The effects of insulin-like growth factor-I (IGF-I) on in vitro maturation (IVM) (experiment I) and on in vitro embryo development (experiment II) of bovine oocytes fertilized in vitro, were evaluated in terms of cleavage (CR), blastocyst (BR) and hatching (HR) rates. For IVM, immature cumulus-oocyte complexes were cultured in TCM-199 medium supplemented with Hepes, sodium bicarbonate, sodium pyruvate, additives, fetal calf serum (B-199 medium) and gonadotropins (14 U/ml PMSG and 7 U/ml hCG). La influencia de la nutrición en la función ovárica se clasifica como “efecto estático”, “efecto dinámico” (Meza-Herrera et al., 2004) y “efecto agudo” (Scaramuzzi et al., 2006). Dichos efectos son mediados por cambios en los niveles de hormonas metabólicas y de la superfamilia de factores de crecimiento (Meza-Herrera et al., 2006), pero su importancia relativa y rutas de acción necesitan ser dilucidados (Martin et al., 2004). The objective of this study was to evaluate the plasma concentrations of insulin-like growth factor-I (IGF-I), and the mRNA hepatic expression of IGF-I and of the growth hormone receptors GHR and GHR 1A, in postpartum beef cows. Four Angus and four crossbred (Angus x Nelore) postpartum suckled beef cows were used. Liver and blood samples were collected every 10 days, from calving to 40 days postpartum, for gene expression and for β-hydroxybutyrate and IGF-I assays, respectively. The objective of this study was to determine the effects of GDF-9, IGF-I, and GH alone or combined on preantral follicle survival, activation and development after 1 and 7 days of in vitro culture. Either fresh (non-cultured) or cultured ovarian tissue was processed for histological and fluorescence analysis. For all media tested, the percent of normal follicles was greater when compared to minimum essential medium supplemented (MEM+) alone, except when ovarian tissue was cultured with GDF-9/IGF-I or GDF-9/GH (P < 0.05). Avaliaram-se os efeitos da utilização de proteína láctea ou zinco suplementar na dieta sobre o desempenho, os pesos relativos do intestino delgado e fígado, a morfologia intestinal e as concentrações séricas de IGF-I (fator de crescimento semelhante à insulina), hormônio do crescimento e zinco em leitões. Una inyección de hormona bovina del crecimiento (bST) en la inseminación mejora el porcentaje de concepción (PC) en vacas repetidoras; sin embargo, no hay clara evidencia de su eficacia en vacas de primer servicio. Se evaluó si una inyección de bST en la inseminación mejora el PC al primer servicio en vacas Holstein. El experimento se hizo con 435 vacas de 15 hatos de Tizayuca, Hidalgo, México. We studied the effect of IGF-I, insulin and testosterone on intracellular Ca2+ in cultured muscle cells. Insulin produced a fast (<1 s) and transient [Ca2+] increase lasting less than 10 s. IGF-I induced a transient [Ca2+] increase, reaching a fluorescence peak 6 s after stimulus, to return to basal values after 60 s. Testosterone induced delayed (35 s) and long lasting (100-200 s) signals, frequently associated with oscillations. IGF-I, testosterone and electrical stimulation-induced Ca2+ signals were shown to be dependent on IP3 production. All of these Ca2+ signals were blocked by inhibitors of the IP3 pathway. Recent studies suggest that IGF-I is a crucial regulatory factor in follicular growth during early post-partum period. The aim of the present study was to determine in detail the changing profiles of metabolic and reproductive hormones in relation to ovulation of the dominant follicle (DF) of the first follicular wave post-partum in high-producing dairy cows. Plasma concentrations of related hormones in 22 multiparous Holstein cows were measured from 4 weeks pre-partum to 3 weeks post-partum, and the development of DF was observed with colour Doppler ultrasound.
