the innate enzymatic potential (amylase) of the PHB producing strain B.thuringiensis IAM 12077 and explored the same for cost-effective production of PHB using agrowastes, eliminating the need for pretreatment (acid hydrolysis and/or commercial enzyme). Comparative polyhydroxyalkanoate (PHA) production by B. thuringiensis IAM 12077 in biphasic growth conditions using glucose and starch showed appreciable levels of growth (5.7 and 6.8 g/L) and PHA production (58.5 and 41.5%) with a PHA yield of 3.3 and 2.8 g/L, respectively. Nitrogen deficiency supported maximum PHA yield (2.46 g/L) and accumulation (53.3%). Anti-fungal potential of ozone against some dermatophytes BJM

ABSTRACT Dermatophytes are classified in three genera, Epidermophyton, Microsporum and Trichophyton. They have the capacity to invade keratinized tissue to produce a cutaneous infection known as dermatophytoses. This investigation was performed to study the effect of gaseous ozone and ozonized oil on three specific properties of six different dermatophytes. These properties included sporulation, mycelia leakage of sugar and nutrients and the activity of their hydrolytic enzymes. Generally, ozonized oil was found to be more efficacious than gaseous ozone. ABSTRACT Pre-harvest sprouting is one of the most important problems of the wheat crop and it is necessary to find alternative uses for the grain because millers reject the damaged wheat. In the field these grains were stimulated to germinate, due to weather conditions and their metabolic activity was enhanced, the hydrolytic enzymes promoted changes in the proteins and reserve carbohydrates, which reflect on the flour quality. Similar to malting germination has started, but with different intensity and under different conditions. Production of hydrolytic enzymes by the plant pathogenic fungus Myrothecium verrucaria in submerged cultures BJM

The capability of the plant pathogenic fungus Myrothecium verrucaria to produce extracellular hydrolytic enzymes in submerged cultures was studied using several substrates. The fungus was able to produce different depolymerases and glycosidases, being xylanase, pectinase and protease the most important. Lipase was found in cultures developed in the presence of olive oil, while protease activity was detected in all cultures. Xylanase and pectinase were optimally active at pH 4.5-5.5, while protease was active in a large range of pH 3.5 to 11.0. All three enzymes were maximally active at 40ºC and they were stable for several hours at temperature up to 50ºC.

Selection of cellulolytic fungi isolated from diverse substrates BABT

The aim of the present work was to select filamentous fungi isolated from diverse substrates to obtain the strains with potential to produce the hydrolytic enzymes. From a total of 215 strains, seven strains from the soils, six from the plants and one from sugarcane bagasse were selected and identified as belonging to the Trichoderma, Penicillium and Aspergillus genera. The best hydrolytic activities obtained by semi-solid fermentation using these strains were approximately: 35; 1; 160; 170 and 120 U/gdm (CMCase, FPase, β-glucosidase, xylanase and polygalacturonase, respectively), demonstrating their potential to synthesize the enzymes compared with the results reported in the literature.

Thermophilic bacteria in Moroccan hot springs, salt marshes and desert soils BJM

The diversity of thermophilic bacteria was investigated in four hot springs, three salt marshes and 12 desert sites in Morocco. Two hundred and forty (240) thermophilic bacteria were recovered, identified and characterized. All isolates were Gram positive, rod-shaped, spore forming and halotolerant. Based on BOXA1R-PCR and 16S rRNA gene sequencing, the recovered isolates were dominated by the genus Bacillus (97.5%) represented by B. licheniformis (119), B. aerius (44), B. sonorensis (33), B. subtilis (subsp. spizizenii (2) and subsp. inaquosurum (6)), B. amyloliquefaciens (subsp. amyloliquefaciens (4) and subsp. plantarum (4)), B. tequilensis (3), B. pumilus (3) and Bacillus sp. Xylanase production by Aspergillus awamori under solid state fermentation conditions on tomato pomace BJM

In this work, tomato pomace, a waste abundantly available in the Mediterranean and other temperate climates agro-food industries, has been used as raw material for the production of some hydrolytic enzymes, including xylanase, exo-polygalacturonase (exo-PG), cellulase (CMCase) and α-amylase. The principal step of the process is the solid state fermentation (SSF) of this residue by Aspergillus awamori. In several laboratory experiments, maximum xylanase and exo-PG activities were measured during the first days of culture, reaching values around 100 and 80 IU/gds (international units of enzyme activity per gram of dried solid), respectively.
